Abstract:Background: One of the most common pathological conditions in the gastrointestinal (GI) tract is increased incidence of malignancy with advancing age. Earlier studies from this laboratory have demonstrated that proliferative activity in the colonic mucosa increases with advancing age, accompanied by a parallel rise in the expression and activation of EGFR. The underlying mechanisms for the age-related rise in GI malignancy, particularly colorectal cancer are poorly understood. Recent evidence suggests that colorectal cancer is driven by self-renewing cancer stem-like cells (CSCs). Earlier, we reported that the age-related increase in adenomas in the colon was associated with increased expression of a number of CSC markers and EGFR in microscopically normal mucosa suggesting a role for CSCs in the development of colorectal neoplasia. We hypothesized that the increase in CSCs and EGFR are related. Methods: Biopsies from colon cancer, adenomatous polyps and normal mucosa from subjects, aged between 40 and 80 years were analyzed for the levels of several markers of CSCs as well as for phospho-EGFR and/or microRNAs by (a) immunohistochemistry, (b) FACS analysis, (c) RT-PCR and/or (d) Western-blot. Results: Analysis of colon cancer specimens revealed a high level of co-expression of total and phospho-EGFR with CD166 (one of the CSC markers). The same phenomenon was also observed in adenomas as well as in normal appearing mucosa of patients with adenomatous polyps. Moreover, co-expression of phospho-EGFR and CD166 in the colonic mucosa was found to be significantly higher in patients with adenomas over 60 years of age than their younger counterparts. In addition, FACS analysis of cells isolated from colonic biopsies of patients with adenomas over 60 years of age revealed a higher proportion of cells expressing CD44+CD166than those below 60 years. This increase was associated with elevated levels of microRNA-21, which is known to regulate the expression of EGFR. Conclusion: Our data suggest that aging leads to an increase in CSCs in the colonic mucosa which is associated with an increase in EGFR expression and this may play a pivotal role in the age-related rise in colorectal cancer.

Identification of STAT3 as a biomarker for cellular senescence in liver fibrosis: A bioinformatics and experimental validation study

Abstract:Background: Cellular senescence is associated with a dysregulated inflammatory response, which is an important driver of the development of liver fibrosis (LF). This study aimed to investigate the effect of cellular senescence on LF and identify potential key biomarkers through bioinformatics analysis combined with validation experiments in vivo and in vitro. Methods: The Gene Expression Omnibus (GEO) database and GeneCards database were used to download the LF dataset and the aging-related gene set, respectively. Functional enrichment analysis of differential genes was then performed using GO and KEGG. Hub genes were further screened using Cytoscape's cytoHubba. Diagnostic values for hub genes were evaluated with a receiver operating characteristic (ROC) curve. Next, CIBERSORTx was used to estimate immune cell types and ratios. Finally, in vivo and in vitro experiments validated the results of the bioinformatics analysis. Moreover, molecular docking was used to simulate drug-gene interactions. Results: A total of 44 aging-related differentially expressed genes (AgDEGs) were identified, and enrichment analysis showed that these genes were mainly enriched in inflammatory and immune responses. PPI network analysis identified 6 hub AgDEGs (STAT3, TNF, MMP9, CD44, TGFB1, and TIMP1), and ROC analysis showed that they all have good diagnostic value. Immune infiltration suggested that hub AgDEGs were significantly associated with M1 macrophages or other immune cells. Notably, STAT3 was positively correlated with α-SMA, COL1A1, IL-6 and IL-1β, and was mainly expressed in hepatocytes (HCs). Validation experiments showed that STAT3 expression was upregulated and cellular senescence was increased in LF mice. A co-culture system of HCs and hepatic stellate cells (HSCs) further revealed that inhibiting STAT3 reduced HCs senescence and suppressed HSCs activation. In addition, molecular docking revealed that STAT3 was a potential drug therapy target. Conclusions: STAT3 may be involved in HCs senescence and promote HSCs activation, which in turn leads to the development of LF. Our findings suggest that STAT3 could be a potential biomarker for LF.

