Abstract:The refractive index (RI) distribution can serve as a natural label for undyed cell imaging. However, the majority of images obtained through quantitative phase microscopy is integrated along the illumination angle and cannot reflect additional information about the refractive map on a certain plane. Herein, a light-field reconstruction method to image the RI map within a depth of 0.2 μm is proposed. It records quantitative phase-delay images using a four-step phase shifting method in different directions and then reconstructs a similar scattered light field for the refractive sample on the focus plane. It can image the RI of samples, transparent cell samples in particular, in a manner similar to the observation of scattering characteristics. The light-field reconstruction method is therefore a powerful tool for use in cytobiology studies.

Abstract:The resolution of reconstructed images is significantly limited by the size of captured holograms in digital holography. A multiplane digital holographic algorithm based on extrapolation iterations is proposed herein. Holograms at different axial distances are captured with a shifting imaging sensor and then extrapolated to a larger hologram after padding. Multiple holograms provide multipoint constraints in diffraction calculation. Constraints based on the energy distribution and transmissions of an object are applied to extract the object function from the holograms. The method achieves a higher convergence speed for solving the inverse problem compared with the single-plane extrapolation method. We demonstrate that the reconstruction in multiple planes yields a better resolution than that in one plane.

Abstract:The strong need in materials and biological science has prompted the development of high-speed quantitative phase imaging. However, for phase retrieval applying digital micromirror devices (DMDs), the accuracy of the retrieved phase will be disturbed by the DMD-induced aberrations. Here, we propose a phase retrieval method based on measuring and correcting errors caused by phase non-uniformity of the device. Using only four binary amplitude masks and corresponding diffraction intensities, the proposed method achieves rapid convergence and high-quality reconstruction. The experiments prove the practical feasibility for general samples and the effective improvement of the retrieved phase accuracy.

Abstract:Digital Holography Microscopy (DHM) is a powerful tool for three-dimensional imaging of micro-objects, since it yields an intensity image, called hologram. This is interference pattern, records the intensity information and allow reconstruction of the phase shift introduced by objects. With the development of computer and fast Fourier transformation technology, DHM has been widely used in biological and material tissues, etc.. In this paper, DHM is used to measure the abrupt edges and surface of reflect objects which are calibrated. Phase compensation uses the physical configuration to compensate the phase aberration, such as Microscope Objective (MO), spherical aberration and other curvature introduced by specimen or instruments in set-up. We apply this method to obtain the pure holograms of objects. The holograms produce more accurate phase-contrast imaging and three-dimensional imaging. Angular Spectrum Propagation (ASP) approach is a strict diffraction integration, and widely used for many applications, such as short-distance propagation, and the off-axis system and tilted plane. Here, we achieve numerical reconstruction of the hologram with ASP under phase compensation condition in an off-axis system.First, we use the fast Fourier transformation for the hologram and get the frequency image. Then, filter out the spectrum about the image which we need. Finally, inverse Fourier transform of the filtered spectrum and get the complex amplitude information. The phase image and intensity image of specimen is obtained after calculating the reconstruction distance.

Abstract:The morphology and dynamics of label-free tissues can be exploited by sample-induced changes in the optical field from quantitative phase imaging. Its sensitivity to subtle changes in the optical field makes the reconstructed phase susceptible to phase aberrations. We import variable sparse splitting framework on quantitative phase aberration extraction based on alternating direction aberration free method. The optimization and regularization in the reconstructed phase are decomposed into object terms and aberration terms. By formulating the aberration extraction as a convex quadratic problem, the background phase aberration can be fast and directly decomposed with the specific complete basis functions such as Zernike or standard polynomials. Faithful phase reconstruction can be obtained by eliminating global background phase aberration. The aberration-free two-dimensional and three-dimensional imaging experiments are demonstrated, showing the relaxation of the strict alignment requirements for the holographic microscopes.

Abstract:Objective measurements of the morphology and dynamics of label-free cells and tissues can be achieved by quantitative phase with low phototoxicity and no photobleaching. Modern quantitative optical imaging possesses a huge information capacity. The morphology and dynamics of label-free tissues can be exploited by sample-induced changes in the optical field from quantitative phase imaging. Its sensitivity to subtle changes in the optical field makes the reconstructed phase susceptible to phase aberrations. We present aberration-free high-bandwidth holographic microscopy which exploits high-throughput label-free quantitative phase imaging. Firstly, a full-bandwidth holographic reconstruction is retrieved from interferograms by establishing a holographic multiplexing framework. Based on the analyticity of band-limited signal under a diffraction-limited system, the maximum space bandwidth utilization limit in a single multiplexing hologram is increased to the maximum sensor limit. Secondly, A variable sparse splitting framework on quantitative phase aberration extraction is imported based on the alternating direction aberration-free method. By formulating the aberration extraction as a convex quadratic problem, the background phase aberration can be fast and directly decomposed with the specific complete basis functions such as Zernike or standard polynomials. Faithful high throughput phase reconstruction can be obtained by eliminating global background aberration. It opens a new route to multiplexing quantitative optical imaging and helps to improve the performance of constraint-free modern optical microscopes in various spectral regimes.

