Abstract:Modulation of apoptosis may potentiate the sensitivity of tumor cells to chemotherapeutic agents, thus improving the clinical outcome of cancer treatment. Bax, an apoptosis-promoting member of the bcl-2 family, may be a key factor influencing the chemosensitivity of tumor cells, however, its involvement in cellular sensitivity to anti-cancer drugs remains uncertain in squamous cell carcinoma (SCC). To investigate the role of bax gene expression in modulating cisplatin (CDDP)-induced apoptosis in vitro, an established CDDP-resistant human head and neck SCC (IMC-3 cell line) was transfected with bax gene-bearing mammalian expression vector. Overexpression of the bax gene in CDDP-resistant IMC-3 cells elevated the CDDP susceptibility of tumor cells to a level similar to that of the parental IMC-3 cells. In an in vivo study, percutaneous transfer of apoptosis-promoting bax gene by particle-mediated (gene gun) delivery caused overexpression of Bax in SCC, which was confirmed by immunohistochemical staining, and inhibited the growth of mouse CDDP-resistant SCC. Furthermore, combination therapy with bax gene transfer and subcutaneous administration of CDDP at 3-day intervals markedly inhibited the growth of mouse SCC. Thus, overexpression of bax in SCC by a gene gun system appears to be a rational approach to improving the efficacy of chemotherapy and treatment outcome. We suggest that exogenous bax expression may have therapeutic applications for enhancing chemotherapy in SCC.
Influence of Bax on Apoptosis and Drug Sensitivity of HCC-9204 Cells

Abstract:Objective To investigate the role of Ba x in apoptotic pathways and the response of human HCC-9204 cells to cell death induced by adriamycin. Methods The whole length of Bax cDNA was transfected into human hepatocellular car cinoma cell line HCC-9204 by lipofectamine transfection. Expression of Bax in protein was analyzed by immunohistochemistry. TUNEL and flow cytometry were used to assess the effect of Bax on apoptosis. Tetrazolium blue (MTT) assay was used to evaluate drug sensitivity of HCC-9204 cells. Re sults The expression of Bax protein increased in Bax transfected cells at 24~48h after the addition of ZnSO_4. Apoptotic index sig nificantly increased in HCC-9204/Bax cells at 24h and 48h (3.6±5.3 vs 27 .2±10.5, t=63.45, P0.05;4.2±4.1 vs 32.3±8.6, t=93.27, P0.05), respectively. Flow cytometry showed a significant sub-G1 peak an d apoptosis in 15.36% HCC-9204/Bax cells at 48h after treatment. MTT assay showed that the effects of Bax on cell viability following ADR exposure were sig nificant as compared to the vehicle-transfected HCC-9204/pMD cells. Conclusion Overexpression of Bax co uld not only induce apoptosis, but also sensitize HCC-9204 cells to cell death induced by adriamycin.
Effect of BZG-4000, a novel multi-targeted kinase inhibitor with potent anticancer activity, on a hepatocellular carcinoma xenograft model

Abstract:The present study was to synthesize a novel multi-targeted kinase inhibitor and evaluated its anticancer effects on a hepatocellular carcinoma xenograft model. In our study, in vivo efficacy was determined in nude mice bearing HuH7 human HCC xenografts. The mice were randomly divided into the following five groups with the use of a randomization chart (n = 8 in each group): high-dose BZG-4000 group, medium-dose BZG-4000 group, low-dose BZG-4000 group, sorafenib group and model group. Tumor size measurements included the length (L) and width (W) measured with calipers and tumor volume was calculated as (LW ∧ 2)/2. Tumor tissues slides were hematoxylin and eosin (HE) stained for histopathological examination. Immunohistochemistry detected CD31 expression and Western blotting measured VEGF protein expression. We found that when BZG-4000 was administered orally to xenograft HuH7 nude mice, tumor growth was inhibited and significant tumor shrinkage was evident. After oral administration of BZG-4000 at 40 mg/kg/day, the tumor weight and volume were significantly lower than tumors of the sorafenib group. BZG-4000 considerably decreased the expression of CD31 and VEGF in tumors compared to tumors treated with positive control drug. It was concluded that BZG-4000 has the potential to inhibit the tumorigenesis of hepatocellular carcinoma in vivo by decreasing the expression of CD31 and VEGF.
Overexpression of Bax Induces Apoptosis and Enhances Drug Sensitivity of Hepatocellular Cancer-9204 Cells.