Abstract:Dysregulation of the aldehyde dehydrogenase (ALDH) family has been implicated in various pathological conditions, including cancer. However, a systematic evaluation of ALDH alterations and their therapeutic relevance in hepatocellular carcinoma (HCC) remains lacking. Herein, we found that 15 of 19 ALDHs were transcriptionally dysregulated in HCC tissues compared to normal liver tissues. A four gene signature, including ALDH2, ALDH5A1, ALDH6A1, and ALDH8A1, robustly predicted prognosis and defined a high-risk subgroup exhibiting immunosuppressive features like regulatory T cell (Tregs) infiltration. Single-cell profiling revealed selective overexpression of tumor necrosis factor receptor superfamily member 18 (TNFRSF18) on Tregs, upregulated in high-risk HCC patients. We identified ALDH2 as a tumor suppressor in HCC, with three novel phosphorylation sites mediated by protein kinase C zeta that enhanced enzymatic activity. Mechanistically, ALDH2 suppressed Tregs differentiation by inhibiting β-catenin/TGF-β1 signaling in HCC. Collectively, our integrated multi-omics analysis defines an ALDH-Tregs-TNFRSF18 axis that contributes to HCC pathogenesis and represents potential therapeutic targets for this aggressive malignancy.

Abstract:Recent studies have discovered aging-associated changes of adult stem cells in various tissues and organs, which potentially contribute to the organismal aging. However, aging-associated changes of liver progenitor cells (LPCs) remain elusive. Employing young (2-month-old) and old (24-month-old) mice, we found diverse novel alterations in LPC activation during aging. LPCs in young mice could be activated and proliferate upon liver injury, whereas the counterparts in old mice failed to respond and proliferate, leading to the impaired liver regeneration. Surprisingly, isolated LPCs from young and old mice did not exhibit significant difference in their clonogenic and proliferative capacity. Later, we uncovered that the decreased activation and proliferation of LPCs were due to excessive reactive oxygen species produced by neutrophils infiltrated into niche, which was resulted from chemokine production from activated hepatic stellate cells during aging. This study demonstrates aging-associated changes in LPC activation and reveals critical roles for the stem cell niche, including neutrophils and hepatic stellate cells, in the negative regulation of LPCs during aging.

Abstract:T cells accumulate in adipose tissue (AT) and change phenotypes with obesity, but the mechanisms for these changes and their contribution to obesity and metabolic functions, particularly related to aging, remain poorly defined. STAT1 is a transcription activator that mediates cellular responses to interferons (IFNs) and other cytokines. Hence, we sought to determine the role of STAT1 in AT T cell phenotypic changes and obesity with aging. Mice with specific Stat1 knockout in αβT cells (tStat1-KO) were generated by crossbreeding Stat1fl/fl mice with CD4-cre mice. Notably, with aging, tStat1-KO mice compared to wild-type (WT) littermates exhibited less obesity, with reduced body weight (31.80 ± 4.6 g in tStat1-KO [n=28] vs 36.21 ± 3.23 g in WT [n=22] at the age of 50 weeks [on normal chow], P<0.01) and decreased fat mass (4.56 ± 0.96 g in tStat1-KO [n=9] vs 8.63 ± 2.91 g in WT [n=4], P<0.05), and improved insulin sensitivity and glucose tolerance indicated by insulin- or glucose-tolerance test. Western blot showed that UCP1 levels were increased in subcutaneous AT of tStat1-KO compared to WT mice at the age of 50 weeks. Flow cytometric analysis indicated that compared to young (~12-week-old) WT mice, aged (~50-week-old) WT mice had higher percentages of total CD3+ T cells and CD4+ and CD8+ T cells in AT stromal vascular fractions (SVF) and even higher percentage of regulatory T cells (Treg) in CD4+ T cells (n=5-12/group, P<0.05). The percentages of total CD3+ T cells and CD8+ cells, but not CD4+ T cells, in AT SVF were even higher in aged tStat1-KO than WT mice (P<0.05). In contrast, the percentage of Treg in CD4+ cells tended to be lower in AT of aged tStat1-KO than of aged WT (P=0.08). The proportions of macrophages in AT were not significantly different among young WT, aged WT, and aged tStat1-KO mice. In summary, T cell knockout of Stat1 in mice reduced aging-associated obesity and improved insulin resistance, with changes in AT T cells, particularly CD8+ T cells and Treg. Disclosure Z.
