Abstract:Sulfated polysaccharides are polysaccharide of which hydroxys of single sugar molecules are replaced by sulfate groups.In recent years,sulfated polysaccharides have aroused more and more concern for their anticoagulant activity and lower side effects compared with heparin.There are two forms of sulfated polysaccharides-natural sulfated polysaccharides and chemically sulfated polysaccharides,both of which have good anticoagulant activity.As the core of improving their functional properties,the structure-activity relationship of sulfated polysaccharides in anticoagulation is extensive studied around world.The sulfate methods,the anticoagulant activity of sulfated polysaccharides and mechanisms are reviewed.
Preliminary Study on Extraction and Purification of Chondroitin Sulfate from Monopterus albus Bone

Abstract:On the basis of dilute alkali-enzyme hydrolysis,chondroitin sulfates were extracted from Monopterus albus bones assisted with ultrasonic wave breaking.The effects of three ways to de-protein were compared,as well as the fractionated precipitation experiment of polysaccharide.Then the crude products were got to further purification by ion-exchange chromatography(CM22) and dialysis.The results showed that 5%TCA had better effect on de-protein after enzymolysis,and two volumes of ethanol were added to precipitate most chondroitin sulfates.Purity determination indicated that the pure products contained sulfate groups,and the content of CHS reached to 92.31%.Agar gel electrophoresis and infrared absorption spectroscopy showed that the purified sample was similar with the standard chondroitin sulfate,so we can confirm that the pure products were chondroitin sulfate polysaccharides.

Abstract:A new RP-HPLC method was established for the determination of chondroitin sulfate,and chon-droitin sulfate A,B,C,D.A C18 column was used,with the mobile phase of acetonitrile-0.5% phosphoric acid-isopro-panol(15∶85∶1),at the detection wavelength of 209nm,and column temperature of 40℃.The calibration curve was linear in the concentration range of 0.4-2mg/ml(r = 0.9997).The average recovery was 99.3%,with RSD of 1.16%.

Abstract:Dermatan sulfate (DS) is one of the hardest impurities to remove from heparin products due to their high structural similarity. The development of a sensitive and feasible method for quantitative detection of DS in heparin is essential to ensure the clinical safety of heparin pharmaceuticals. In the current study, based on the substrate specificity of chondroitin B lyase, ultraviolet spectrophotometric and strong anion-exchange high-performance liquid chromatographic methods were established for detection of DS in heparin. The former method facilitated analysis in heparin with DS concentrations greater than 0.1mgmL(-1) at 232nm, with good linearity, precision and recovery. The latter method allowed sensitive and accurate detection of DS at concentrations lower than 0.1mgmL(-1), exhibiting good linearity, precision and recovery. The linear range of DS detection using the latter method was between 0.01 and 0.5mgmL(-1).

Abstract:Heparins are sulfated polysaccharides with a heterogeneous mixture derived from animal tissues, subject to supply limitations and the risk of animal virus residues. Patients using heparin also face the risks of spontaneous bleeding and thrombocytopenia. Here we reported an efficient riclinoctaose-based approach for rapid chemical synthesis of a structurally defined heparin-like anticoagulant sulfated octasaccharide (SRO). We used sulfur trioxide-pyridine, sulfur trioxide-trimethylamine, and sulfur trioxide-triethylamine complexes as solvents for one-pot O-sulfation and determined the optimal conditions for synthesizing SRO. Sulfur trioxide-trimethylamine provided reasonable control over the degree of substitution between 1.85 and 1.88, revealing a single molecule with a theoretical molecular weight of 2952.96 g/mol. The structural features of the SRO were carried out by Fourier transform infrared spectroscopy and one- and two- dimensional 1 H and 13 C NMR analysis, revealing sulfation repeatedly present at the fixed positions of C-6/C-2/C-3 and reducing terminals. The anticoagulant activity of SRO was demonstrated by efficiently blocking coagulation in the blood of mice and human. SRO dose-dependently decreased ferric chloride-induced experimental thrombosis in mice. Like heparin, SRO specifically inhibits coagulation factor Xa, but significantly reduces the risk of bleeding compared to heparin. Therefore, we named it octaparin. These results support that octaparin is expected to replace animal-sourced heparin.

