Abstract:Optical Molecular imaging is an emerging alternative to nuclear-based imaging techniques for detecting cancer. In this paper, we investigate reflectance mode multi-frequency domain optical imaging with novel theranostic nanophotonic probes integrating silica core gold nanostructures with Indocyanine Green dye. From phase sensitive images obtained from homodyne optical measurements we analyze amplitude and phase contrast to determine optimal measurement acquisition parameters for nanoshell enhanced fluorescent dyes.

Abstract:We study the spectral dependence of the scattered light from Au nanorods illuminated by 50-fs pulses resonant with the longitudinal plasmon, and determine the origin of the scattering in terms of the plasmon modes. We project the expected spectrum under illumination from a squeezed optical pump.

Abstract:Anti-Stokes one-photon luminescence from a single gold nanorod is experimentally investigated. The anti-Stokes emission of gold nanorods is enhanced and strongly modulated by localized surface plasmon resonance (LSPR). It is found that the polarization dependence of the anti-Stokes emission is in strong correlation with that of the Stokes emission. Further experiments provide evidence that LSPR significantly enhanced both excitation and emission processes. Moreover, the line shape of the anti-Stokes emission is dependent on the surface temperature, which is related to the distribution of free electrons near the Fermi level. This discovery provides an effective method in principle to probe localized temperature at nanoscale dimension. Here, the reported results about the anti-Stokes emission provide more understanding for the photoemission process from the plasmonic nanostructures.

Abstract:Metallic nanoparticles have opened the possibility of imaging, tracking, and manipulating biological samples without time limitations. Their low photoluminescence quantum yield, however, makes them hard to detect under high background conditions. In this study we show that it is possible to image gold nanorods by detecting their anti-Stokes emission under resonant excitation. We show that even in the membrane of a cell containing the fluorescent dye Atto 647N, the signal/background of the anti-Stokes emission can be >10, while it is impossible to image the particles with the Stokes emission. The main advantage of this technique is that it does not require any major change in existing fluorescence imaging setups, only the addition of an appropriate short-pass filter in the detection path.
Simultaneous Excitation and Emission Enhancement of Fluorescence Assisted by Double Plasmon Modes of Gold Nanorods

Abstract:Here we propose a scheme utilizing the double plasmon modes of gold nanorod (GNR) to efficiently enhance the fluorescence of surrounding emitters. The transversal and longitudinal surface plasmon resonance (TSPR and LSPR, respectively) modes of GNR are simultaneously utilized to enhance the excitation and emission efficiency, respectively. To demonstrate the idea, GNRs coated with an Oxazine-725 dye molecule-doped silica shell are employed. For comparison, gold nanospheres with the same shell are also studied, of which the single plasmon resonance mode matches only with the excitation wavelength of Oxazine-725. The experimental results, in agreement with theoretical simulations using the discrete dipole approximation method, successfully demonstrate the efficient excitation and emission enhancement of fluorescence assisted by the double SPRs of GNRs.

Abstract:Mesoporous encapsulation of gold nanorods (GNRs) in a silica shell of controllable thickness (4.5-25.5 nm) was realized through a single-step coating method without any intermediary coating. The dependence of localized surface plasmon resonance (LSPR) extinction spectra of the coated GNRs on the thickness of the silica shell was investigated with both simulation and experiments, which agreed well with each other. It was found that cetyltrimethyl ammonium bromide (CTAB) molecules, which act as surfactants for the GNRs and dissociate in the solution, greatly affect the silica coating. Mesoporous silica-encapsulated GNRs were also shown to be highly biocompatible and stable in bio-environments. Based on LSPR enhanced scattering, mesoporous silica-encapsulated GNRs were utilized for dark field scattering imaging of cancer cells. Biomolecule-conjugated mesoporous silica-encapsulated GNRs were specifically taken up by cancer cells in vitro, justifying their use as effective optical probes for early cancer diagnosis. Mesoporous silica can also be modified with functional groups and conjugated with certain biomolecules for specific labeling on mammalian cells as well as carrying drugs or biomolecules into biological cells.

