Abstract:In this research paper, purified goat mammary epithelial cell line was obtained by culturing dispersed cells routinely and purifying them with time-controlled trypsinization. Results indicated that: large numbers of dispersed mammary cells could be obtained by digesting goat mammary tissue with 150 U/mL collegenaseⅠand 150 U/mL hyaluronidase for 4 h at 37 ℃, which is the feasible method for collection goat mammary cells. Adding hydrocortisone, insulin-like growth factor-I (IGF-I), epithelial growth factor (EGF) and insulin-transferrin-seuim (ITS) to RPMI1640 or DEME/F12 medium, which supplied with 10 fetal bovine serum and antibiotics, was benefical to the goat mammary cell growth significantly. The light microscope structure and perspective electron microscope ultrastructure of goat mammary cell indicated that: goat mammary cells formed island monolayer aggregates and dome-like structure. The seventh generation mammary cell cytoplasms had plenty of secretive vesicles and Golgi apparatus, which showed that the seventh generation mammary had vigorous secretive activity and there wasn’t any sign of apoptosis.

Abstract:This study sought to establish an in vitro lactating BMEC (bovine mammary epithelial cell) model, which may maintain the native function for a period of time. Mammary tissues of midlactation Holstein dairy cows were dispersed and cultured in a medium containing insulin, prolactin, hydrocortisone, transferrin, epidermal growth factor and fetal calf serum. After the cells migrating from the tissue reached similar to 80% of confluency, the tissues were removed, and secretory epithelial cells were enriched by digesting with 0.25% trypsin repeatedly to remove fibroblasts. The BMEC cells plated on plastic dishes displayed a monolayer, cobblestone, epithelial-like morphology and formed alveoli-like structures and island monolayer aggregates which are the typical characteristics of the mammary epithelial cells. The isolated cells were identified as of epithelial origin by staining with antibody against cytokeratin 18. A one-half logarithmically growth curve and abundant microvilli and cytoplasmic lipid droplets were observed in these cells. The transcription of the alpha(s1) casein gene and synthesis of alpha(s) caseins were also detected in the model. Thus, our lactating BMEC model can be an effective model in vitro for studies of milk synthesis in the bovine mammary gland.

Abstract:The objective of this study was to determine how bovine mammary protein profiles vary during lactation and the dry period. Three lactating and 3 nonlactating cows were selected for mammary gland tissue sampling. Compared with the mammary proteins in nonlactating cows, a total of 60 differentially expressed proteins (DEP, including 57 upregulated and 3 downregulated) were identified in lactating cows using 2-dimensional difference gel electrophoresis combined with mass spectrometry. These DEP included enzymes and proteins associated with various macromolecular metabolic processes, and appeared to promote the increased metabolic activity associated with, milk synthesis and secretion. The increased DEP were primarily related to initiation, maintenance, and involution of lactation, and included proteins involved in glycolysis/gluconeogenesis, the tricarboxylic acid cycle, the pentose phosphate pathway, oxidative phosphorylation, aminoacyl-transfer RNA biosynthesis, and fatty acid biosynthesis. Identified DEP were further validated by real-time, reverse-transcription PCR and Western blot. Five new DEP associated with lactation were uniquely identified. This work provided some protein-associated insights to facilitate further investigation of the mechanisms underlying lactation in dairy cows.
The Effects of Insulin and Prolactin on an Epithelial Cell Line from Mammary Gland of Dairy Goat

Abstract:Mammary epithelial cells with lactational function can be a valuable cellular model for research of the development and regulation of the mammary gland.This paper describes some aspects of function of an epithelial cell line from the mammary gland of the dairy goat.SDS-PAGE,triglyceride and lactose content of cultured cells were used to assess synthetic function of cells and the effects of exposure to insulin and prolactin.Results show that goat mammary epithelial cells can synthesize fat,proteins and lactose when they were cultured in DMEM-F12 medium with added EGF,IGF-1,ITS and FBS.There were no obvious changes after 48h treatment with additional insulin.Prolactin added to the basal medium significantly increased synthesis of proteins and lactose.A mammary gland epithelial cell line from goats which has lactational function has been established.This outcome provides a valuable and convenient model system.

Abstract:A ruminant mammary tissue culture that accurately reproduces mammary function in vitro would be a valuable tool in studies of ruminant lactation. This study was conducted to establish an in vitro model for culture of lactating bovine mammary tissue. The tissue still maintained the activity at 72 h, which was indicated by the value of MTT and oligonucleosomal fraction of cellular DNA. Under the conditions used in this study, the gene expression of alphas1 casein was highest at 8 h, and then declined at 12 and 24 h. The results suggest that this primary tissue culture system may be useful in studying the effects of different combinations of amino acids on milk protein in terms of alphas1 casein in vitro.

