Abstract:The chemiluminescence of a luminol-H(2)O(2) system is found to be remarkably enhanced by the Ce(IV) complexes of EDTA-bridged cyclodextrin dimers. The dimers were proved to work much more efficiently than the corresponding monomer. The cavity shape of cyclodextrin moieties and their cooperation displayed an important role in amplifying the chemiluminescence. Further modification of either the cyclodextrin rims or the EDTA linker altered significantly the catalytic abilities of the cyclodextrin dimers, and the examination of the effect of substituents on the chemiluminescence outputs suggested that the proximity between the cyclodextrin cavity and the metallic center might account for the amelioration of the chemiluminescence output.

Abstract:CdSe/ZnS QDs enable the optical probing of the biocatalytic oxidation of tyrosine derivatives and of the scission of peptides by thrombin. CdSe/ZnS QDs were modified with tyrosine methyl ester or with a tyrosine-containing peptide. The tyrosine units were reacted with tyrosinase/O2 to yield the respective l-DOPA and quinone derivatives. The luminescence of QDs modified by the enzyme-generated quinone units is quenched. The quinone-functionalized peptide associated with the QDs was cleaved by thrombin, a process that restored the luminescence of the QDs.

Abstract:Background Chemiluminescence (CL) bioassay is one of the most advanced and used detection method in clinical diagnosis and biomedical research because of the advantages of low background, easy operation, and wide-field imaging without a light source or microscope. The luminol/hydrogen peroxide/horseradish peroxidase (luminol/H 2 O 2 /HRP) system is the most popular CL system, but its application in high-throughput imaging detection is challenged due to its low luminescence efficiency and flash-type emission which is difficult in ensuring the reproducibility and consistency of detection results. Results We reported a glow-type CL system of luminol@CD/H 2 O 2 /HRP by using a supramolecular enhancer of cyclodextrin (CD). This luminol@CD/H 2 O 2 /HRP system exhibited a luminescence lifetime of 41 min for sensitive and accurate imaging analysis. The long-lasting CL emission was attributed to the formation of a 1:1 host–guest complex between luminol and CD, which could stabilize the emitter and effectively reduce nonradiative relaxation. The formation of luminol@CD complex was determined through NMR experiments and theoretical analysis. Under optimum conditions, the luminol@CD/H 2 O 2 /HRP system showed higher sensitivity and much better precision than classical luminol/H 2 O 2 /HRP system for imaging detection of HRP. Especially, this glow-type luminol@CD/H 2 O 2 /HRP system realized CL imaging of microwell arrays on microfluidic chips. In addition, the luminol@CD/H 2 O 2 /HRP system was successfully applied for point-of-care detection of 17 β- estradiol based on a competitive mechanism of host–guest recognition. Significance An efficient CL system is crucial for obtaining reproducible and consistent results for accurate detection. Our luminol@CD/H 2 O 2 /HRP system emitted strong and persistent luminescence, resulting in reliability and efficiency at both CL macroscopic and microscopic imaging detection. We expected the luminol@CD/H 2 O 2 /HRP CL system to be applied in various detection fields.

Abstract:An innovative approach for covalent‐bond‐activated mechanoresponse by complexing rhodamine or spiropyran with cyclodextrin (CD) is reported. This approach endows diverse fluorophores with perfect mechanochromism by introducing a supramolecular system. Unique characteristics such as noncovalent chemical modification and convenient preparation make this approach promising for practical applications. The strong hydrogen bonds provided by CD play a crucial role in triggering the mechanochromic switch. First, the hydrogen bonds seize both sides of the fluorophore's weak chemical bonds and tightly lock the fluorophore in the cavity of CD. Second, the hydrogen bonds prompt the aggregation of complex inclusions in large ordered arrays and strengthen the molecular interactions. In this way, the weak chemical bonds can focus more external force and stretch more easily upon shearing (quantified). This is the first report of supramolecule‐triggered mechanochromic switches. This study opens an avenue to correlate a mechanochemical reaction with a supramolecular system.
Single-atom Nanozymes with Axial Ligand-Induced Self-Adaptive Conformation in Alkaline Medium Boost Chemiluminescence