Abstract:AIM:To investigate the role of overexpression of Bax in apoptotic pathways and the response of human hepatocellular cancer (HCC)-9204 cells to cell death induced by adriamycin.METHODS:The whole length of Bax cDNA was transfected into human HCC-9204 cells by the method of lipofectamine transfection. An inducible MT-II regulatory system was constructed, which allowed controlled expression of protein upon addition of ZnSO4 (100 micromol/L) as an external inducer. Stable transfecting inducible expression vector containing Bax gene was performed. Expression of Bax in protein was analyzed by immunohistochemistry and Western blotting. TUNEL and flow cytometry were used to assess the effect of Bax on apoptosis. Colony assay and tetrazolium blue (MTT) assay were used to evaluate the difference in drug sensitivity of HCC-9204 cells after Bax-transfection.RESULTS:Immunohistochemistry and Western blotting demonstrated that the expression of Bax protein markedly increased in Bax-transfected cells 4 h after the addition of ZnSO4. Bax positive signal was frequently found on the cytoplasm and perinuclear region of HCC-9404 cells, and there was ectopic expression in cells with marked condensation of chromatin and cytoplasm (apoptotic cells). Apoptotic index significantly increased in Bax-transfected HCC-9204/Bax cells (3.6 vs 27.2, 4.2 vs 32.3, P<0.05). Flow cytometry analysis showed a significant sub-G1 peak and apoptosis in 15.4% HCC-9204/Bax cells 24 h after treatment. Furthermore, colony survival rate decreased from 66% (HCC-9204/pMD) to 45% (HCC-9204/Bax) 2 d after ADR withdrawal. MTT assay result showed that the effects of Bax on cell viability following ADR exposure were significant as compared to the vehicle-transfected HCC-9204/pMD cells (21% vs 44%, P<0.01).CONCLUSION:Overexpression of Bax not only induces apoptosis, but also sensitizes HCC-9204 cells to cell death induced by adriamycin.
Significance of Expression of Bcl-2 Mrna and Bax Mrna in Arsenic Trioxide-Induced Apoptosis of Hepatocellular Carcinoma Cells

Abstract:ObjectiveTo explore of significance of expression of bcl-2 mRNA and bax mRNA in arsenic trioxide-induced apoptosis of hepatocellular carcinoma (HCC) cells. MethodsAfter human HCC cell line bel-7402 was treated with arsenic trioxide at various concentrations, the apoptosis was detected with transmission electron microscopy, flow cytometry and DNA laddering. Meanwhile, the expression of bcl-2 mRNA and bax mRNA was determined with semiquantitative reverse transcription polymerase chain reaction (RT-PCR). ResultsCell apoptosis was not detected in 0.5 μmol/L As_2O_3 group but in 8 μmol/L As_2O_3 group. The expression of bcl-2 mRNA were not seen in the control group and drug-treated group. Enhancement of expression of bax mRNA along with increase of As_2O_3 concentration. ConclusionsProliferation is inhibited via inducing apoptosis by As_2O_3 in bel-7402 cell line. Enhancement of bax mRNA expression but not that of bcl-2 mRNA expression is related to this process.