Abstract:Plasmon-resonant gold nanorods (GNRs) are demonstrated as strong absorption contrast agents for optical coherence tomography (OCT). OCT imaging of tissue phantoms doped with GNRs of different resonant wavelengths and concentrations is studied. To utilize the high absorption property of GNRs, a differential absorption OCT imaging is introduced to retrieve the absorption information of GNRs from conventional backscattered signals. It is shown that the contrast of the OCT image can be enhanced significantly when the plasmon resonant wavelength of the GNRs matches the central wavelength of the OCT source.

Nonlinear Optical Properties Of Gold Nanorods (Gnrs) Under Fs Laser Excitation Near The Third Optical Tissue Window And Application For Multichannel Cellular Imaging

Abstract:Gold nanorod (GNR) was a promising bioimaging probe in the third optical tissue window (1600-1800 nm). In this paper, hydrophilic GNRs with proper dimensions were synthesized according to the classical seed mediated method. Their nanostructures were taken with a transmission electron microscope and their extinction spectra were recorded on a UV-vis scanning spectrophotometer. Their nonlinear optical features were recorded by a home built measurement system, under the femto second (fs) laser excitation of 1580 nm, which was selected as the demonstration of the third optical tissue window. A sharp third harmonic generation (THG) signal and a mild three photon luminescence (3PL) signal could be clearly distinguished. These signals could be utilized for multichannel nonlinear bioimaging in the third optical tissue window.Negatively charged polystyrene sulfonate (PSS) and positively charged polyallylamine hydrochloride (PAH) were separately coated onto the positively charged GNRs through electrostatic adherence. The PAH-PSS coated GNRs were added in the culture medium of HeLa cells. Multichannel nonlinear optical imaging of cancer cells stained with GNRs was performed with a 1580 nm fs laser coupled scanning microscope. The signals were filtered by a 665 nm dichroic mirror, and then splitted by another 560 nm dichroic mirror into the green and red imaging channels. HeLa cells can be clearly visualized in both green and red channels with good contrast. For HeLa cells without GNRs treatment, there were no such signals detected in these channels. This was a pioneer work of the multichannel nonlinear bioimaging in the third optical tissue window with GNRs and would be helpful for our further studies on in vivo bioimaging in this window with deeper penetration

Abstract:Light emission from gold nanoparticles was investigated with ultra-narrow-band notch filters to obtain the complete spectral shape. The anti-Stokes emission band was observed at all excitation wavelengths. The spectral shape of the anti-Stokes emission could be well fitted by a Fermi–Dirac-like line shape, while the spectral profile of the Stokes emission could be fitted by a Lorentzian line shape. The electron distribution and local surface plasmon resonance jointly determined the spectral shape. Additionally, we found that the anti-Stokes emission intensity increased more rapidly compared with that of the Stokes emission as illumination power was increased. This phenomenon can be understood from the temperature dependence of the electron distribution owing to photothermal effects.

Abstract:Lanthanide-doped upconversion nanocrystals (UCNCs) have recently become an attractive nonlinear fluorescence material for use in bioimaging because of their tunable spectral characteristics and exceptional photostability. Plasmonic materials are often introduced into the vicinity of UCNCs to increase their emission intensity by means of enlarging the absorption cross-section and accelerating the radiative decay rate. Moreover, plasmonic nanostructures (e.g., gold nanorods, GNRs) can also influence the polarization state of the UC fluorescence—an effect that is of fundamental importance for fluorescence polarization-based imaging methods yet has not been discussed previously. To study this effect, we synthesized GNR@SiO2@CaF2:Yb3+,Er3+ hybrid core–shell–satellite nanostructures with precise control over the thickness of the SiO2 shell. We evaluated the shell thickness-dependent plasmonic enhancement of the emission intensity in ensemble and studied the plasmonic modulation of the emission polarization at the single-particle level. The hybrid plasmonic UC nanostructures with an optimal shell thickness exhibit an improved bioimaging performance compared with bare UCNCs, and we observed a polarized nature of the light at both UC emission bands, which stems from the relationship between the excitation polarization and GNR orientation. We used electrodynamic simulations combined with Förster resonance energy transfer theory to fully explain the observed effect. Our results provide extensive insights into how the coherent interaction between the emission dipoles of UCNCs and the plasmonic dipoles of the GNR determines the emission polarization state in various situations and thus open the way to the accurate control of the UC emission anisotropy for a wide range of bioimaging and biosensing applications.

