Abstract:A regioselective electrochemical C–H amination method to synthesize N2-substituted 1,2,3-triazole using easily accessible ethers has been developed. Various substituents, including heterocycles, have a good tolerance, and 24 examples were obtained in moderate to good yields. Control experiments and DFT calculation investigations demonstrate that the electrochemical synthesis undergoes a N-tosyl 1,2,3-triazole radical cation process promoted by the single-electron transfer of the lone pair electrons of the aromatic N-heterocycle, and the desulfonation is responsible for the high N2-regioselectivity.

Abstract:A new catalytic enantioselective approach for the formation of allyl alpha-amino acid derivatives by reaction of N-tosyl alpha-imino esters with allyl stannanes and silanes catalyzed by chiral copper(I) complexes has been developed. A series of different BINAP and phosphine-oxazoline (P,N) ligands have, in combination with various Lewis acids, been tested as chiral catalysts for allylation of N-tosyl alpha-imino esters. It has been found that both type of ligands, in combination with copper(I) salts, give highly valuable unsaturated alpha-amino acid derivatives. The reaction has been investigated for different allyl stannanes and silanes, and it has been found that tri-n-butyl allyl stannane gives the best results of the simple allyl compounds tested, leading to gamma,delta-unsaturated alpha-amino acid derivatives in up to 94% yield and with up to 83% ee, which can be improved to be >95% ee by recrystallization. The reaction has also been investigated using different acyclic and cyclic allyl stannanes leading to various types of unsaturated a-amino acid derivatives in very high yield (up to 95%) and with up to 98% ee. The stereochemistry and absolute configurations of the allyl alpha-amino acid derivatives have been determined by X-ray analysis, and it is suggested that the reaction takes place as an ene-like reaction.

Abstract:A general and convenient selective direct arylation of the 3‐position of triimidazo[1,2‐a:1',2'‐c:1'',2''‐e][1,3,5]triazine (1) with (hetero)aryl halides in DMA was successfully achieved in the presence of K2CO3 as the base and a catalyst precursor consisting of Pd(OAc)2 and P(2‐furyl)3. Electron‐poor and ‐rich (hetero)aryl moieties, including the strongly deactivated and sterically encumbered 2,4,6‐trimethoxyphenyl unit, are well tolerated in the electrophilic partner. The data obtained in this synthetic study support a reaction mechanism involving an electrophilic attack of an arylpalladium‐(II) halide species onto the triazine ring.

Abstract:A series of bifunctional phosphine-thiourea organic catalysts based on natural amino acid scaffolds were designed and prepared. L-threonine-derived bifunctional phosphine catalysts were found to be very efficient in promoting asymmetric Morita-Baylis-Hillman (MBH) reaction of acrylates with aromatic aldehydes, affording the desired MBH adducts with up to 90% ee. To gain mechanistic insights into the reaction, the effects of adding various additives on the MBH reaction were investigated. We propose that the hydrogen bonding interactions between the thiourea moiety of the catalyst and the enolate intermediate are crucial for the stereochemical outcome of the reaction. The method described in this report may provide a practical solution to the enantioselective MBH reaction of simple acrylates.

Abstract:Six Cp*Ir complexes containing NN-bitentate chelate ligands [Cp*IrCl(C5H4CH2C5H3OH)][Cl] (1), [Cp*IrCl(C5H4CH2C5H3O)] (2), [Cp*IrCl(C5H4C5H3OH)] [Cl] (3), [Cp*IrCl(C5H4CH2C5H4)][Cl] (4), [Cp*IrCl(CH3OC5H3CH2C5H3OCH3)][Cl] (5), and [Cp*IrCl(CH3OC5H3CH2C5H3OH)][Cl] (6) were synthesized and characterized. Complex 1 could be transformed to 2 when reacted with (NaOBu)-Bu-t or NEt3 via -OH deprotonation. These six complexes were tested as catalysts for mono-N-alkylation of amines with trialkyl amines, and complex 1 exhibited highest activity. The coupling reactions proceed under air condition, with 1 mol % catalyst loading without extra base in methanol at 120 degrees C and can be further accelerated by adding NR3.HCl.