Abstract:There is a growing tendency of the morbidity of hilar cholangiocarcinoma in recent years. Some studies indicate that Bax gene expression was slightly increased in adjacent tissues of cholangiocarcinoma. Therefore, it is speculated that this condition may have certain correlation with the invasive ability of cholangiocarcinoma cell. This research aimed to detect the effect of Bax on cell invasive ability. The cholangiocarcinoma cell line RBE was cultured. Bax high-expression vector and Bax small interfere RNA (Bax-siRNA) were named pLBax and pLBax-siRNA, respectively. The constructed vector was transfected into the RBE cell line by using liposome transfection followed by analysis of the relationship between Bax expression and invasive ability of cholangiocarcinoma cell. The Bax gene expression in cells transfected with both vectors showed increased and decreased expression, respectively. The RBE cell apoptosis was significantly increased after high expression of Bax, while cell invasive ability was decreased with significant differences among pLBax, pLBax-siRNA and control group ( p < 0.05). Bax was downregulated and RBE cell showed reversed changes with significant differences among different groups ( p < 0.05). The change of the Bax expression in cholangiocarcinoma cell can affect cell invasive ability and apoptosis. Bax expression was negatively correlated with cell invasive ability.

Experimental Study of Effects of Bax Gene on Human Colorectal Cancer Cell Line HT-29

Abstract:Objective To evaluate the therapeutic effects of bax gene on human colorectal cancer cell line HT-29. Methods Human colorectal cancer cell line HT-29 was transfected with adenovirus mediated bax gene. The morphological changes, growth and apoptosis of transfected cells were examined under phase-microscope, MTT method and flowcytometry. The bax protein expression in bax transduced cells was detected by Western Blot. Results Marked morphological changes of HT-29 were found, the effects were more significant when combined with Ad/PGK-GV16. Cell growth inhibition and apoptosis were 57.4%, 28.2% respectively, higher than Ad/GT-bax alone (29.2%, 12.6%, P0.05). bax protein expression in HT-29 cells transfected with dual vectors containing the bax gene was much higher than that transfected with Ad/GT-bax alone or other vectors. Conclusions bax gene transduction effectively induced the apoptosis of HT-29 cell.

Abstract:The chicken anemia virus protein, apoptin, which manifests selectivity and specificity to tumor cells, induces a p53-independent and Bcl-2-insensitive type of apoptosis in various human tumor cells. In this study, the apoptin gene was cloned from the total DNA of chicken anemia virus, and the recombinant vector was constructed. We used oligonucleotide microarray to study the changes of four genes, including Bcl-2, Bcl-(XL), Bad and Bax. The post -transfection with the recombinant was also studied. The pro-apoptotic genes(Bad and Bax) and anti-apoptosis genes(Bcl-2 and Bcl-(XL)) were up-regulated in contrast to the controls. According to the published data, either Bcl-2 or Bel-(XL) can form non-functional heterodimers by Bad and Bax binding together, resulting in blocking partly the. release of cytochrome c from mitochondria. However, apoptosis could be inhibited by neither the endogenous Bcl-XL nor Bcl-2 over-expression. The experiments show that the apoptin-induced apoptotic pathway is related to the up-regulation of Bad and Bax. Bad was up-regulated by apoptin; then this up-regulated product of Bad was in favor of displacing Bax from binding to Bcl-XL or Bcl-2. Consequently. Bax exerted a pro-apoptotic dysfunction to mitochondria, thereby inducing the release of cytochrome c. Finally, apoptin induced the apoptosis of HHCC cells. These results indicate that the oligonucleotide microarray can reveal the genes related to the apoptosis induced by apoptin in HHCC cells.
Research on Relationship Between Expressions of Apoptosis-Related Gene Bcl-X and Bax in Hepatocellular Carcinoma and Its Clinical Pathological Features

Abstract:Aim To study relationship between expressions of apoptosis-related gene bax and bcl-x in heopatocellular carcinoma (HCC) tissues and its clinical feature. Mothods The expression of Bax and Bcl-x were determined by immunohistochemistry with EnvisionTM system. Results Bcl-x and Bax proteins were detectable in 20 and 19 out of 40 HCCs, respectively. Their expressions in HCC cells were lower than those in cirrhosis(P＜ 0.05), The expression of Bax and Bcl-x in highly or intermediatly differentiated HCC cells were highed than that in low-diffe-rehtiated HCC cells suggesting that there was significant relationship among bcl-x and bax expressions and differential degree， clinical stages of HCC cells， and AFP level. However, the expression of this proteins showed no relationship with patient's sex and age(＞ 60 or ＜ 60 years old) (＞ 0.05). Conclusion Apoptosis-related genes bcl-x and bax participate partially in apoptotic regulation of HCC cells, and show that their expressions have correlation with differential degree, clinical stage of HCC cells and AFP levels.
A rudiment approach to hepatitis B virus X gene's effect on the apoptosis of normal HL7702 cells