Abstract:In the digital holographic microscopy, the phase aberration is a key problem limiting the quantitative phase measurement, especially for the microstructure whose phase distribution is dense with few and discontinuous background region. A numerical phase aberration compensation method with global curve fitting preprocessing and automatic extraction of the background region for microstructure testing is proposed. The global curve fitting is implemented to compensate most of the tilt, quadratic and partial high order aberrations and make the profile of the measured object significantly distinguishable. Then the background region is automatically extracted by directly segmenting the converted grayscale phase image. At last, the Zernike polynomial fitting is performed on the extracted background region, and the obtained coefficients are used to calculate the phase aberration of the whole phase image. Simulation and experimental results demonstrated that the proposed method is able to accurately extract background region and compensate the phase aberration for the complicated dense microstructure.

Abstract:Digital holography is an effective technology in image reconstruction as amplitude and phase information of cells can be acquired without any staining. In this paper, we propose a holographic technique with an improved Gerchberg-Saxton (GS) algorithm to reconstruct cell imaging based on phase reconstruction information. Comparative experiments are conducted on four specific models to investigate the effectiveness of the proposed method. The morphological parameters (such as shape, volume, and sphericity) of abnormal erythrocytes can be obtained by reconstructing cell hologram of urinary sediment. Notably, abnormal red blood cells can also be detected in mussy circumstances by the proposed method, owing to the significantly biophysical contrast (refractive index distribution and mass density) between two different cells. Therefore, this proposed method has a broad application prospect in cell image reconstruction and cell dynamic detection.

Abstract:This paper proposes a new method that reconstructs the information of specimen by using random phase shift step in digital holographic microscopy (DHM). The principles of the method are described and discussed in detail. In practical experiment, because the phase shifter is neither perfectly linear nor calibrated, digital holograms with inaccurate phase shift step are recorded by the charge-coupled device (CCD). The phase could be accurately reconstructed from the recorded digital holograms by using the random phase-shifting algorithm, which makes up for reconstructed phase error caused by ordinary phase-shifting algorithm. The phase aberration compensation is also discussed. In order to verify the flexibility of the proposed method, numerical simulation of random phase-shifting DHM was carried out. The simulation results illustrated that the presented method is effective when the phase shift step is unknown or random in DHM.

Abstract:This paper presents the performance of electronic holography for imaging applications of biological and medical interest involving turbid media. It demonstrates that digital holography is useful for biological studies in the areas of estuary sediment erosion and imaging unstained living cells. In particular, we demonstrate the recording geometries of classical 'in-line' back illumination using a collimated beam for measuring micro-panicles eroded in a cylindrical observation chamber; we also demonstrate a phase contrast set-up using a magnifying lens for observing biomedical micro-objects of unstained living cells in physiological saline. Practical results are presented for the studies of sediment erosion and transport, which provide information about how polymeric substances produced by the organisms in sediment and erosion shear velocity affect the sediment particle size when erosive stresses are applied to sediment beds.

Abstract:In 1948, Dennis Gabor proposed the concept of holography, providing a pioneering solution to a quantitative description of the optical wavefront. After 75 years of development, holographic imaging has become a powerful tool for optical wavefront measurement and quantitative phase imaging. The emergence of this technology has given fresh energy to physics, biology, and materials science. Digital holography (DH) possesses the quantitative advantages of wide-field, non-contact, precise, and dynamic measurement capability for complex-waves. DH has unique capabilities for the propagation of optical fields by measuring light scattering with phase information. It offers quantitative visualization of the refractive index and thickness distribution of weak absorption samples, which plays a vital role in the pathophysiology of various diseases and the characterization of various materials. It provides a possibility to bridge the gap between the imaging and scattering disciplines. The propagation of wavefront is described by the complex amplitude. The complex-value in the complex-domain is reconstructed from the intensity-value measurement by camera in the real-domain. Here, we regard the process of holographic recording and reconstruction as a transformation between complex-domain and real-domain, and discuss the mathematics and physical principles of reconstruction. We review the DH in underlying principles, technical approaches, and the breadth of applications. We conclude with emerging challenges and opportunities based on combining holographic imaging with other methodologies that expand the scope and utility of holographic imaging even further. The multidisciplinary nature brings technology and application experts together in label-free cell biology, analytical chemistry, clinical sciences, wavefront sensing, and semiconductor production.

