Abstract:Reaction of nucleoside triphosphates (NTPs) with amines in pyridine mediated by trimethylsilyl chloride produced nucleoside 5'-phosphoramidates in moderate yields without any preprotection of nucleosides and amino acid methyl esters. The reaction pathway is very similar to the mechanism of the RNA capping reaction, DNA or RNA ligation reaction, and catalysis of hydrolases and nucleases involving the formation of covalent enzyme-NMP (nucleoside 5'-monophosphate) intermediates in biological systems, which could provide a valuable clue for the enzymatic reactions.

Abstract:A new catalytic enantioselective approach for the formation of allyl alpha-amino acid derivatives by reaction of N-tosyl alpha-imino esters with allyl stannanes and silanes catalyzed by chiral copper(I) complexes has been developed. A series of different BINAP and phosphine-oxazoline (P,N) ligands have, in combination with various Lewis acids, been tested as chiral catalysts for allylation of N-tosyl alpha-imino esters. It has been found that both type of ligands, in combination with copper(I) salts, give highly valuable unsaturated alpha-amino acid derivatives. The reaction has been investigated for different allyl stannanes and silanes, and it has been found that tri-n-butyl allyl stannane gives the best results of the simple allyl compounds tested, leading to gamma,delta-unsaturated alpha-amino acid derivatives in up to 94% yield and with up to 83% ee, which can be improved to be >95% ee by recrystallization. The reaction has also been investigated using different acyclic and cyclic allyl stannanes leading to various types of unsaturated alpha-amino acid derivatives in very high yield (up to 95%) and with up to 98% ee. The stereochemistry and absolute configurations of the allyl alpha-amino acid derivatives have been determined by X-ray analysis, and it is suggested that the reaction takes place as an ene-like reaction.

Abstract:An approach to various phosphoramidates has been developed through one-pot chemoselective reaction of nucleoside di- and triphosphates with 3'-amino-3'-deoxythymidine, D-glucosamine, and peptides mediated by trimethylsilyl chloride, and the corresponding conjugates via the linkage of phosphate were obtained. The method could be generally applied for the synthesis of phosphoramidates without any preprotection of polyphosphates and amines containing multifunctional groups.

Abstract:In the presence of imidazole N-phosphoamino acids were prompted to undergo ester exchanges on the phosphorus as well as N to O migration 10-38 times faster than in its absence. Only phosphoserine and threonine methyl esters, were activated by imidazole to give similar reactions. For the diastereomeric N-phosphonoserine methyl ester containing a phosphorus-carbon bond, a stereoselective reaction occurred to provide an O-phosphonoserine derivative with an isomeric ratio of 65 to 35. The mechanism may involve the formation of pentacoordinate intermediates promoted by imidazole.

Abstract:Amino acid-nucleotide conjugates have important biological functions and therapeutic applications. For example, aminoacyl adenylates are key intermediates in aminoacyl tRNA synthetase reactions. They may also be involved in the prebiotic synthesis of polypeptides. Finally, various amino acid carbomethoxy aryl phosphoramidates of nucleotide prodrugs may be activated through a mechanism involving a pentacoordinated phosphorane intermediates. In order to understand better the chemistry of these compounds, a phenylalanyl adenylate pentacoodinated phosphorane has been synthesized in 72% yield and its decomposition in aqueous solution studied. Hydrolysis gave 2′,3′-O-isopropylidene adenosine 5′-monophosphate, 2′,3′-O-isopropylidene adenosine, and phenylalanine. The results provide model chemistry for the enzymatic degradation mechanism of antiviral aryl amino acid phosphodiester amidates in cells, which leads to their activation.

Abstract:Coupling of N-phosphoamino acids 2 and amino acid methyl esters by dichlorotriphenylphosphorane 3 produced N-phosphopeptide methyl esters 5, which were sequentially hydrolyzed in triethylamine or dilute NaOH aqueous solution. The target products N-phosphodipeptides 6 were obtained in reasonable yields and high purity after isolation using extraction method. The convenient and efficient approach could be generally used for synthesis of polypeptides.