Abstract:Chondroitin sulfate (CCS) was purified from discarded codfish (Gadus macrocephalus) bones, and its chemical structure and anticoagulant activity were assessed. CCS was obtained via enzymatic lysis and ion-exchange column chromatography, with a yield of approximately 0.15%. High-performance gel performance chromatography revealed CCS to be a largely homogeneous polysaccharide with a relatively low molecular weight of 12.3 kDa. FT-IR spectroscopy, NMR spectroscopy, and SAX-HPLC indicated that CCS was composed of monosulfated disaccharides (ΔDi4S 73.85% and ΔDi6S 19.06%) and nonsulfated disaccharides (ΔDi0S 7.09%). In vitro anticoagulation analyses revealed that CCS was able to significantly prolong activated partial thromboplastin time (APTT) and thrombin time (TT) (p < 0.05). At a CCS concentration of 5 μg/mL and 25 μg/mL, APTT and TT were approximately 1.08 and 1.12 times higher, respectively, compared to the negative control group. The results indicated that CCS might offer value as a dietary fiber supplement with the potential to prevent the incidence of coagulation-related thrombosis.

Abstract:The aim is to compare the structures, anticoagulant and antithrombotic activities of two fucosylated chondroitin sulfates isolated from sea cucumbers Isostichopus badionotus (fCS-Ib) and Pearsonothuria graeffei (fCS-Pg), which were known to have different sulfation patterns on the fucose branches.The structures of fCSs were identified using 2D NMR. Anticoagulant activities were measured by activated partial thromboplastin time (APTT) and thrombin time (TT), and inhibition of factors IIa, Xa and XIIa was assessed in vitro. Antithrombotic activity was determined ex vivo by measuring the length and weight of the thrombus generated.The two fCSs had identical chondroitin sulfate E backbones and similar fucose branches, but different sulfation patterns of the fucose branches. The fucose branch in fCS-Ib was mainly 2,4-O-sulfated whereas that in fCS-Pg was mainly 3,4-O-sulfated. In vitro assay indicated that fCS-Pg possessed much lower potency than fCS-Ib in prolonging APTT/TT and in inhibition of thrombin. However, they both exhibited similar inhibitory effects on factor X activation by intrinsic tenase complex, and on thrombus generation. Furthermore, both fCSs significantly activated factor XII, which has been proved to be associated with adverse clinical events associated with heparin contaminated by oversulfated chondroitin sulfate.The 2,4-O-sulfated fucose branch is the key structural factor of fCSs for prolonged APTT/TT and inhibition of thrombin, whereas the inhibitory effect of fCSs on factor X, XII activation and thrombus generation was attributed to the overall structure of fCS polysaccharide. GENERAL IMPORTANCE: Both fCSs have well defined structures and can be readily quality-controlled, and therefore may be potential alternatives for heparin as anticoagulant and antithrombotic drugs.

Abstract:In this paper, a combination of hot-pressure, enzymatic hydrolysis and membrane separation process is used for efficiently and environmentally friendly extraction of chondroitin sulfate (CS) from large hybrid sturgeon cartilage, namely, HPCS. The recovery and yield of CS were 93.68% and 36.47% under the optimized conditions. Fourier transform infrared (FT-IR) spectroscopy, nuclear magnetic resonance (NMR) spectroscopy and high-performance liquid chromatography (HPLC) indicated that the HPCS was composed of monosulfated disaccharides in position 6 and 4 of the N-acetyl-D-galactosamine (58.38% and 27.34%, respectively) and nonsulfated disaccharide (14.29%), which was similar to the composition of CS extracted by dilute alkali-enzymatic hydrolysis-chemical precipitation from large hybrid sturgeon cartilage (SCS). The wound healing results indicated that HPCS could promote cell migration and proliferation, alleviate inflammation and facilitate angiogenesis, which results in its excellent wound treatment activity. These results provide theoretical and practical significance for the production and application of chondroitin sulfate.

