Abstract:Neurogenin 1 (ngn1), with the functions of controlling the differentiation of neurons, determining specific neuronal subtype, and inhibiting glial differentiation, is quantitatively detected for the first time. By using specifically modified silver nanoparticles (AgNP) as the signaling element, a label-free, rapid and sensitive colorimetric immunoassay for the synthetic peptide fragment of ngn1 (amino acid sequence: AQDDEQERRRRRGRTR) is reported. The detection procedure is based on an anti-aggregation mechanism, by which ngn1 inhibits the aggregation of the probe in the presence of salt (NaClO(4)). The anti-ngn1 antibody conjugated AgNP (denoted as AgNP-Ab) is negatively charged, and mono-binding of the like-charged ngn1 to the probe will increase the surface charge density, hence enhancing the interparticular electrostatic repulsion. Along with the increase of ngn1 concentration, the color of the solution varies from red to yellow, thereby developing a feasible approach for the detection of ngn1. Using a UV/vis spectrophotometer, this assay exhibits a linear response range of two orders of magnitude, from 50 to 800 ng mL(-1), and a detection limit of 30 ng mL(-1). On the basis of these qualities, the antibody-conjugated AgNP may become a useful tool for point-of-care diagnosis of ngn1 and such a method offers a new insight on the detection of the analogous antigen fragment as well.
Label-free Colorimetric Immunoassay for the Simple and Sensitive Detection of Neurogenin3 Using Gold Nanoparticles

Abstract:Neurogenin3 (ngn3), as a marker for pancreatic endocrine precursor cells and an essential ingredient in the development of islet cells, was quantitatively detected for the first time. Based on a non-cross-linking specific interaction mechanism, a label-free colorimetric immunoassay for the synthetic peptide fragment of ngn3 (SKQRRSRRKKAND) using glutathione (-Glu-Cys-Gly, GSH) functionalized gold nanoparticles (GNPs) is reported. The anti-ngn3 antibody conjugated GNPs (GNP-Ab) was formed through electrostatic interaction upon the addition of anti-ngn3 antibody to the GSH-modified GNPs solution. Monobinding of the positively charged ngn3 to the negatively charged GNP-Ab will minimize the electrostatic repulsion between nanoparticles by neutralizing the surface charge, and then agglomeration is induced by an increasing salt concentration. Under the optimal conditions, the assay showed a linear response range of 50-300 ng/mL for the peptide with a detection limit being 20 ng/mL. The preliminary study on ngn3 opens up an innovative insight to detect short synthetic peptide fragment of antigen, and may own an opportunity for practical applications in clinical diagnosis and therapeutics.

Abstract:A rapid and simple method of detection immunoglobulin was developed by using quantum dots labeled immunoassay. The Goat anti-Mouse IgG labelled quantum dots are mixed with Mouse IgG. After the immunoreaction, the quantum dots are aggregated to form a network, which can quench the fluorescence of themselves in the reaction system. The more Mouse IgG is, the bigger the network is, and the fluorescence intensity of quantum dots decreases continually. The concentration of Mouse IgG can be detected by the measurement of relative fluorescence intensity.

Abstract:A new method for detecting the nerve growth factors (NGF) is proposed based on the fluorescence quenching effect of carboxyl quantum clots. The research results show that in the pH7.5 buffer solution, the NGF antigen specifically binds to the carboxyl quantum dot-labeled NGF antibody, which can quench the fluorescence of the carboxyl quantum clots. The fluorescence quenching degree and the NGF antigen mass concentration has a good linear relationship in the range of 1-20 ng/mL, and the linear correlation coefficient is 0.9696 with the detection limit of 1 ng/ml,. This proposed method has good anti-interference characteristics and has great potential applications in diagnosis and detection.