Their geometric stereoisomers were isolated and characterized. The reaction was catalyzed by acetyl chloride and good to excellent yields were obtained under these reaction conditions.

Abstract:An enyne dimer (1) of methyl propiolate was reacted with amines to form dimethyl (E,E)-2-amino-2,4-hexadiene dioates with remarkable chemospecificity, regiospecificity, and stereospecificity. This enyne was also reduced by Ph3P stereospecifically to form dimethyl (E,E)-muconic ester. Hydrogenation of the conjugated amino-diene led to an efficient production of dimethyl α-aminoadipate. A lactam of dimethyl α-aminoadipate was obtained in high yield by simply varying the hydrogenation conditions.

Abstract:The catalytic hydrophosphination of imines represents the most straightforward and atom-economical strategy for the preparation of alpha-aminophosphines, which are useful as biologically active molecules and ligands for catalysis. The first catalytic protocol for the rapid and high-yielding hydrophosphination of common imines enabled by an ene-diamido samarium methoxide catalyst was developed. The reaction allowed the efficient and diastereoselective synthesis of various bis(alpha-amino)phosphines through the double addition of PhPH2 to common imines. The catalytic reaction has a relatively wide substrate scope and offers high diastereoselectivities (up to 99: 1 dr for meso products) and high yields (up to 97%). DFT calculations disclosed an inner-sphere mechanism involving unprecedented activation of a Sm-O bond by a phosphine and stabilization of the addition intermediates by the metal center.

Alkylthiophosphoramidates derivatives were synthesized y reaction of O-isopropyl phosphorodichloridothioate with an amino acid este followed by hydrolysis in 2M NH 4 OH. Abstract:α-Aminophosphonic acids are phosphorus analogues of α-amino acids. Compounds of this type find numerous applications in medicine and crop protection due to their unique biological activities. A crucial factor in these activities is the configuration of the stereoisomers. Only a few methods of stereoselective transformation of α-amino acids into their phosphorus analogues are known so far and all of them are based on asymmetric induction, thus involving the use of a chiral substrate. In contrast, we have focused our efforts on the development of an effective method for this type of transformation using a racemic mixture of starting N-protected α-amino acids and a chiral catalyst. Herein, a simple and efficient stereoselective organocatalytic α-amidoalkylation of dimethyl phosphite by 1-(N-acylamino)alkyltriphenylphosphonium salts to enantiomerically enriched α-aminophosphonates is reported. Using 5 mol% of chiral quinine- or hydroquinine-derived quaternary ammonium salts provides final products in very good yields up to 98% and with up to 92% ee. The starting phosphonium salts were easily obtained from α-amino acid derivatives by decarboxylative methoxylation and subsequent substitution with triphenylphosphonium tetrafluoroborate. The appropriate self-disproportionation of enantiomers (SDE) test for selected α-aminophosphonate derivatives via achiral flash chromatography was performed to confirm the reliability of the enantioselectivity results that were obtained.

Abstract:The isomeric α-amino acyl adenylates and amino acid phosphoramidates of adenosine were synthesized and analyzed in detail by electrospray ionization tandem mass spectrometry (ESI-MS(n)). In ESI-MS/MS of α-amino acyl adenylates, the novel rearrangement ion [cAMP-H](-) observed as the most intense signal was formed through the pentacoordinate phosphorus intermediate with a six-membered ring by nucleophilic attack of the 3'-hydroxyl group on the phosphorus atom. In contrast, for the amino acid phosphoramidate of adenosine, the phosphorus atom could be attacked not only by the carboxylic group to form the cyclic aminoacyl phosphoramidates (CAPAs), but also by the nitrogen atom on the nucleobase leading to intramolecular phosphoryl group migration. It was found that the sodium ion having multidentate binding ability played an essential role in this characteristic rearrangement. The proposed mechanisms were supported by the MS/MS study, deuterium-labeled experiments, high-resolution tandem mass spectrometry and moderate calculations at the B3LYP/6-31G* level. The characteristic fragmentation patterns of α-amino acyl phosphates and amino acid phosphoramidates allows identification of stereoisomers when either the phosphorylation is at the N-terminus or C-terminus of amino acids.