Abstract:Mimicking the structure of natural enzymes for designing advanced alternatives provides great opportunities to address the bottleneck of enzyme-involved chemiluminescence (CL). Herein, according to theoretical calculations, we found that an endogenous axial ligand of M-N-C single-atom nanozymes (SAzymes), originating from OH− spontaneously bonding to the metal center in an alkaline medium, can self-adaptively change its strength to facilitate intermediate steps. Furthermore, the lowest energy barrier of the rate-determining step and the strongest affinity and fastest electron transfer with luminol anion endow Co-N-C with the highest peroxidase-like activity. Guided by the theoretical calculations, a series of M-N-C SAzymes (M=Fe, Co, Ni) were synthesized to boost CL, where Co-N-C SAzymes with superior catalytic activity and high selective generation of O2•− were validated. As a proof-of-concept, Co-N-C SAzymes were employed for sensitive detection of acetylcholinesterase and organophosphorus pesticide.

Abstract:In view of the shortcomings of the current coreactant electrochemiluminescence (ECL) and inspired by natural oxygen (O2) reduction metalloenzymes, a novel ECL amplification strategy was established. A pyrolytic iron- and nitrogen-doped (Fe-N-C) nanosheet rich in singly ionized oxygen vacancy (VO•) defects was rationally designed by destroying the highly saturated coordination with a preorganized ligand 1,10-phenanthroline-2,9-dicarboxylic acid (PDA). Extraordinary catalytic activity for O2 activation was obtained via screening a special pyrolysis temperature using spectroscopic and electrochemical methods. The high-spin ferric centers of highly dispersed FeC nanoclusters and abundant carbon and oxygen vacancy defects fully contributed to the inherent catalytic activity. ECL amplification was achieved by integrating the material with luminol to generate redox-active radicals in situ from dissolved O2 and simultaneously shorten the transferring distance of radicals. Tetracycline (TC), which posed a growing threat to aquatic biodiversity and environmental safety, as a model antibiotic was successfully detected with a detection limit of 3.88 nM (S/N = 3), clarifying a promising application prospect of this new effective ECL amplification strategy in biological analysis and environmental monitoring.

Abstract:Local enrichment of free radicals at the electrode interface may open new opportunities for the development of electrochemiluminescence (ECL) applications. The sensing platform was constructed by assembling ECL-emitting luminol derived carbon dots (Lu CDs) onto the heterojunction Tungsten disulfide/Covalent organic frameworks (WS2@COF) for the first time, establishing a nanoconfinement-reactor with significantly heightened ECL intensity and stability compared to the Lu CDs-H2O2 system. This enhanced performance is credited to the COF domain's restricted pore environment, where WS2@COF exhibits a more negative adsorption energy for H2O2, effectively enriching H2O2 in the catalytic edge sites of WS2. Furthermore, the internal electric field at the WS2 and COF interface accelerates electron flow, boosting WS2's catalytic activity and achieving domain-limited catalytic enhancement of ECL. Self-designed DNA nanomachines combined with cascading molecular keypad locking mechanisms are integrated into the biosensors, effectively guaranteeing the accuracy of the sensing process while providing crucial safeguards for molecular diagnostics and information security applications. In essence, this innovative approach represents the first system to enhance local free radical concentrations by enriching co-reactants on the electrode surface through nanoconfinement catalysis, yielding heightened ECL luminescence intensity. The potential impact of this novel strategy and sensing mechanism on real-bioanalysis applications is promising.

Abstract:Electrocheluminescence (ECL) immunoassay amplified by coreaction accelerators has experienced major breakthroughs in ultrasensitive detection of biomarkers. Herein, CeO2/SnS2 heterostructures were synthesized and applied as a novel coreaction accelerator to enhance the ECL efficiency of the luminol-dissolved O2 system for the first time. Benefiting from the well-matched lattice spacing, ultrafine CeO2 nanoparticles (NPs) were grown in situ on layered SnS2 nanosheets (NSs) with improved dispersion. CeO2/SnS2 as an electroactive substrate can remarkably accelerate the generation of abundant superoxide anion radicals (O2•-) to react with luminol anion radical (L•-), achieving about 2-fold stronger ECL intensity than that of pure CeO2 NPs. To avoid harsh chemical synthesis of conventional ECL labels and simplify the antibody conjugation process, ferritin (Ft) was served as a natural nanocarrier to immobilize luminol molecules (Lum@Ft) via a one-step linkage, whose protein nanocage can easily connect with the detection antibody. Moreover, a robust site-oriented immobilization strategy using HWRGWVC heptapeptide as specific capturer was further adopted to maintain the bioactivity of the capture antibody on the amine-functionalized CeO2/SnS2 surface, which promoted the incubation efficiency markedly. On account of this advanced sensing strategy, a brand new biosensor was constructed for the accurate detection of heart failure biomarkers, which performed with favorable linearity in the range of 0.0001-50 ng/mL and achieved the detection limit of 36 fg/mL.