Abstract:A highly selective, ultrasensitive, fluorescence detection method for DNA and antigen based on self-assembly of multiwalled carbon nanotubes (CNTs) and CdSe quantum dots (QDs) via oligonucleotide hybridization is reported. Mercaptoalkyloligonucleotide molecules bind to the quantum dots, while amineoalkyloligonucleotides bind to CNTs with -COCl surface groups. QDs and CNTs further assemble into nanohybrids through DNA hybridization in the presence of target complementary oligonucleotides. The method is achieved with good repeatability with the detection limit of 0.2 pM DNA molecules and 0.01 nM antigen molecules. This novel detection system can also be used for multicomponent detection and antigen-antibody immunoreaction. The novel system has great potential in applications such as ultrasensitive pathogen DNA or antigen or antibody detection, molecular imaging, and photoelectrical biosensors.

Abstract:We report a novel peptide probe for the detection of neurokinin-1 receptor using disaggregation-caused signal enhancement. The probe was obtained via the aggregation of a modified substance P in a terpyridine-Fe (II) complex with Gd (III)-DOTA into well-defined nanostructures, which effectively weaken ligand fluorescence and slow the exchange rate of inner-sphere water molecules. This probe disaggregates upon binding to the neurokinin-1 receptor and activates the contrast agents to generate a fluorescent signal that positively enhances magnetic resonance imaging contrast and allows for the detection of overexpressed receptors on tumor cells and the identification of lung cancer using serum samples.

Abstract:The use of polystyrene fluorescent microspheres conjugated to antigens and CdTe quantum dots (QDs) conjugated to antibodies as sensitive fluorescent labels in fluoroimmunoassay is described. CdTe QDs were attached to antibodies via electrostatic/hydrophilic self‐assembly. Both CdTe QD–labeled antibodies (anti‐IgGQD) and microsphere‐labeled antigens (IgGFM) were used to form a typical immunoassay on the surface of fluorescent microspheres. The immunoreaction system with different microsphere emissions and QDs can be detected effectively at a single excitation wavelength, because fluorescent QDs have broad excitation spectra and narrow emission bandwidths. This approach allowed detection of goat‐anti‐rabbit IgG in the range of 0.5 to 10 µg/mL. Furthermore, the immunocomplexes of anti‐IgGQD–IgGFM were confirmed by coincidence detection of the fluorescence signals of CdTe QDs and microspheres on a microfluidic on‐chip device.

Abstract:In this paper, a new fluorescence bioassay for Golgi protein-73 (GP73), a promising marker for monitoring liver tumor, was developed by using anti-GP73 antibody (GP73 Ab) capped quantum dots (QDs) coupled with protein A/G agarose beads in an attempt to improve the analysis time, cost and operation. First, carboxylic-functionalized Mn modified CdTe/CdS QDs were synthesized and covalently conjugated with GP73 Ab, then protein A/G agarose beads were specifically combined with the QDs-conjugated Ab to form the QDs-Ab-beads conjugate, which could capture and separate GP73 from the sample through simple centrifugation. It was found that the fluorescence intensity of the above QDs-Ab-beads biosensor could be specifically quenched by GP73 added. A simple, rapid and specific quantitative method for GP73 protein was proposed using the as-prepared QDs-Ab-beads as a biosensor. Under the optimized conditions, the calibration curve of the proposed assay showed good linearity with a correlation coefficient of 0.9935 in the concentration range of 20-150 ng/mL of GP73 protein. The limit of detection (defined as 3σ/K) was 10 ng/mL. The method built exhibited a great potential in the clinic test of GP73.

Abstract:N-acetylneuraminic acid (Neu5Ac) is widely spread in many biologically significant glycans of mammals, commonly as a terminal alpha-glycoside. It is of great significance to develop analytical techniques for detection of Neu5Ac. Herein, a high-sensitive fluorescent biosensor for Neu5Ac has been developed based on FRET between CdSe/ZnS quantum dots (QDs) and BHQ(2), as well as exonuclease III (Exo III)-assisted recycling amplification strategy. Employing the specially designed three-level FRET systems and fluorescent signal recovery mechanism, together with five-step recycling signal amplification chain reactions, an ultralow detection limit of 24 fM was achieved. Meanwhile, good linear response ranges within 0.2-12.5 pM and 12.5-1000 pM were founded. The assay has excellent performance in real sample detection, and thus offers great potential for detection of sialic acids modified glycans/lipids in the fields of medical diagnosis and food testing.