Abstract:Mastitis in cows is caused by the inflammation of the mammary glands due to an infection by external pathogenic bacteria. Mammary gland epithelial cells, which are in direct contact with the external environment, are responsible for the first line of defense of the mammary gland against pathogenic bacteria, playing an essential role in immune defense. To investigate the mechanism of bovine mammary epithelial cells in the inflammatory process, we treated the cells with LPS for 12 hours and analyzed the changes in mRNA by transcriptome sequencing. The results showed that compared to the control group, the LPS treatment group had 121 up-regulated genes and 18 down-regulated genes. GO and KEGG enrichment analysis revealed that these differential genes were mainly enriched in the IL-17 signaling pathway, Legionellosis, Cytokine-cytokine receptor interaction, NF-kappa B signaling pathway, and other signaling pathways. Furthermore, the expression of GRO1 and CXCL3 mRNAs increased significantly after LPS treatment. These findings provide new insights for the treatment of mastitis in cows in the future.
Establishment of goat mammary organoid cultures modeling the mammary gland development and lactation

Abstract:Background: Although several cell culture systems have been developed to investigate the function of the mammary gland in dairy livestock, they have potential limitations, such as the loss of alveolar structure or genetic and phenotypic differences from their native counterparts. Overcoming these challenges is crucial for lactation research. Development of protocols to establish lactating organoid of livestock represents a promising goal for the future. In this study, we developed a protocol to establish a culture system for mammary organoids in dairy goats to model the mammary gland development and lactation process. Results: The organoids cultured within an extracellular matrix gel maintained a bilayer structure that closely resembled the native architecture of mammary tissue. The expansion of mammary organoids was significantly promoted by growth factors containing epidermal growth factor and fibroblast growth factor 2 whereas the proliferative index of the organoids was significantly inhibited by the treatment with WNT inhibitors. Upon stimulation with a lactogenic medium containing prolactin, the mammary organoids exhibited efficient lactation, characterized by the accumulation of lipid droplets in the lumen space. The lactation could be sustained for more than 3 weeks. Importantly, the expression patterns of genes related to fatty acid synthesis and milk proteins in lactating organoids closely mirrored those observed in mammary tissues. These observations were confirmed by data from proteomic analysis that the bulk of milk proteins was produced in the lactating organoids. Conclusion: This study is the first to establish a mammary organoid culture system modeling the mammary gland development and lactation process in ruminants. The efficient induction of lactation in ruminant mammary organoids holds promises for advancing the field of cell-based milk bio-manufacture in the food industry.
Establishment of a bovine rumen epithelial cell line

Abstract:Rumen epithelium plays an essential role in absorption, transport, and metabolism of short-chain fatty acids, the main products of rumen fermentation, and in preventing microbes and other potentially harmful rumen contents from entering the systemic circulation. The objective of this study was to generate an immortal rumen epithelial cell line that can be used as a convenient model of rumen epithelial cells in vitro. We isolated primary rumen epithelial cells from a steer through trypsin digestion and transduced them with lentiviruses expressing the Simian Virus (SV) 40 T antigen. We cloned the transduced cells by limiting dilution. Western blotting analysis confirmed the expression of the SV40 T antigen in two single-cell clones. Cells from one clone, named bovine rumen epithelial clone 1 (BREC1), displayed a flat and squamous morphology in culture. RNA sequencing revealed that BREC1 cells expressed many markers of epithelial cells, including keratins, the epidermal growth factor receptor, and the short-chain fatty acid transporters monocarboxylic acid transporter (MCT) 1 (MCT-1) and MCT-4. RNA sequencing revealed that BREC1 cells expressed key enzymes such as 3-hydroxymethyl-3-methylglutaryl-CoA lyase and 3-hydroxy-3-methylglutaryl-CoA synthase 1 involved in ketogenesis, a unique function of rumen epithelial cells. RNA sequencing also revealed the expression of genes encoding tight junctions, desmosomes, anchoring junctions, and polarized plasma membranes, structures typical of epithelial cells, in BREC1 cells. Cell proliferation assays indicated that BREC1 cells were similar to primary rumen epithelial cells in response to insulin-like growth factor 1, insulin, and butyrate. In conclusion, BREC1 is not only a convenient but an appropriate model for studying the factors and mechanisms that control proliferation, apoptosis, differentiation, nutrient transport, metabolism, and barrier function in rumen epithelium.