Abstract:Objective To investigate HBx's effect on the apoptosis of HL7702 and the expression of apoptotic factors. Methods The HBx gene eukaryon expression vector pcDNA3.1 (+) - X was transiently transfected into HL7702 cells by lipid - media transfection,with HL7702 transfected with pcDNA3.1 (+) and HL7702 untransfected taken as controls. 72 h after transfection,the cells were collected to proceed with the following tests; expression of HBX mRNA by RT -PCR,expression of HBx protein in cells by immunofluorescency, apoptotic rate by flow cytometry, evaluation of the mRNA levels of Bax and Bcl-2 by semi - quantified RT - PCR using β- actin as an internal control. Results HBX mRNA was detected in HL7702/ pcDNA3.1 (+) - X cells by RT - PC,showing strong immunofluorescence; whereas no expression of HBx was detected in the controls. As compared with the untransfected HL7702 group (1.40% ± 0. 37% ) and HL7702/pcDNA3.1(+)group (1.25% ±0. 30% ) ,the apoptotic rate in HL7702/pcDNA3.1 (+) - X group was obviously increased (3.38% ±0.67% , P <0.05). Semiquantitative RT - PCR analysis showed Bax mRNA level was significantly higher in HBX transfection group than those in the controls (P<0.05) , whereas the Bcl-2 mRNA level was significantly lower in he HBX transfected group than in the controls (P<0.05). No significant differences in apoptotic rate, Bax mRNA level or Bel - 2 mRNA level were found between the two control groups. Conclusion HBX gene can be transfected into HL7702 cells successfully. HBx up - regulates Bax and down - regulates Bcl-2, suggesting HBx can induce liver cell apoptosis.
A Study on the Mechanism of Adriamycin-Induced Apoptosis of Human Hepatic Carcinoma Cells

Abstract:ObjectiveTo investigate the mechanism of adriamycin-induced apoptosis of hepatic carcinoma HCC-9204 cells, and study the expression of apoptosis-associated genes Bcl-2 and Bax. MethodsTUNEL method and flow cytometry were used to detect the effect of adriamycin on the multiplication cycle of human hepatic cancer ACC-9204 cells, and the cell apoptosis. The expression of Bel-2 and Bax protein in (ACC-9204) cells was examined. ResultsAdriamycin markedly induced apoptosis of HCC-9204 cells, and typical characteristics of apoptosis were detected by staining. On cell cytometric analysis 24h after drug (treatment), a significant sub-G1 peak was detected. HCC-9204 cells treated with adriamycin showed (significant) down regulation of Bel-2 protein expression, but it was not significantly different from that of the control group(P0.05). ConclusionsAdriamycin can induce apoptosis of cancer cells, and this is an important mechanism for its anticancer effect. This effect may be related to the down regulation of Bel-2 (expression).
[Effect of HBx Antisense Oligodeoxynucleotide on Formation of Transplanted Hepatocellular Carcinoma in Nude Mice].

