Abstract:Objective:To improve the synthetic technology of arotinolol hydrochloride.Methods and Results:2,5-Thiophenedicarboxylic acid as starting material was firstly treated with methanol to give 2,5-dicarboxythiophene dimethyl ester,from which 5-acetyl-thiophene-2-carboxylic acid,a key intermediate,was produced via Claisen condensation with ethyl acetate and by hydrolysis and decarboxylation.Then 5-(2-mercapto-4-thiazolyl)-2-thiophenecarboxamide obtained from the key intermediate through four-step reactions was reacted successively with epichlorohydrin,tert-butylamine and concentrated hydrochrolic acid to gain the target compound,of which the overall yield was 16.3% and the structure was confirmed by ESI-MS and 1H NMR.Conclusion:The improved synthetic technology of arotinolol hydrochloride simplifies its synthesis process,increases its yield and promotes its industrial production.
