PROSTAGLANDIN E2 INDUCES THE PHOSPHORYLATION OF CREB AND GABABR2 IN CULTURED RAT ASTROCYTES

Abstract:Prostaglandin E2 (PGE2) has long been reported to be an important mediator in inflammation. Phosphorylation of transcription factor cAMP response element binding protein (CREB) and GABABR2 subunit affects various neurophysiological functions in different ways. To study the phosphorylation of CREB and GABABR2 subunit induced by PGE2 in astrocytes, immunocytochemistry and Western blotting were used to determine the phosphorylation of CREB and GABABR2 (Ser892) in cultured astrocytes treated by 10 μmol/L PGE2 for various periods. The results showed that basic phosphorylation levels were present for CREB and GABABR2 in normal astrocytes. The levels of phosphorylation of CREB and GABABR2 (Ser892) could be observed elevated after treated with PGE2 in a time-dependent manner compared with control groups. The results demonstrate that PGE2 can induce phosphorylation of CREB and GABABR2 (Ser892) in cultured astrocytes and indicate that PGE2 may play a considerable role in inflammatory situation in central nervous system.