Abstract:An individual gold nanorod as an optical antenna to modulate single-molecule fluorescence spontaneous emission behaviors is investigated theoretically. 2D and 3D numerical finite-difference time-domain methods are implemented to investigate changes in the excitation rate, spontaneous emission rate, quantum efficiency, and emission spectral shape as functions of the separation between the emitter and nanorod. Our simulations reveal that the 3D relative configuration between the gold. nanorod and the single dipole definitely affects the quantum efficiency and emission spectral shape. The orientation of the dipole and the polarization of the excitation light are also investigated to clarify the polarization dependence of the plasmonic-enhanced fluorescence. Furthermore, we calculate the modified quantum efficiency and emission spectral shape of a single Cy5 dye molecule in the vicinity of a single gold nanorod taking experimental requirements into account.

Abstract:Gold nanorods (GNRs) have been widely used for bio-imaging. However, GNRs assisted optical in vivo deep tissue imaging is severely restricted due to signal attenuation, low contrast, complex process or low real-timing. To overcome these problems, we functionalized GNRs with both near-infrared (NIR) fluorescence and surface enhanced Raman scattering (SERS) and utilized these co-functionalized GNRs for purely optical in vivo imaging of live mice. Our proposed technology has the combined advantages of high real-timing, high imaging contrast and deep detection ability. The distribution and excretion of intravenously injected GNRs in deep tissues of live mice were observed in vivo for the first time through purely optical imaging. We also demonstrated successfully in vivo biomedical applications of the co-functionalized GNRs to sentinel lymph node (SLN) mapping and tumor targeting of mice. The present technology has great future potentials for disease diagnosis and clinical therapies.

Abstract:Imaging electrochemical processes has attracted increasing attention in past decades. Particularly, monitoring electrochemical reactions rapidly at the nano-scale is still a challenge due to the ultra-low current detection and long scanning time required. The development of optical techniques provide a new way to demonstrate electrochemical processes through optical signals which enhance sensitivity and spatial resolution. Herein, we developed a novel method to image electrocatalytic processes on single gold nanorods (GNRs) during Cyclic Voltammetry (CV) scanning based on plasmon resonance scattering information by using dark-field microscopy. The electrocatalytic oxidation of hydrogen peroxide was selected as a typical reaction and the catalytic mechanism was revealed using the obtained spectra. Notably, observation on single GNRs avoided the averaging effects in bulk systems and confirmed that the individual nanoparticles had variable catalytic properties with different spectral change during the reaction process. Furthermore, a color-amplified system was introduced to convert light intensity into imaging information via the Matlab program which was able to image thousands of nanoparticles simultaneously. This approach offered the statistical intensity distribution of all of the nanoparticles in a dark-field image which dramatically enhanced the detection accuracy and avoided random events.

Abstract:We measure nonlinear optical scattering from individual Au nanorods excited by ultrafast laser pulses on resonance with their longitudinal plasmon mode. Isolating single rods removes inhomogeneous broadening and allows the measurement of a large nonlinearity, much greater than that of nanorod ensembles. Surprisingly, the ultrafast nonlinearity can be attributed entirely to heating of conduction electrons and does not exhibit any response associated with coherent plasmon oscillation. This indicates a previously unobserved damping of strongly driven plasmons.

Abstract:Biological applications of core/shell near-infrared quantum dots (QDs) have attracted broad interest due to their unique optical and chemical properties. Additionally, the use of multifunctional nanomaterials with near-infrared QDs and plasmonic functional nanoparticles are promising for applications in electronics, bioimaging, energy, and environmental-related studies. In this work, we experimentally demonstrate how to construct a multifunctional nanoparticle comprised of CdSe/ZnS QDs and gold nanorods (GNRs) where the GNRs were applied to enhance the photoluminescence (PL) of the CdSe/ZnS QDs. In particular, we have obtained the scattering PL spectrum of a single CdSe/ZnS QD and GNR@CdSe/ZnS nanoparticle and comparison results show that the CdSe/ZnS QDs have an apparent PL enhancement of four-times after binding with GNRs. In addition, in vitro experimental results show that the biostability of the GNR@CdSe/ZnS nanoparticles can be improved by using folic acid. A bioimaging study has also been performed where GNR@CdSe/ZnS nanoparticles were used as an optical process for MCF-7 breast cancer cells.