Abstract:An immunoassay for Escherichia coli O157:H7 using quantum-dot (QD) labeling and subsequent release of the QD labels from immunocomplexes has been developed. The assay principle is that anti-E. coli O157:H7 conjugated immunomagnetic beads are used to capture E. coli O157:H7; this is followed by the binding of QD labeled antibodies to form sandwich immunocomplexes (Bead-Cell-QD); a dissociation buffer then elutes QDs from immunocomplexes by denaturing antibody or lysing cell; the fluorescence signal of the eluted QDs is measured to quantify E. coli O157:H7. This proposed approach avoids the interference of bead autofluorescence in signal transduction and, thus, enhances the detection resolution, while keeping the fast magnetic separation and sandwich binding of two selective antibodies for high specificity.

Abstract:A magnetic bead based sensitive sandwich assay for C-reactive protein (CRP) has been developed. In this assay, magnetic microbeads conjugated with monoclonal anti-CRP are used to capture CRP in human serum, and then sequentially incubated with biotinylated monoclonal anti-CRP and streptavidin coated quantum dots (QDs) to form sandwich immunocomplexes. The immunocomplexes are further mixed with an immunoaffinity separation buffer to release QDs from the magnetic bead surface by dissociating antibody-antigen pairs. After magnetic bead separation, the fluorescence signal of QDs released in the immunoaffinity separation buffer is measured to quantify CRP. This proposed approach takes advantages of magnetic beads for fast magnetic separation, sandwich binding of two monoclonal antibodies for high specificity, and QD fluorescence labeling for high detection sensitivity. Moreover, it adopts immunoaffinity separation to avoid the interference of magnetic bead autofluorescence in signal transduction and thus enhances the detection resolution of the assay. The detection limit of CRP is 0.5 fM, corresponding to 10 pM CRP in 50 microL of sample volume, and the detection range is of around 3 orders of magnitude.

Abstract:A novel immunosensor of human immune globulin (IgG) was fabricated based on the fluorescence transfer between luminescent semiconductor quantum dots (QDs) and gold nanoparticles (AuNPs). AuNPs and CdSe/ZnS QDs were respectively labeled with immune reaction pair:IgG and goat anti-human immunoglobulin (GIgG), by optimizing the conditions including pH value and protein amount. In the assembled QD-GIgG-IgG-AuNP fluorescence resonance energy transfer (FRET) immunocomplex system, the presence of AuNP-IgG directly reduced the fluorescence intensity of the GIgG conjugated QDs. As a result, the concentration of AuNP-IgG had a linear relationship with the fluorescence decrease in a range of 0-1.57 microg/mL. Furthermore, the mechanism of the QDs' fluorescence decay has also been discussed and attributed to the light-induced photobleaching. This novel sensing method achieves quantitative detection of trace proteins, suggesting the potential of biomolecule-AuNPs conjugation based analytical methods in further application.

Abstract:A novel quenching electrochemiluminescence (ECL) immunosensor for trace detection of neuron-specific enolase (NSE) was developed. Doping CdS with Zn2+ to obtain ZnCdS not only significantly improves the original properties of CdS, but also can be loaded onto the surface of g-C3N4 to form a special complex structure, which effectively improves the ECL performance of g-C3N4. Rh0.6Ru0.4 alloys with flower-like structures were prepared by simple hydrothermal synthesis, with excellent electrical conductivity and catalytic performance. Due to its special flower-like structure with large specific surface area, Ag nanoparticles can be uniformly loaded on its surface to form Rh0.6Ru0.4@Ag composites. Through spectral comparison, it is found that Rh0.6Ru0.4@Ag has strong UV-vis absorption in the ECL emission wavelength range of g-C3N4, which can generate resonance energy transfer, thereby effectively quenching the ECL signal. The immunosensor has a wide detection range with a detection limit of 16 fg/mL (S/N=3), good stability and biocompatibility, and has great potential in clinical applications.