Abstract:Ever-increasing attention is being drawn towards single-atom nanozymes (SAzymes) for colorimetric biosensing applications. A series of atomically dispersed cerium (Ce) nanozymes are synthesized with different feeding molar ratios of Ce/Zn or under various pyrolysis temperatures, and their enzymatic performance, including peroxidase-, oxidase-, and catalase-activities, is explored. Both theoretical and experimental results demonstrate that Ce species symmetrically coordinate with nitrogen atoms to form CeN 4 moieties which exhibit peroxidase-like activity. The best Ce-based SAzyme achieves remarkable peroxidase-like specificity and enables the colorimetric sensing of H 2 O 2 . This sensitivity provides a versatile reporter platform for detecting the activities of natural enzymes involved in H 2 O 2 generation and further monitoring their substrates as well as inhibitors, for example, organophosphorus pesticides (OPs) when integrating with the acetylcholine esterase-choline oxidase biocatalyzed cascade. More importantly, this proposed sequential catalytic biosensor exhibits the capability to monitor trace OPs in the real samples with high sensitivity yet insignificant interference. This work sheds new light on further design of highly active lanthanide-based SAzymes to boost enzyme-mimicking reactions for the applications in rapid on-site colorimetric analysis of emerging contaminants in environmental and food samples.

Abstract:Chemiluminescence (CL) has been widely used for bioanalysis owing to its high sensitivity, low background and simplicity. However, most of the CL systems need acidic/alkaline conditions or organic solvent to enhance their luminescent efficiency, and the non-physiological conditions can usually lead to the misfunction of biomolecules during biosensing. Herein, we report a highly luminous CL system under physiological conditions based on carbon dots-bis(2-carbopentyloxy-3,5,6-trichlorophenyl) oxalate (CDs-CPPO) micelles, and further used it in biosensing application. In the CL system, the amphiphilic surfactant packed CPPO and hydrophobic CDs together to form CDs-CPPO micelles. Such micelles solution not only isolated the CPPO from water to prevent its hydrolysis but also made the close proximity between CPPO and CDs, thus significantly enhancing the CDs quantum yield. The CL quantum yield was calculated to be 5.26 × 10−4 einsteins/mol, about 200-fold higher than that of the most commonly used luminol CL system. The oxidases (e.g., glucose oxidase) were tested to be susceptible to the organic solvent and non-physiological pH. Hence, the CL system was used for the detection of oxidase substrates (exemplified by glucose) in serum samples, and the limit of detection was as low as 8.4 nmol/L. The highly luminous CL system that can work under physiological conditions is promising for biosensing applications

Abstract:The development of enhanced strategies with excellent biocompatibility is critical for electrochemiluminescence (ECL) imaging of single cells. Here, we reported an ECL imaging technique for single cell membrane protein based on Co3O4 nanozyme catalytic enhancement strategy. Due to the remarkable catalytic performance of Co3O4 nanozymes, H2O2 can be efficiently decomposed into reactive oxygen radicals, and the reaction with L012 was enhanced, resulting in stronger ECL emission. The anti-carcinoembryonic antigen (CEA) was coupled with nanozyme particles to construct a probe that specifically recognized the overexpressed CEA on MCF-7 cell membrane. According to the locally enhanced visualized luminescence, the rapid ECL imaging of single cell membrane protein was realized eventually. Accordingly, Co3O4 nanozymes with highly efficient activity will provide new insights into ECL imaging analysis of more biological small molecules and proteins.