Abstract:OBJECTIVES:To study the inhibitory effect of HBx antisense oligodeoxynucleotide on the formation of transplanted hepatocellular carcinoma in nude mice.METHOD:50 nude mice were randomly divided into 5 groups: 1 control group and 4 experimental groups. Log-phase Hep3B cells endogenously expressing HBX were injected subcutaneously in nude mice. From the second day, the PAGE purified AS1, AS2, AS3 and AS4 HBx antisense oligodeoxynucleotides were injected intraperitoneally into the 4 experimental groups, respectively, on alternate days for 5 times, and distilled water was injected into the control group. Growth information of subcutaneous transplantation tumor in nude mice was recorded for 30 days. Incidence rate of transplanted tumor in different groups was compared and analyzed by survival analysis. Statistics software SPSS12.0 was used to analyze the data.RESULTS:Incidence rate of transplanted tumor was 100% in AS1, AS2, AS3 and control groups, and 90% in AS4 group (x2 = 3.995, P = 1.0). The median latency period for transplanted tumor formation was 19 days (17.48-20.52), 12 days (9.93-14.07), 11 days (9.45 to 12.55), 21 days (19.48 to 22.52), and 10 days (8.99 to 11.01) in AS1, AS2, AS3, AS4 and control group, respectively. The latency period for tumor formation was prolonged by treatment of mice with AS1 and AS4 antisense oligodeoxynucleotide (P less than 0.01).CONCLUSION:Antisense oligodeoxynucleotide targeting to the appropriate sites of HBx gene can prolong the latency period of subcutaneously transplanted tumor in nude mice, however, the formation of transplanted tumor can not be completely blocked by limited treatment with these antisense oligos. In addition, our results suggest that peritoneal injection may be an effective way to deliver antisense oligodeoxynucleotide to living organisms.
[Effect of Hepatitis B Virus X Gene on the Apoptosis of HepG2 Cells].

Abstract:OBJECTIVE:To investigate the effect of hepatitis B virus X gene on the apoptosis in X gene-transfected HepG2 cells.METHODS:HBX gene eukaryon expression vector pcDNA3.1-X was transfected into HepG2 cells by lipid-mediated transfection to establish HepG2/HBX cell model for HBX expression. HepG2 transfected with pcDNA3.1 was used as controls. At 24 h, 48 h, 72 h and 96 h after transfection, cell apoptotic rates were detected by flow cytometry. RT-PCR and Western blot were applied to evaluate the expression levels of HBX, Fas, FasL, and the levels of p-JNK and p-c-Jun protein.RESULTS:HBX mRNA was detected in HepG2/ HBX cells 24 h after transfection, and the expression of HBX mRNA level was gradually up-regulated after transfection (P < 0.05); whereas no expression of HBX gene in the control cells. At 24 h, 48 h, 72 h and 96 h after transfection, all of the apoptotic rate in HepG2/HBX group were much higher than those in the controls (P < 0.05). The expression levels of Fas and FasL protein as well as the levels of p-JNK and p-c-Jun protein were significantly higher than those in the controls (P < 0.05). A positive correlationship was observed between the expression of HBX mRNA level and the hepatocyte apoptotic rate (P < 0.05), the same correlation of HBX mRNA level with the levels of Fas, FasL, p-JNK and p-c-Jun were also observed. (P < 0.05).CONCLUSION:HBX can induce hepatocyte apoptosis by up-regulating the levels of p-JNK and p-c-Jun to promote the expression of FasL.
An enhanced anti-tumor effect of apoptin-cecropin B on human hepatoma cells by using bacterial magnetic particle gene delivery system

Abstract:The gene therapy of cancer, due to the limit of its efficiency and safety, has not been widely used in clinical. Recently, bacterial magnetic particles (BMPs), which are membrane-bound nanocrystals found in magnetotactic bacteria, have been exploited as a new gene delivery system. However, its application on gene therapy remains to be explored. In our previous study, we found that a combination of cecropin B (ABPs) and apoptin (VP3) could serve as an effective gene therapeutic agent. Thus, in this study, we used BMPs to deliver the co-expression plasmid of these two gene, namely pVAX1-VA, and evaluated its therapeutic effect on human hepatocellular carcinoma (HepG2). Our results showed that BMPs significantly improved the efficiency of gene transfection (almost 3-fold than Lipofectamine 2000 at 48 h, P < .001), which led to stronger apoptosis (in a peak almost 2-fold than Lipofectamine 2000-pVAX1-VA, P < .01) and growth inhibition of HepG2 cells. More importantly, compared with Lipofectamine 2000-pVAX1-VA group, BMP-pVAX1-VA strikingly inhibited tumor growth (0.60 ± 0.09 g vs. 0.88 ± 0.11 g, P < .05) in nude mouse tumor models and increased the tumor-infiltrating lymphocytes considerably without apparent cytotoxicity. These findings suggest that BMPs could be an attractive gene delivery system for gene therapy and provide a potential available treatment for human hepatocellular carcinoma and maybe some other kinds of tumors.
Effect of Ad-Gax transfection on apoptosis of human lung adenocarcinoma A549 cells and its mechanism