Abstract:Background: Bovine oviduct epithelial cells are widely used in co-culture experiments to improve early embryonic development and in vitro fertilization in embryo transfer programmes for domestic animals. Methods: The present study compares different methods for harvesting and culture of bovine oviduct epithelial cells in order to optimize handling. Bovine oviduct epithelial cells were mechanically or enzymatically isolated and cultured on glass, on permeable membranes, or in suspension. Growth of the cells and their state of differentiation was examined by means of classical staining methods, immunohistochemistry and electron microscopy. Results: Initial cell suspensions contained sheets of ciliated and nonciliated (secretory) cells; 24 h after seeding, free floating epithelial cells formed vesicles with cilia on their external surface. First adhesion of cells was seen 72 h after seeding. Later on, cells grew continuously and confluent monolayers were formed after 7 days. Results were identical after mechanical or enzymatical cell harvesting and were identical on both substrata tested, i.e., on glass and on permeable membranes. Light and electron microscopy proved the monolayers to resemble a polarized, simple, cuboidal to columnar epithelial membrane with intact junctional complexes and numerous apical microvilli. Their epithelial nature was established by immunostaining for cytokeratins. Cilia were missing and secretory granules were scarce. A layer of acidic glycoprotein material was demonstrated on the apical surface. Monolayers of bovine oviduct epithelial cells stored lipid droplets and large quantities of glycogen. About 50% of the seeded cells did not adhere but survived in the culture medium as free floating cells. These suspended cells maintained morphological criteria of differentiation (cilia and secretory granules) until day 12 of culture. Proliferation rates of cultivated cells were determined by counting mitoses and by immunostaining with MIB1 antibody. Results showed coincidence of rapid proliferation and morphological dedifferentiation of monolayers. Suspended cells, by contrast, did not proliferate but retained cellular differentiation under identical culture conditions. Conclusions: The results strongly suggest that monolayers of bovine oviduct epithelial cells will not fully substitute for original oviduct epithelium when used in co-culture experiments after in vitro fertilization.
Effect of Semen Vaccariae and Taraxacu Mogono on Cell Adhesion of Bovine Mammary Epithelial Cells

Abstract:The aim of this study is to reveal the regulation mechanism of the effect of Semen vaccariae and Taraxacu mogono on the cell-cell adhersion molecule, E-cadherin and β-catenin on the proliferation role and secretion function of bovine mammary epithelial cells cultured in vitro. Firstly, the epithelial character of bovine mammary epithelial cells was authenticated using immunofluorescence, then the cell grow curve was observed and investigated after S. vaccariae and T. mogono treatment. On the effect of S. vaccariae and T. mogono, cell adhesion molecules E-cadherin, β-catenin and CycinD1 mRNA and protein were detected by qRT-PCR and Western blotting, respectively. The results showed that the cellular keratin 18 expressed positively and proliferated vigorously after S. vaccariae and T. mogono treament. The mRNA and protein levels of E-cadherin and CycinD1 were remarkably higher (P<0.05) in 36 h after S. vaccariae and T. mogono treatment. The cell proliferation at 36 h was increased significantly (P<0.05). In conclusion, S. vaccariae and T. mogono have a positive impact on the cell proliferation and an effect on the adhesion molecules E-cadherin, β-catenin and CycinD1 in the Wnt signaling pathway.

Abstract:The mammary gland, crucial for milk production in mammals, presents challenges for in vitro study due to its complex structure and limited cell lifespan. We addressed this by introducing the SV40 large T antigen into primary mammary epithelial cells (MECs) from sheep, creating an immortalized T-tag MEC line. This line, stable for over 50 passages, maintained typical epithelial cell morphology during long-term culture. Through transcriptome sequencing and validation, we discovered 3833 differentially expressed genes between MECs and T-tag MEC line, encompassing key biological processes and signaling pathways like cell cycle, p53, and cancer. The cell line, expressing MEC markers (KRT8, KRT18, proliferating cell nuclear antigen, SV40, CSN2, and acetyl-CoA carboxylase alpha), proved capable of synthesizing milk fat and protein. Despite its infinite proliferation potential, the T-tag MEC line showed no tumor formation in mice or cell migration in vitro, indicating stability. This development offers a valuable resource for studying MECs in dairy sheep, facilitating the advancement of long-term culture systems and in vitro lactation bioreactors.
Transdifferentiation of goat ear fibroblasts into lactating mammary epithelial cells induced by small molecule compounds

