An Experimental Study on the Influence of Nicotine to bone Reparing Capability of Calvarial Defects in Rats

Abstract:Objective:To study the influence of nicotine to osteogenic efficiency of calvarial defects of rats. Methods: 24 SD rat were randomly assigned to one of the following groups: Group 1 - control (n = 12) and Group 2 - subcutaneous nicotine administration (n = 12). Nicotine (0.5mg/kg/d ) was injected into rats of Group 2 and 0.9% sodium chloride solution was injected into rats of Group 1. Twenty days later, a critical-sized calvarial defects was created. Injection scheme was continued as before. The rats were scarified on 21th day and 56th day after operation. The repairing condition of bone defects was evaluated by tetracycline fluorescence marker, quantitative bone histomorphometry and mineral content mensuration of new bone. Results: On the 21th day following operation, new bone formed within the bone defects in both groups. The region labled with tetracycline within bone defects were sparser in Group 2. Significant difference was found between Group 1 and Group 2 about the distance between two fluorescent lines of incorporated tetracycline and the mean mineralization deposition. On the 56th day after operation, repairing bone proportion was greater in control group. Mineral content analysis of new bone in defects revealed statistically significant decrease in Group 2. Conclusion: Nicotine induced disadvantageous effect in bone repairing efficiency.