Abstract:Luminol, the first discovered and man-made effective chemiluminescence (CL) system, is the best known and one of the most widely used CL materials. The chemiluminescent process of the luminol CL has not yet been fully elucidated, although the decomposition of 1,2-dioxane-3,6-dione dianion (CP2-) is verified to be the key step to produce light emitter. However, the mechanisms of the CP2- decomposition and the effective singlet chemiexcitation are totally unknown, which is the outstanding obstacle to comprehending luminol CL. In the present work, by means of the state-of-the-art multireference computation and the nonadiabatic molecular dynamics (NAMD) simulation, the decomposition mechanism of CP2- is clearly revealed. A stepwise single electron transfer from the aminophthaloyl to the O-O bond initiates the decomposition of CP2-, and the light emitter is produced via the two crossings of the potential energy surfaces of the ground state (S0) and the first singlet excited state (S1). The NAMD simulated quantum yield of the light emitter demonstrates that the located two conical intersections control an effective nonadiabatic pathway in luminol CL. The proposed mechanism of "two conical intersections" is suitable not only to luminol but also to other CL materials with cyclic peroxide as the chemical energy provider.
Co-amplification of Luminol-Based Electrochemiluminescence Immunosensors Based on Multiple Enzyme Catalysis of Bimetallic Oxides CoCeOx and NiMnO3 for the Detection of CYFRA21-1

Abstract:The accelerated energy supply of co-reactants is an extremely effective strategy for achieving highly sensitive electrochemiluminescence analysis, and binary metal oxides would be an excellent tool for this purpose owing to the nano-enzyme acceleration of mixed metal valence states. Herein, an electrochemiluminescent (ECL) immunosensor for monitoring the concentration of cytokeratin 19 fragment antigen 21-1 (CYFRA21-1) was developed based on a co-amplification strategy triggered by two bimetallic oxides, CoCeOx and NiMnO3, with luminol as the luminophore. CoCeOx derived from an MOF exhibits a large specific surface area and excellent loading capacity as a sensing substrate, and the peroxidase properties enable the catalysis of hydrogen peroxide to provide energy supply to the underlying radicals. The dual enzymatic properties of flower-like NiMnO3 were employed as probe carriers for luminol enrichment. The peroxidase properties built on Ni2+/Ni3+ and Mn3+/Mn4+ binary redox pairs resulted in the integration of highly oxidative hydroxyl radicals, and the oxidase properties provided additional superoxide radicals via dissolved oxygen. The practically proven multi-enzyme-catalyzed sandwich-type ECL sensor easily accomplished an accurate immunoassay of CYFRA21-1, harvesting a detection limit of 0.3 pg mL(-1) in the linear range of 0.001-150 ng mL(-1). In conclusion, this work explores the cyclic catalytic amplification of mixed-valence binary metal oxides with nano-enzyme activity in the field of ECL and develops an effective pathway for ECL immunoassay.

Abstract:As a peroxidase mimic, cupric oxide nanoparticles were found to enhance the chemiluminescence (CL) of luminol-H(2)O(2) system up to 400 folds. The CL spectra and radical scavengers were conducted to investigate the possible CL enhancement mechanism. It was suggested that the enhanced CL could be attributed to the peroxidase-like activity of CuO nanoparticles, which effectively catalyzed the decomposition of hydrogen peroxide into hydroxyl radicals. The effects of the reactant concentrations and some organic compounds were also investigated. The proposed method could be used as a sensitive detection tool for hydrogen peroxide and glucose.

Abstract:Cuprous oxide (Cu2O) crystals are usually used as the photocatalysts of water splittings and carbon monoxide oxidations. Herein, we report the new catalytic properties of Cu2O to the electrochemilumenescence (ECL) reactions of luminol and oxygen. Adjusting the shape of Cu2O microcrystals from cube to octahedra also facilities the improved electrocatalytic acticity, where octahedral Cu2O microcrystals possess higher ECL signal by 25% at -0.6 V (versus Ag/AgCl) at pH 7.4. The octahedral Cu2O microcrystals are synthesized by the coordination of iodide ions to the surface, which alters the crystals shapes from cubes to octahedra. Size-distribution of octahedra is improved with this mehod compared to the previous studies with only hydroxide ions as the coordinates. The catalytic activity of octahedral Cu2O crystals is expected to be compared with the noble metal nanomaterials and constructed the high-efficiency and low-cost ECL biosensors.

