Abstract:Hepatic satellite cells (HSCs) are the main cell type involved in the development of liver fibrosis and have been recognized as the important cellular source of extracellular matrix (ECM). NADPH oxidase (NOX) catalyzes the generation of reactive oxygen species (ROS), regulates signal transduction in HSCs, and thereby plays a key role in the pathogenesis of hepatic fibrosis. ROS generated by NOX promotes proliferation and inhibits apoptosis of HSCs by activation of mitogen-activated protein kinase and phospha-tidylinositol-3 kinase/Akt signaling pathways, thus contributing to the development of liver fibrosis. Inhibition of NOX activation to generate ROS and NOX-mediated signal transduction induces HSC apoptosis. Therefore, drugs that target specific NOX can be expected to be useful in arresting the progression of liver fibrosis.