Abstract:Background and objective: Apoptosis is closely related to development of lung cancer. It is a strategy of lung cancer therapy to induce apoptosis. The aim of this study is to explore the effects of growth arrest-specific homeobox (Gax) transfection on apoptosis and expression of Bcl-2 and Bax proteins of human lung adenocarcinoma A549 cells. Methods: A549 cells were transfected with Gax gene by a replication-deficient adenovirus expressing the hemagglutinin-tagged Gax cDNA (Ad-Gax). Apoptosis of A549 cells was observed by transmission electronic microscope and terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNED positive staining. Apoptotic rate of A549 cells was evaluated by flow cytometry. Expressions of Bcl-2 and Bax proteins in A549 cells were detected by immunocytochemistry. Results: Before Ad-Gax transfection, none or few of TUNEL-positive A549 cells were detected. After Ad-Gax transfection, a marked increase in TUNEL-positive staining occurred, especially at 24 h later. The ratio of apoptosis of A549 cells in non-transfection group and transfection groups at 12 h, 24 h, 48 h were 0.25%, 12.57%, 17.29%, 15.03%, respectively. Compared with non-transfection group, the apoptotic rates of transfection groups increased significantly (χ2 value was 7.357, 11.126 and 9.943 respectively, P<0.01). The average optical density (AOD) of Bcl-2 protein in A549 cells in non-transfection group and transfection groups at 12 h, 24 h, 48 h were 2.02 ± 0.07, 1.79 ± 0.02, 1.25 ± 0.51 and 1.21 ± 0.24 respectively. Compared with non-transfection group, AOD of Bcl-2 protein in A549 cells in transfection groups decreased significantly (t value was 6.651, 7.089 and 7.438 respectively, P<0.01). On the other hand, Bax protein expression in transfection groups increased, the AODs of Bax were 4.49 ± 0.61, 4.24 ± 0.37 and 3.95 ± 0.43, respectively. Compared with non-transfection group (3.12 ± 0.42), AOD of Bax protein in A549 celle in transfection groups increased significantly (t value was 7.469, 7.287 and 6.473 respectively, P<0.01). In the Ad-Gax transfection groups the lower Bcl-2/Bax ratio was, the higher the apoptotic rate of A549 cells was (r= -0.49, P<0.01). Conclusion: Ad-Gax transfection can induce A549 cells apoptosis. Possible mechanism is that Gax can downregulate Bcl-2 protein expression and upregulate Bax protein expression, and A549 cells apoptosis is related to the Bcl-2/Bax ratio.
Apoptosis Induction of ZBB-006, a Novel Synthetic Diterpenoid, in the Human Hepatocellular Carcinoma Cell Line HepG2 in Vitro and in Vivo.

Abstract:Diterpenes, present in many medicinal plants, have been the focus of continuous studies for the development of new anticancer agents. ZBB-006 is a new synthetic diterpenoid derivative which exhibited significant anti-proliferation activity against various cancer cell lines in our previous study. Here, we investigated the antitumor effect of ZBB-006 and its potential mechanisms in the human hepatocellular carcinoma cell line HepG2, both in vitro and in vivo. We found that oral administration of ZBB-006 effectively suppressed the growth of HepG2 xenograft tumor in nude mice without body weight decline as compared with the control group. Meanwhile, the growth inhibitory effect of ZBB-006 on HepG2 cells was observed with MTT assay. Apoptosis induced by ZBB-006 in HepG2 cells was evidenced by DAPI staining and Annexin V/PI double staining assay. ZBB-006 also dissipated the mitochondrial membrane potential (ΔΨm) apparently as revealed by JC-1 staining. Furthermore, the cleavage of PARP, activation of caspase-3 and caspase-9 but not caspase-8 was demonstrated by western blot assay both in vitro and in vivo. Additionally, the proapoptotic protein Bax was markedly elevated, while the antiapoptotic protein Bcl-2 was downregulated. Collectively, our data indicated that ZBB-006 exerted a strong antitumor effect on HepG2 cells by initiating the mitochondrial-dependent apoptosis, and it has potential to be explored as a new promising therapeutic agent against human hepatoma.
Beta-Catenin Sirna Regulation Of Apoptosis- And Angiogenesis-Related Gene Expression In Hepatocellular Carcinoma Cells: Potential Uses For Gene Therapy