Abstract:Sulfate in sea cucumber polysaccharide(crude sample),sea cucumber chondroitin sulfate(SC-CHS)and sea cucumber fucan(SC-FUC)were hydrolyzed by TFA,separated and determinated by ICS-2000 system.The average recovery was 100.90%,RSD=2.12%,the detection limitation reached 3.9×10-2μg/mL.Compared with the traditional determination method of sulfate,this method was reproducible,much simple and more accurate.The method applied to quantitative determination of sulfate in other sulfated polysaccharides.The results showed that sulfate in all kinds of SC-CHS had a higher content than in SC-FUC,characteristical differences of sulfate content existed among different kinds of sea sucumber polysaccharide.

Abstract:A type of water-soluble carboxymethyl chitosan (O--CMC), with 76% degree of substitution determined by conductivity method, was prepared by using chloroacetic acid to react with C6-OH of chitosan. The solubility of O--CMC was characterized also. Animal experiment in rabbits showed that O--CMC could lubricate arthron, inhibit proliferation of fibroblast cells on rabbits' knee joints, benefit the process of repairing pathologic articular cartilage, and produce good therapeutic effect on rheumatoid arthritis.
Heparan sulfate functions are altered in the osteoarthritic cartilage

Abstract:Background: Heparan sulfate (HS) proteoglycans (PG) may be found at the chondrocyte surface and in the pericellular cartilage matrix, and are involved in cell-cell and cell-matrix interactions. An important function of HS chains is to regulate cell fate through specific interactions with heparin-binding proteins (HBP) modulated by their complex sulfation pattern. Osteoarthritis (OA) is a joint disorder characterized by the degradation of articular cartilaginous extracellular matrix. The aim of this study was to investigate HS structure and functions in osteoarthritic cartilages compared to normal cartilages (controls). Methods: Glycosaminoglycans (GAG) were extracted from human macroscopically normal cartilages (controls, n = 7) and (OA cartilages n = 11). HS were isolated and quantified using the DMMB quantification method. Their structure and functions were then compared using respectively a HPLC analysis and HBP binding tests and their phenotypic effects on murine chondrocytes were studied by RQ-PCR. Statistical analyzes were performed using a one-way ANOVA followed by a Dunnett's test or a t test for pairwise comparisons. Results: In OA, HS were characterized by increased sulfation levels compared to controls. Moreover, the capacity of these HS to bind HBP involved in the OA pathophysiological process such as FGF2 and VEGF was reduced. Chondroitin sulfates and keratan sulfates regulated these binding properties. Finally, HS from OA cartilages induced the mRNA levels of catabolic markers such as MMP3, MMP13, and TS4 and inhibited the mRNA levels of anabolic markers such as COL2, ACAN, SOX9, and VEGF in murine articular chondrocytes. Conclusion: The sulfation of HS chains was increased in OA cartilages with changes in HBP binding properties and biological effects on chondrocyte phenotypes. Thus, modified HS present in altered cartilages could be a novel therapeutic target in OA.
Effect of oversulfation on the chemical and biological properties of fucoidan

Abstract:Fucoidan was sulfated using chlorosulfonic acid–pyridine complex and isolated as the sodium salt. Infrared analysis of the native and sulfated fucoidans gave identical results in respect to the O–H stretching, hemiacetal stretching and SO stretching. Absorption around 840cm−1 was also present in both the fucoidans representing the sulfate at the axial C-4 position except for a shoulder at 820cm−1, which was present only in the sulfated compound indicating the presence of sulfate groups at the equatorial C-2 position. The sulfated compound showed four times higher anticoagulant activity in doubling prothrombin time of normal citrated human plasma in comparison with native fucoidan. Earlier studies using native fucoidan showed that the activation of glutamic plasminogen (Glu-Plg) by tissue plasminogen activator (t-PA) or by urokinase (u-PA) was enhanced when the in vitro studies were conducted using 0.05M Tris buffer (pH 7.4) and that the addition of physiological concentration of NaCl (0.9%) to the buffer reversed the activation. The results of the current studies showed that sulfated fucoidan gave higher stimulations of Glu-Plg activation by u-PA and by t-PA in comparison to native fucoidan when the in vitro studies are conducted using Tris buffer containing 0.9% NaCl.

