Identification and analysis of the extract isolated from three herbs which induced collagen synthesis

Abstract:Alterations of the extracellular matrix are critical in the aged and damaged skin. Any substances keep extracellular matrix can protect skin from aging and damaging. Type I and type III collagens are predominant components in the dermis. We have investigated increasing effect of the extract isolated from Salvia miltiorrhiza, Astragalus membranaceus, and Angelica on collagen synthesis in human foreskin fibroblasts and mouse embryonic fibroblast. Effect of the extract on the expression of type I and type III collagen genes were measured by quantitative real-time polymerase chain reaction. The extract content was analyzed by the reverse-phase isocratic mode of high performance liquid chromatography. Real-time polymerase chain reaction results showed that the extract significantly stimulated type I and type III collagen expression in both human foreskin fibroblasts and mouse embryonic fibroblast. High performance liquid chromatography analysis revealed that the extraction process was stabilized and reduplicative. Three characteristic peaks could be used as quality control. The present study suggested that the extract from Salvia miltiorrhiza, Astragalus membranaceus, and Angelica could be effective against aged and damaged skin since its activity on inducing type I and type III collagen synthesis. © 2012 American Scientific Publishers.