Abstract:Electrochemiluminescence (ECL) of highly efficient and non-toxic carbon quantum dots (CQDs) plays an important role in immunoassays. However, the high absolute value of the excitation potential and poor controllability make precise detection of low levels difficult. In this work, novel CQDs with low excitation potential were reported to be used for the ultra-sensitive immunoassay of neuron-specific enolase (NSE). It is noteworthy that the narrow bandgap CQDs exhibit a trigger potential of -0.5 V and show excellent applicability of ECL signal at an excitation potential of -0.8 V. This property eliminates side reactions and electrode passivation and significantly improves ECL efficiency. Based on this, the magnetic zeolite-imidazole framework (MZIF) nanoreactor with co-reaction acceleration was employed as a carrier for the in-situ encapsulation of CQDs. The constrained-enhanced ECL sensor platform demonstrated the ability to achieve stable and controllable signal output. A sandwich-type biosensor was constructed by introducing copper sulfide (Cu7S4) as a dual-mechanism quencher, with NSE serving as the target analyte. Moreover, the electrochemical detection method based on the differential pulse voltammetry (DPV) properties of Cu7S4, was successfully validated against ECL. The sensors constructed on magnetic electrodes exhibited a bimodal signal output of ECL and DPV, encompassing a wide detection range (0.001 ng/mL - 500 ng/mL) and low detection limit (0.18 pg/mL for ECL and 0.15 pg/mL for DPV). This design offers a novel approach to understanding the efficient emission of carbon dots and provides a solution for the sensitive detection of biomarkers in clinical research.
Sensitive fluorescence detection of DNA using isothermal exponential amplification coupled quantum dots coated silica nanospheres as label

Abstract:A new strategy to combine isothermal exponential amplification (IEA) with CdTe quantum dots (QDs) functionalized silica nanospheres as a label was designed for highly sensitive fluorescent detection of target DNA. A well plate was used as a support to immobilize a molecular beacon (MB) as a recognition probe and perform the IEA procedure. After the MB recognized target DNA and opened its cycle, the stem part could hybridize with a primer to initiate the polymerization of the DNA strand, which led to the release of target to open another MB molecule and start the next cycle of strand-replacement polymerization. Meanwhile, the formed double-stranded DNA was recognized by nicking endonuclease, leading to an endonuclease-based strand-replacement polymerization, which produced a DNA trigger to open more MB. The opened MB molecules were finally bound to the label by biotin-streptavidin coupling. Upon a dissolving process, the released cadmium cation could sensitize the fluorescent emission of Rhod-5N to achieve cascade signal amplification. The proposed method could detect target DNA ranging from 10(-17) to 10(-11) mol L-1 with a detection limit down to similar to 50 copies. It also showed high selectivity. This highly sensitive and specific assay has potential to become a promising DNA quantification method in biomedical research and clinical diagnosis.

Abstract:A versatile immunosensor using CdTe quantum dots as electrochemical and fluorescent labels has been developed for sensitive protein detection. This sandwich-type sensor is fabricated on an indium tin oxide chip covered with a well-ordered gold nanoparticle monolayer. Gel imaging systems were successfully introduced to develop a novel high-efficient optical immunoassay, which could perform simultaneous detection for the samples with a series of different concentrations of a target analyte. The linear range of this assay was between 0.1 and 500 ng/ mL, and the assay sensitivity could be further increased to 0.005 ng/mL with the linear range from 0.005 to 100 ng/mL by stripping voltammetric analysis. The immunosensor showed good precision, high sensitivity, acceptable stability, and reproducibility and could be used for the detection of real sample with consistent results in comparison with those obtained by the ELISA method.

