Insight of co-fermentation of carbon monoxide with carbohydrate-rich wastewater for enhanced hydrogen production: Homoacetogenic inhibition and the role of pH

Abstract:<p>Co-fermentation of CO and wastewater for H<sub>2</sub> generation could be a promising technology to maximize energy recovery and minimize the operation requirement, but H<sub>2</sub> consumption by homoacetogenesis remains the main challenge. The present study investigated the effect of different pH (initial and constant) and CO loading on CO and carbohydrate-rich wastewater co-fermentation for H<sub>2</sub> production and focused on homoacetogenesis behavior. After initial pH experiment, pH 5, 6, 7 and 8 were selected for constant pH experiment, and it was found that the highest H<sub>2</sub> production was obtained at constant pH 5 and CO loading of 2000 mL/d. Significant H<sub>2</sub> consumption was observed at constant pH 7 and 8, but not at pH 5 and 6. The injection of CO further promoted H<sub>2</sub> consumption at neutrophil pH 7–8. 16S rRNA gene sequencing and qPCR analysis were used to probe the underlying mechanisms. It was observed that at constant pH 5, CO injection resulted in a significant increase in the abundance of hydrogenogens (<em>Thermosinus</em> and <em>Moorella</em>) and high copies of CODH, accompanied with no obvious increase in the abundance of homoacetogens (<em>Desulfitobacterium</em>, and <em>Clostridium</em>) and low FHS copies. Thus, pH 5 is not only optimal for co-fermentation of CO and wastewater to generate H<sub>2</sub>, but also optimal to prevent H<sub>2</sub> consumption. Contrary results were obtained at constant pH 7 and 8. This study demonstrated a potential technique to achieve H<sub>2</sub> production economically from CO and wastewater co-fermentation system by enhancing hydrogenogenesis while constructing a balanced microbial community composition that restrains homoacetogenesis, which would provide a great foundation for industrial applications.</p>