Abstract:Mammary epithelial cells are the only cells in the mammary glands that are capable of lactation and they are ideal for studying cellular and molecular biology mechanisms during growth, development and lactation of the mammary glands. The limiting factors in most of the currently available mammary epithelial cells are low cell viability, transgenerational efficiency and lactation function that renders them unsuitable for subsequent studies on mammary gland's cellular and lactation mechanisms and utilizing them as bioreactors. Hence, new methods are required to obtain mammary epithelial cells with high transgenerational efficiency and lactation function. In this study, transdifferentiation of goat ear fibroblasts (GEFs) into goat mammary epithelial cells (CiMECs) was induced in only eight days by five small molecule compounds, including 500 μg/mL VPA, 10 μM Tranylcypromine, 10 μM Forskolin, 1 μM TTNPB, 10 μM RepSox. Morphological observation, marker genes comparison, specific antigen expression and comparison of gene expression levels by transcriptome sequencing between the two types of cells that led to the primary deduction that CiMECs have similar biological properties to goat mammary epithelial cells (GMECs) and comparatively more lactation capacity. Therefore, we establish a novel reprogramming route to convert fibroblasts into CiMECs under fully chemically conditions. This study is expected to provide an in vitro platform for understanding cellular mechanisms such as mammary epithelial cells' fate determination and developmental differentiation, and also to find a new way to obtain a large number of functional mammary epithelial cells in vitro.

Abstract:The mammary gland is a unique organ with the ability to undergo repeated cyclic changes throughout the life of mammals. Among domesticated livestock species, ruminants (cattle and buffalo) constitute a distinct class of livestock species that are known milk producers. Cattle and buffalo contribute to 51 and 13% of the total milk supply in the world, respectively. They also play an essential role in the development of the economy for farming communities by providing milk, meat, and draft power. The development of the ruminant mammary gland is highly dynamic and multiphase in nature. There are six developmental stages: embryonic, prepubertal, pubertal, pregnancy, lactation, and involution. There has been substantial advancement in our understanding of the development of the mammary gland in both mouse and human models. Until now, there has not been a thorough investigation into the molecular processes that underlie the various stages of cow udder development. The current review sheds light on the morphological and molecular changes that occur during various developmental phases in diverse species, with a particular focus on the cow udder. It aims to explain the physiological differences between cattle and non-ruminant mammalian species such as humans, mice, and monkeys. Understanding the developmental biology of the mammary gland in molecular detail, as well as species-specific variations, will facilitate the researchers working in this area in further studies on cellular proliferation, differentiation, apoptosis, organogenesis, and carcinogenesis. Additionally, in-depth knowledge of the mammary gland will promote its use as a model organ for research work and promote enhanced milk yield in livestock animals without affecting their health and welfare.

Abstract:The mammary gland is an important organ for mammals to feed the offspring. It provides sufficient nutrients to the offspring by secreting milk. The healthy development of the mammary gland is of great significance to lactation and the survival rate of the offspring. The mammary gland develops through different stages, including embryonic period, puberty, pregnancy, lactation and regression period. The mammary gland is a highly dynamic organ. At different developmental stages, its morphology will undergo significant changes and the cells undergo extensive changes. Adipose tissue is an important part of the mammary gland. In the process of mammary gland development and circulatory remodeling, adipose tissue exhibits regular morphological and functional changes. The dynamic change of mammary gland adipose tissue is a significant feature of mammary gland circulatory development and reconstruction, and it is mainly manifested as changes in morphology and quantity. For example, adolescent mammary gland accounts for more adipocytes, the ratio of pregnancy and lactation decreases, and the ratio of degenerative phase increases, and this process will be repeated in different reproductive cycles. Studies have confirmed that adipose tissue can secrete special physiological factors to regulate the function of epithelial cells and the development of mammary glands, and has the potential to convert with epithelial cells. It is one of the indispensable cellular components in the development cycle of mammary glands. In addition, the abnormal development of adipose tissue also affects the development of epithelial cells and the function of secreting milk. However, it is still unclear how mammary gland adipocyte change in different periods, what is the mechanism, and what functions they play in mammary gland development. In addition, the research on mammary adipose tissue mainly focuses on mice, and there are few researches on other animals, which makes the research on mammary adipose tissue lack species integrity. In summary, this review synthesizes the relevant progress of mammary gland adipose tissue in recent years, and provides basic data for the follow-up study of the mechanism of adipose tissue regulating mammary gland development.
In vitro multipotent differentiation and barrier function of a human mammary epithelium