Abstract:The phosphorylation of adenosine withtrimetaphosphate in solution, in solid phase and usingwet-dry cycles was carried out and it was found thatwet-dry cycles were the most efficient. The catalytic effectsof some metal ions on the phosphorylation were alsostudied and it was discovered that Ni(II) is the mosteffective. The combination of wet-dry cycles (4 cycles)and catalysis by Ni(II) led to an unprecedented highconversion of adenosine to phosphorylated products (30%)near neutral pH. The main phosphorylated products were2', 3'-cyclic AMP (10.4%) and 5'-ATP (13.0%).

Abstract:Both C-2- and C-3-symmetric proline-derived beta-amine alcohol ligands were designed, synthesized, and successfully applied to the enantioselective direct addition of trimethylsilylacetylene to N-phosphinoylimines. Aromatic, heteroaromatic, and aliphatic N-(diphenylphosphinoyl) imines and several N-(diethoxyphosphoryl) imines were tested, and optically active propargylic amides in good yields (up to 92%) and excellent enantioselectivities (up to 95%) were obtained by the simple experimental procedure. The convenience, mild conditions, and easy deprotection of the phosphonamide products made the present method very attractive. Furthermore, the Michael-type addition process of C=N alkynylation was studied and proposed on the basis of React P-31 NMR investigation.

Abstract:The oligouridylates formation with N-(O,O-diisopropyl)-phosphoryl alanine in the presence of poly(beta-(N-7-adeninyl)ethyl methacrylate) was studied. An increased yield by the presence of the polymeric nucleic acid analog was confirmed by anion exchange column HPLC, C18 reverse phase HPLC, and turbo ionspray MS.

Abstract:The mass spectral behavior of amino acid methyl ester phosphoramidate derivatives have been investigated using electrospray ionization multistage mass spectrometry (ESI-MSn) and moderate theoretical calculations at the B3LYP/6-31G(d) level. A novel methyl group migration to the phosphoryl group with the formation of the intermediate cyclic aminoacylphosphoramidate was found. The proposed structures of the rearrangement ions were confirmed by high-resolution tandem mass spectrometry. A possible mechanism involving the pentacoordinate phosphoric-carboxylic phosphate anhydride was proposed, in which the metal ion coordination with the phosphoryl and carbonyl groups and the intrinsic properties of phosphoryl group might be the key factors responsible for this novel migration. Abstract:Various alpha,beta-unsaturated (E)-alkenylphosphonamidates were prepared in excellent stereo- and regioselectivities via the electrophilic hydrophosphonylation of cyclic and acyclic aldimines with alkylphosphonochloridates in the presence of base. For unsaturated aldimines, only their respective imine bonds were hydrophosphonylated regiospecifically. The reaction provided a new strategy for the synthesis of (E)-alk-1-enylphosphonamidates via a tandem phosphorylation, deprotonation, and intramolecular 1,5-hydride transfer sequence process from readily available aldimine and alkylphosphonochloridate starting materials. The strategy established the first one-pot electrophilic rather than nucleophilic hydrophosphonylation of aldimines without external hydride donors.

Abstract:S-adenosylmethionine-dependent methyltransferases are involved in countless biological processes, including signal transduction, epigenetics, natural product biosynthesis, and detoxification. Only a handful of carboxylate methyltransferases have evolved to participate in amide bond formation. In this report we show that enzyme-catalyzed F-methylation of carboxylate substrates produces F-methyl esters that readily react with N- or S-nucleophiles under physiological conditions. We demonstrate the applicability of this approach to the synthesis of small amides, hydroxamates, and thioesters, as well as to site-specific protein modification and native chemical ligation.