Determination of chondroitin sulfate in synovial fluid and drug by ratiometric fluorescence strategy based on carbon dots quenched FAM-labeled ssDNA

Abstract:Chondroitin sulfate (CS) plays an increasingly important role in clinical settings and pharmacy quality control. However, sensitive and simple methods for CS detection remain limited. In this work, positively charged nitrogen doped carbon dots (P-NCDs) with internal luminescence and quenching property to FAM-labeled random-sequence ssDNA (F-ssDNA) were prepared by a simple heating method. P-NCDs attached and quenched F-ssDNA through electrostatic interaction to form the system of P-NCDs and F-ssDNA (P-NCDs/F-ssDNA) with retained fluorescence intensity of P-NCDs. The highly negatively charged CS reacted electrostatically with P-NCDs and then replaced F-ssDNA in P-NCDs/F-ssDNA to recover the fluorescence intensity of the original quenched F-ssDNA while retaining the internal fluorescence intensity of P-NCDs. Thus, by using restored F-ssDNA as the signal controlled by adding CS to P-NCDs/F-ssDNA, a ratiometric fluorescence strategy based on the retained fluorescence of P-NCDs as reference signal was fabricated through synchronous fluorescence spectrometry for the sensitive detection of CS. Under the optimal experimental conditions, a linear equation for CS was obtained for CS concentration within the range of 0.05–2.0 μg/mL. The method was also successfully applied for the accurate determination of CS in joint fluid samples of arthritic patients, chondroitin sulfate tablets, and chondroitin sulfate eye drops, suggesting its appreciable application potential in the clinic.

Abstract:The method for preparing low molecular weight fucosylated chondroitin sulfate from sea cucumber Isostichopus badionotus using partial acid hydrolysis was reported, and its hydrolysis mechanism was also investigated. The sea cucumber chondroitin sulfate FCS was hydrolyzed under different conditions (80 degrees C 3 h and 6 h), then isolated and purified on a Bio-P-4 geltration to prepare low molecular weight fractions (LMWF-FCS). The chemical compositions of LMWF-FCS showed the branched fucose (Fuc) was cleaved during acid hydrolysis process, whereas the mole ratio of acetyl-galactosamine (GalNAc) and glucuronic acid (GlcA) in the backbone remained the same, which indicated the backbone was a typical chondroitin sulfate structure. The disaccharide composition analysis of LMWF-FCS suggested that the sulfation patterns of GalNAc in the backbone chain changed and the substitution value was reduced. Furthermore, the D-1 NMR analysis illustrated the branched-Fuc was cleaved during acid hydrolysis, but their substitution patterns were not influenced, which was distinct from the previous reports that the substitutions of branched-Fuc in FCS were easy to change. Simultaneously, the sulfation pattern of GalNAc in backbone chain changed obviously in the acid hydrolysis process. The anticoagulant activity in vitro illuminated the anticoagulant activity of the degradation products over time in the acid hydrolysis are gradually declined, but still kept good. Therefore, the LMWF-FCS prepared could be developed as a new anticoagulant and antithrombotic drug like low molecular weight heparin.
Depolymerization of Fucosylated Chondroitin Sulfate with a Modified Fenton-System and Anticoagulant Activity of the Resulting Fragments.

Abstract:Fucosylated chondroitin sulfate (fCS) from sea cucumber Isostichopus badionotus (fCS-Ib) with a chondroitin sulfate type E (CSE) backbone and 2,4-O-sulfo fucose branches has shown excellent anticoagulant activity although has also show severe adverse effects. Depolymerization represents an effective method to diminish this polysaccharide's side effects. The present study reports a modified controlled Fenton system for degradation of fCS-Ib and the anticoagulant activity of the resulting fragments. Monosaccharides and nuclear magnetic resonance (NMR) analysis of the resulting fragments indicate that no significant chemical changes in the backbone of fCS-Ib and no loss of sulfate groups take place during depolymerization. A reduction in the molecular weight of fCS-Ib should result in a dramatic decrease in prolonging activated partial thromboplastin time and thrombin time. A decrease in the inhibition of thrombin (FIIa) by antithromin III (AT III) and heparin cofactor II (HCII), and the slight decrease of the inhibition of factor X activity, results in a significant increase of anti-factor Xa (FXa)/anti-FIIa activity ratio. The modified free-radical depolymerization method enables preparation of glycosaminoglycan (GAG) oligosaccharides suitable for investigation of clinical anticoagulant application.
Chemical O-sulfation of N-sulfoheparosan: a Route to Rare N-sulfo-3-O-sulfoglucosamine and 2-O-sulfoglucuronic Acid.