Abstract:The observation of thioester-mediated acyl transfer processes in nature has inspired the development of novel protein synthesis and functionalization methodologies. The chemoselective transfer of an acyl group from S-to-N is the basis of several powerful ligation strategies. In this work we sought to apply the reverse process, the transfer of an acyl group from N-to-S, as a method to convert stable chiral amides into more reactive thioesters. To this end, we developed a novel cysteine-derived oxazolidinone that serves both as a chiral imide auxiliary and an acyl transfer agent. This auxiliary combines the desirable features of rigid chiral imides as templates for asymmetric transformations with the synthetic applicability of thioesters. We demonstrate that the auxiliary can be applied in a range of highly selective asymmetric transformations. Subsequent intramolecular N-to-S acyl transfer of the chiral product and in situ trapping of the resulting thioester provides access to diverse carboxylic acid derivatives under mild conditions. The oxazolidinone thioester products can also be isolated and used in Pd-mediated transformations to furnish highly valuable chiral scaffolds, such as noncanonical amino acids, cyclic ketones, tetrahydropyrones, and dihydroquinolinones. Finally, we demonstrate that the oxazolidinone thioesters can also serve as a surrogate for SNAC-thioesters, enabling their seamless use as non-native substrates in biocatalytic transformations.

Abstract:The first enantioselective synthesis of chiral heterocyclic α-amino phosphonates by nucleophilic dearomatization of quinolines and pyridines using an anion-binding organocatalysis approach is described. Chiral tetrakistriazoles were employed as efficient hydrogen-bond donor catalysts by forming a chiral close ion-pair with the in situ formed N-acyl salts and 2,2,2-trichlorethoxycarbonyl chloride (TrocCl). The ion-pair was subsequently treated with various phosphorus nucleophiles, such as silyl-protected dialkyl- and trialkylphosphites. Thus, the corresponding products were obtained in complete or high regioselectivities and up to 97:3 e.r. for quinolines or up to 89:11 e.r. for the more challenging pyridine substrates. This method allows for rapid access to substituted chiral cyclic α-amino phosphonates, which can be easily transformed into phosphonic acid derivatives.

Abstract:Nonionic, chiral macrocyclic peptoids are efficient phase-transfer catalysts in the C-4 enantioselective alkylation of 2-[4-(trifluoromethyl)phenyl]-2-oxazoline-4-carboxylic acid esters. Screening of the structural features of cyclic peptoids, namely the ring size, symmetry elements, number of proline residues, and substituents on the side chains, showed that the alternated N-(4-methoxybenzyl)glycine/l-proline cyclohexapeptoid is the optimal catalyst (good yields and up to 75% ee) for the stereocontrolled construction of α-alkylated serine tert-butyl esters.

Abstract:A radical benzylic amination of α-cyanoarylacetates without the assistance of any transition-metal catalysts or external photosensitizers was realized. This protocol is operationally simple, mild and of high efficiency. The products could be easily transformed to useful vicinal diamines and α-amino amides. DFT calculations revealed the origin of benzylic rather than aromatic amination regioselectivity in the present reaction.

Abstract:Triarylmethanols are well-known core structures in natural products and pharmacologically relevant compounds. Triarylmethanols are well-known core structures in natural products and pharmacologically relevant compounds. In general, transition metal-based catalysts or highly reactive organometallics are employed for the synthesis of these compounds. Herein, we report the regioselective tandem C(sp 3 )–H arylation/oxidation of diarylmethanes with nitroarenes to generate arylated alcohols. The present method is general, mild, green, and conducted in air at room temperature. Furthermore, use of triarylmethanes as pro-nucleophiles provides straightforward access to select tetraarylmethanes through a cross-dehydrogenative coupling process.