Abstract:The molecular mechanism responsible for hepatocellular carcinoma (HCC) development remains to be defined although a number of gene pathways have been shown to play an active role, such as Wnt/beta-catenin signaling. In this study, beta-catenin small interfering RNA (siRNA) was designed, synthesized, and transfected into HCC HepG(2) cells. RT-PCR and Western blot assays were performed to detect expression of altered genes and proteins, and the MTT assay was used to detect cell viability. Our data showed that beta-catenin mRNA and protein expression levels were effectively knocked down by beta-catenin siRNA and subsequently, tumor cell proliferation was significantly suppressed. Flow cytometry assay showed that tumor cells were arrested at the G(0)/G(1) phase of the cell cycles. Molecularly, expression of Smad3, p-caspase-3, and Grp78 protein were upregulated after 72 h of beta-catenin siRNA transfection, whereas expression of TERT, caspase-3, XIAP, MMP-2, MMP-9, VEGF-A, VEGF-C, and bFGF protein were reduced. However, there was no change between the expression of STAT3 and the HSP27 protein following transfection. The results from the current study demonstrated the importance of the Wnt/beta-catenin signaling pathway in regulation of gene expression in HCC. Further studies are required to investigate the role of this pathway in HCC development and targeting of this pathway to control HCC.
[Hepatitis B Virus X Protein Suppressing Adriamycin-Induced Apoptosis of HepG2 Cells].

Abstract:OBJECTIVES:To investigate the effect of hepatitis B virus X protein (HBx) on adriamycin-induced apoptosis of hepatocellular carcinoma cells. METHODS:HBx gene fragment was amplified from subtype adr HBV plasmid by PCR, and inserted into Hind III and Kpn I sites of green fluorescent protein (GFP) eukaryotic expression vector pEGFP-C1 to construct recombinant pGFP/HBx. The pEGFP-C1 and pGFP-HBx were introduced into HepG2 cells by Lipofectamine 2000 to obtain HepG2 cells expressing GFP. GFP-HBx fusion protein was selected using G418. The expression of HBx gene was demonstrated by RT-PCR analysis. HepG2, HepG2/GFP and HepG2/GFP-HBx cells were treated with adriamycin (2.5 microg/ml), and apoptosis of the cells was determined by their morphological changes, trypan blue exclusion, and flow cytometry analysis. RESULTS:Under a fluorescence microscope, visible expression of GFP and GFP-HBx fusion proteins were observed in HepG2/GFP and HepG2/GFP-HBx cells, even after growing over 70 generations. RT-PCR analysis showed that HBx gene was expressed in HepG2/GFP-HBx cells. Trypan blue exclusion showed adriamycin induced time-dependent cell death in HepG2 and HepG2/GFP cells while no significant cell death was observed in HepG2/GFP-HBx cells. Flow cytometry analysis showed that apoptosis rates in HepG2/GFP-HBx (3.94%) cells were significantly lower than those in HepG2 (59.03%) and HepG2/GFP cells (61.38%) at 36 hours after the adriamycin treatment (P < 0.01). No significant differences of apoptosis rates of HepG2/GFP-HBx (3.94%) and of the untreated cells (2.12%, 2.78%, 2.55%) (P > 0.05) were observed. CONCLUSION:A HepG2 cell line expressing GFP and GFP-HBx fusion proteins was successfully established. HBV X protein blocks adriamycin-induced apoptosis of these HepG2 cells.