Abstract:Heparosan, the capsular polysaccharide of E. coli K5 is currently used as the starting material in the chemoenzymatic synthesis of heparan sulfate and the structurally related anticoagulant drug heparin. Base hydrolysis of N-acetyl groups and their subsequent N-sulfonation, are used to prepare N-sulfoheparosan an intermediate of biosynthesis. In the present study, when excess sulfonation reagent was used during N-sulfonation, some O-sulfation also took place in the N-sulfoheparosan product. After a nearly full digestion, a hexasaccharide fraction exhibited resistance to heparin lyase II. Excessive digestion by heparin lyase II and structural identification by NMR and mass spectroscopy indicated that the resistant hexasaccharide fraction has two structures, ΔUA-GlcNS-GlcA2S-GlcNS-GlcA-GlcNS and ΔUA-GlcNS-GlcA- GlcNS3S-GlcA-GlcNS in similar amounts. The 2-sulfated structure exhibited partial resistance to heparin lyase II; however the structure of ΔUA-GlcNS-GlcA-GlcNS3S was completely resistant to heparin lyase II.

Abstract:Chondroitin sulfate (CS) is widely known for its various biological activities which are closely related to the sulfate substitution and the molecular weight. Effective degradation methods without striping sulfate groups are in a need. In the present study, a photocatalytic degradation method using H2O2 and TiO2 has been developed and it could decrease the average molecular weight of CS into 5 kDa within 6 h. The chemical composition of CS before and after degradation were compared by FT-IR, NMR, etc., and no removement of sulfate group was observed. Then the identification of the oligosaccharides in the degradation product by mass spectroscopy revealed that glucuronic acid or its derivative, arabinuronic acid, was at most of the reducing ends, and the depolymerization mechanism was proposed. Furthermore, the absorption of CS in rats was enhanced by the degradation while the excertion profile of the degradation product was similar to that of CS.

Investigation of the biological functions of heparan sulfate using a chemoenzymatic synthetic approach

Abstract:Chitosan was prepared by alkaline N-deacetylation of β-chitin and hydroxypropyl derivatives with different degrees of substitution (DS) were synthesized. It was characterized by Fourier transform infrared (FT-IR) and elemental analysis. The DS of hydroxypropyl chitosan (HPCS) calculated by an element analyzer were 0.42, 0.75, 1.20, 1.82 and 2.25. HPCS showed better foam capacity and stability than that of chitosan, and the effectiveness correlated well with the DS of HPCS. The highest bile acid-binding capacity of all five HPCS reached 56.02 mg/g, which was 4.0-fold higher than that of chitosan. The scavenging ability of HPCS against hydroxyl and ABTS radicals improved with increasing concentration. The correlation between the hydroxypropyl content (DS) of HPCS and scavenging ABTS radical ability was positive. The hydroxyl radicals scavenging activity of HPCS correlated well with its increasing concentration, and EC50 values were below 12.5 mg/mL. These results indicated that hydroxypropylation is a possible approach to obtain chitosan derivatives with desirable physiochemical properties.

Abstract:Heparan sulfate (HS) is a class of linear, sulfated, anionic polysaccharides, called glycosaminoglycans (GAGs), which present on the mammalian cell surfaces and extracellular matrix. HS GAGs display a wide range of critical biological functions, particularly in cell signaling. HS is composed of repeating units of 1 → 4 glucosidically linked uronic acid and glucosamine residues. Heparin, a pharmacologically important version of HS, having higher sulfation and a higher content of iduronic acid than HS, is a widely used clinical anticoagulant. However, due to their heterogeneity and complex structure, HS and heparin are very challenging to analyze, limiting biological studies and even resulting in safety concerns in their therapeutic application. Therefore, reliable methods of structural analysis of HS and heparin are critically needed. In addition to the structural analysis of heparin, its concentration in blood needs to be closely monitored to avoid complications such as thrombocytopenia or hemorrhage caused by heparin overdose. This review summarizes the progress in biotechnological approaches in the structural characterization of HS and heparin over the past decade and includes the development of the ultrasensitive approaches for detection and measurement in biological samples.