Abstract:Using a heterogeneous catalyst, Co(II)-ethanolamine complex sorbed on Dowex-50W resin, the chemiluminescence (CL) of luminol in unbuffered or weakly acidic solution was studied in the presence of H2O2. The maximum luminol CL wavelength at pH 5.7 was 448 nm, 23 nm longer than that in a basic solution (pH 10.5). Three different ligands, mono-, di-, and triethanolamine, and six transition metal ions, Co(II), Cu(II), Ni(II), Mn-(II), Fe(II), and Fe(III) were compared by CL measurements. The CL intensity decreased in the order mono- > di- > triethanolamine and Co(II) > Cu(II) > Ni(II) > Fe-(III) > Mn(II) > Fe(II). This heterogeneous CL system was developed as H2O2 and glucose flow-through sensors. Detection limits (S/N = 3) of H2O2 and glucose using Dowex-50W-X4-Co(II)-monoethanolamine as catalyst are 1 x 10(-7) M and 1 x 10(-6) M, respectively. On the basis of the studies of the CL, fluorescence, UV-vis and ESCA spectra and the effect of dissolved oxygen in luminol solution, a mechanism for CL emission in unbuffered solution was considered as the formation of a superoxide radical ion during the decomposition of H2O2 catalyzed by the Co(II)-ethanolamine immobilized resin. Then the superoxide radical ion acted on luminol and the CL was emitted. The applications of the proposed method to determine H2O2 in rainwater without any special pretreatment and glucose in human urine and orange juice samples give satisfactory results.
Supramolecular Catalyst for Improving the Sensitivity of Biomolecule Detection

Abstract:Although traditional natural enzymes exhibit excellent selectivity and high efficiency in catalyzing reactions, their sensitivity to environmental changes (e. g., pH, temperature) limits their wide range of applications. To address this problem, supramolecular catalysts have emerged, which are capable of mimicking the catalytic mechanism of natural enzymes through non‐covalent interactions to form structures with specific functionalities, thus enabling efficient detection of biomolecules. In this paper, we will focus on the application of four supramolecular catalysts, cyclodextrin (CD), cucurbituron (CB), porphyrin (PP), and columnar aromatics (CA), in catalyzing the reaction and improving the detection sensitivity. Supermolecule refers to two or more molecules that are bonded together by secondary bonds between molecules to form a multimolecular system at the level of more than molecules. At this point, the molecules form an orderly aggregation, showing its unique structure and function. Supramolecular catalysts can form structures with specific functions through non‐covalent interactions (e. g., hydrogen bonding, π‐π stacking, etc.) to mimic the catalytic mechanism of natural enzymes and achieve susceptible and specific detection of biomolecules. The application of supramolecules is mainly focused on the self‐assembly of components for the construction of sensing platform. Therefore, supramolecular catalysts have a huge potential in biomolecule detection. This article focuses on the application of cyclodextrins(CD), cucurbit[n]uril(CB), porphyrins(PP) and Calix[n]arenes(CA) in the field of catalysis and in improving detection sensitivity. Research progress of supramolecular catalysts in polymerization amplification is summarized in this paper.

Direct Probing of Single-Molecule Chemiluminescent Reaction Dynamics under Catalytic Conditions in Solution

Abstract:Chemical reaction kinetics can be evaluated by probing dynamic changes of chemical substrates or physical phenomena accompanied during the reaction process. Chemiluminescence, a light emitting exoenergetic process, involves random reaction positions and kinetics in solution that are typically characterized by ensemble measurements with nonnegligible average effects. Chemiluminescent reaction dynamics at the single-molecule level remains elusive. Here we report direct imaging of single-molecule chemiluminescent reactions in solution and probing of their reaction dynamics under catalytic conditions. Double-substrate Michaelis-Menten type of catalytic kinetics is found to govern the single-molecule reaction dynamics in solution, and a heterogeneity is found among different catalyst particles and different catalytic sites on a single particle. We further show that single-molecule chemiluminescence imaging can be used to evaluate the thermodynamics of the catalytic system, resolving activation energy at the single-particle level. Our work provides fundamental insights into chemiluminescent reactions and offers an efficient approach for evaluating catalysts.