Abstract:A paper-based visual fluorescence immunoassay is presented for the detection of matrix metalloproteinase-7 (MMP7) that is related to renal cancer. The method is based on the distance-dependent fluorescence quenching of CdTe quantum dots (QDs) on a nitrocellulose membrane by Ag+ following a sandwich-type immunoreaction on microtiter wells using silver nanoparticle (AgNP)–labeled secondary antibody– and primary antibody–coated microtiter wells. The silver nanoparticles captured in the well are dissolved with HNO3, while the quenching effect of QDs is based on silver ion-exchange reaction under 365-nm excitation light irradiation. Increasing concentration of released Ag+, thus higher concentration of the protein, leads to an increased distance of quenching on the nitrocellulose membrane. The paper-based immunoassay by combination of AgNP-assisted ion-exchange reaction with QD gives good distance-dependent responses and allows the detection of MMP7 at a concentration as low as 7.3 pg mL−1. The coefficients of variation are less than 6.9% and 12.4% for intra-assay and inter-assay, respectively. High specificity and long-term stability are achieved during the assay. Importantly, the testing of human serum samples using our strategy shows well-matched results with commercial human MMP7 ELISA kits.

Rapid detection of specific genes from human genomic DNA using the microbead-quantum dot complexes in microfluidic chip

Abstract:In the clinic, it is important to prepare for single stranded DNA from genomic DNA to detect the target gene. In this study, we have investigated the detection of single stranded DNA (ssDNA) obtained by ultrasonication of human genomic DNA via fluorescence quenching on microfluidic chip with pillars at the channel to entrap the microbead-QD complexes (MQCs). The QDs with carboxyl group bind to microbeads with amine group by EDC/NHS coupling reaction. The thiolated probe DNA conjugates strongly with the metal ions on the surface of QDs. The MQCs were packed into a chamber on the channel blocked by pillars. ssDNA and TOTO-3 (intercalating dye) were introduced into the microchannel. After hybridization of probe DNA and target DNA, fluorescence quenching was observed at the surface of the MQDs by FRET between QD and TOTO-3. This experiment shows the possibility of rapid detection of genomic DNA from clinical samples via microbead-QD complexs on microfluidic chip.

Abstract:An immunoassay readout method based on fluorescent imaging analysis with laser confocal scanning is described. The ZnS-coated CdSe quantum dots (ZnS/CdSe QDs) were linked to a detection antibody. Immunoassay was carried out on a glass chip using a sandwich assay approach, where antibody covalently bound to a glass chip was allowed to capture antigen specially. Afterwards, the detection antibody labeled with QD was allowed to bind selectively to the captured antigen. The fluorescent signals of the sandwich conjugate were detected by a laser confocal scanner. A diode laser was used to excite efficiently the fluorescent signals while bovine serum albumin was used to eliminate nonspecific binding sites. The detection limit of this approach was up to 10−9 M under current experimental conditions. The specificity of the QDs-labeled immunoglobulin (IgG) was tested by an experiment using goat IgG and human IgG samples. The result was consistent with the binding specificity in a sandwich-type assay. The potential of this method to function as a simple and efficient readout strategy for immunoassay in biochip is discussed.
A sensitive fluorescence assay based on aggregation-induced emission by copper-free click reaction for rapid ctDNA detection

Abstract:A fluorescence assay was developed based on aggregation-induced emission for circulating tumor (ctDNA) rapid detection. AIE-based probe (TPE-DNA) was constructed via strain-promoted azide-alkyne cycloaddition reaction between the azide-functionalized TPE-N 3 and the dibenzocyclooctyne functionalized DNA. AIE-based assay displayed significant fluorescence signal enhancement for the ctDNA targets. The experimental results were verified and explained based on density functional theory simulations. As a proof-of-concept, the ctDNA as the target was detected to determine the feasibility of the assay. The detection could be done within 10 min with the limit of detection of 39 pM and good selectivity without the assistance of enzyme or nano-materials. Compared with traditional click reaction, the assay has advantages of high efficiency and good biocompatibility. It is promising in rapid diagnostic of cancer marker nucleic acid with further efforts to develop intelligent miniaturized equipment.