Abstract:We present a technically simple implementation of quantitative phase imaging in confocal microscopy based on synthetic optical holography with sinusoidal-phase reference waves. Using a Mirau interference objective and low-amplitude vertical sample vibration with a piezo-controlled stage, we record synthetic holograms on commercial confocal microscopes (Nikon, model: A1R; Zeiss: model: LSM-880), from which quantitative phase images are reconstructed. We demonstrate our technique by stain-free imaging of cervical (HeLa) and ovarian (ES-2) cancer cells and stem cell (mHAT9a) samples. Our technique has the potential to extend fluorescence imaging applications in confocal microscopy by providing label-free cell finding, monitoring cell morphology, as well as non-perturbing long-time observation of live cells based on quantitative phase contrast.

Abstract:Phase aberrations must be precisely compensated for phase-contrast imaging in digital holographic microscopy. In this paper, an automatic and accurate phase aberration compensation method is proposed based on iteratively reweighted least squares fitting. Instead of selecting the known-to-be-flat region with manual operation or image segmentation, this method starts from conventional least squares fitting in the whole image and attributes lower weights to the pixels with higher fitting residuals in an iterative reweighting procedure, during which the influence of specimen and noise is automatically reduced. Both phase curvature and high-order aberrations could be corrected without extra operation. Numerical simulation demonstrates that the proposed method is more accurate than the state-of-the-art numerical methods. Experimental result proves that this method is robust and fast enough for dynamic imaging of living cells.

Abstract:In this paper, a novel approach was implemented to image the marine plankton using the in-line digital holographic technology. the digital holography can image all plankton in a certain volume and more information can be recorded including the intensity and phase information. Moreover, the lensless system cases no aberrations and reduces the complexity of structure. In the process of hologram reconstruction, numerical algorithms were developed based on the angle spectrum theory. In the experiments of marine plankton, some technical issues, such as reconstruction algorithm, numerical refocusing, zero-order term suppression, were discussed. We can obtain the reconstructing image layer by layer at different distances by changing the distance step, which demonstrates that digital holographic imaging is capable of digital refocusing. Digital holographic imaging has clear advantages over other optical methods for the analysis of marine plankton, which contributes to further microorganism identification in the oceanographic observation by using the digital image processing and microscopy techniques.

Abstract:We present a single-image numerical phase retrieval method for Zernike phase-contrast microscopy (ZPM) that addresses halo and shade-off artifacts, as well as the weak phase condition, without requiring hardware modifications. By employing a rigorous physical model of ZPM and a gradient descent algorithm for its inversion, we achieve quantitative ZPM imaging. Our approach is experimentally validated using biological cells and its quantitative nature is confirmed through comparisons with digital holography observations.

Abstract:Holographic imaging is degraded severely by the speckle or incoherent noise. In this work, resampling mask method based on phase-shifting digital holography is proposed to reduce the reconstruction noise. The zero-order and the conjugate term of the hologram can be eliminated with four-step phase-shifting digital holography (PSDH). The complex amplitude of the object after propagation can be calculated by the phase-shifting algorithm. A phase-only spatial light modulator is used to realize PSDH in the experiment. The complex amplitude is encoded into several parts by resampling masks. A high quality reconstruction image with low noise is achieved through the superposition of the individually reconstruction of coded complex amplitudes. The experiment data go well with the asymptotic function. This method can be used for digital holographic imaging of the biological samples and microstructures. Experimental results prove the feasibility of this proposed method.

Abstract:Quantitative phase imaging by digital holographic microscopy (DHM) is a nondestructive and label-free technique that has been playing an indispensable role in the fields of science, technology, and biomedical imaging. The technique is competent in imaging and analyzing label-free living cells and investigating reflective surfaces. Herein, we introduce a new configuration of a wide field-of-view single-shot common-path off-axis reflective DHM for the quantitative phase imaging of biological cells that leverages several advantages, including being less-vibration sensitive to external perturbations due to its common-path configuration, also being compact in size, simple in optical design, highly stable, and cost-effective. A detailed description of the proposed DHM system, including its optical design, working principle, and capability for phase imaging, is presented. The applications of the proposed system are demonstrated through quantitative phase imaging results obtained from the reflective surface (USAF resolution test target) as well as transparent samples (living plant cells). The proposed system could find its applications in the investigation of several biological specimens and the optical metrology of micro-surfaces.

Abstract:Digital holographic microscopy is a promising method for measuring complex micro-structures with high slopes. A quasi-common path interferometric apparatus is adopted to overcome environmental disturbances, and an acousto-optic tunable filter is used to obtain multi-wavelength holograms. However, the phase shifting error caused by the acousto-optic tunable filter reduces the measurement accuracy and, in turn, the reconstructed topographies are erroneous. In this paper, an accurate reconstruction approach is proposed. It corrects the phase-shifting errors by minimizing the difference between the ideal interferograms and the recorded ones. The restriction on the step number and uniformity of the phase shifting is relaxed in the interferometry, and the measurement accuracy for complex surfaces can also be improved. The universality and superiority of the proposed method are demonstrated by practical experiments and comparison to other measurement methods.