Abstract:As demonstrated by a variety of animal studies, barrier function in the mammary epithelium is essential for a fully functioning and differentiated gland. However, there is a paucity of information on barrier function in human mammary epithelium. Here, we have established characteristics of a polarizing differentiating model of human mammary epithelial cells capable of forming a high-resistance/low-conductance barrier in a predictable manner, viz., by using MCF10A cells on permeable membranes. Inulin flux decreased and transepithelial electrical resistance (TEER) increased over the course of several days after seeding MCF10A cells on permeable membranes. MCF10A cells exhibited multipotent phenotypic differentiation into layers expressing basal and lumenal markers when placed on permeable membranes, with at least two distinct cell phenotypes. A clonal subline of MCF10A, generated by culturing stem-like cells under non-adherent conditions, also generated a barrier-forming epithelial membrane with cells expressing markers of both basal and lumenal differentiation (CD10 and MUC1, respectively). Progressive changes associated with differentiation, including wholesale inhibition of cell-cycle genes and stimulation of cell and tissue morphogenic genes, were observed by gene expression profiling. Clustering and gene ontology categorization of significantly altered genes revealed a pattern of lumenal epithelial-cell-specific differentiation.

Abstract:Although research on dairy goat mammary gland have referred extensively to molecular mechanisms, research on lines of dairy goat mammary epithelial cells (MECs) are still rare. This paper sought to establish an immortal MEC line by stable transfection of human telomerase. MECs from a lactating (45 days post-parturition) Xinong Saanen dairy goat were cultured purely and subsequently transfected with a plasmid carrying the sequence of human telomerase. Immortalized MECs by human telomerase (hT-MECs) exhibited a typical cobblestone morphology and activity and expression levels of telomerase resembled that of MCF-7 cells. hT-MECs on passage 42 grew vigorously and 'S' sigmoid curves of growth were observed. Moreover, hT-MECs maintained a normal chromosome modal number of 2n=60, keratin 8 and epithelial membrane antigen (EMA) were evidently expressed, and beta-casein protein was synthesized and secreted. Beta-casein expression was enhanced by prolactin (P<0.05). Lipid droplets were found in hT-MECs, and messenger RNA levels of PPARG, SREBP, FASN, ACC and SCD in hT-MECs (passage 40) were similar to MECs (passage 7). In conclusion, the obtained hT-MEC line retained a normal morphology, growth characteristics, cytogenetics and secretory characteristics as primary MECs. Hence, it can be a representative model cell line, for molecular and functional analysis, of dairy goat MECs for an extended period of time.
Transcriptome analysis of mammary epithelial cell between Sewa sheep and East FriEsian sheep from different localities

Abstract:Mammary epithelial cells, the only milk-producing cell type in the mammary gland, undergo dynamic proliferation and differentiation during pregnancy, culminating in lactation postpartum. The East FriEsian sheep ranks among the world's most prolific dairy breeds, while the Sewa sheep, a unique dual-purpose breed autochthonous to the Qinghai-Tibet Plateau, exhibits significantly lower milk production. Employing tissue culture methods, we successfully established mammary epithelial cell lines from both breeds. Morphological assessment of mammary epithelial cells and immunofluorescence identification of Cytokeratin 7 and Cytokeratin 8 confirmed the epithelial identity of the isolated cells. Subsequent RNA-seq analysis of these in vitro epithelial cell lines revealed 1813 differentially expressed genes (DEGs). Among these, 1108 were significantly up-regulated and 705 were down-regulated in Sewa epithelial sheep cells compared to East FriEsian epithelial cells. KEGG enrichment analysis identified cellular processes, environmental information processing, human diseases, metabolism, and organismal systems as the primary functional categories associated with DEGs. Gene ontology (GO) terms annotation, categorized into molecular function, biological processes, and cellular component, yielded "binding and catalytic activity," "molecular function regulator activity," and "cellular process," "biological regulation," and "regulation of biological process" as the top three terms within each domain, respectively. Clusters of Orthologous Groups of proteins (KOG) classification further revealed that "signal transduction mechanisms" accounted for the largest proportion of DEGs among all KOG categories. Finally, based on these analyses, ATF3 and MPP7 were identified as promising candidate genes for regulating lactation.

Abstract:The mammary gland bioreactor is an efficient and novel technique to produce functional proteins,which has great commercial value.The article reviews the current status of construction of expression vector and ways of gene transfer,the two key procedures in mammary gland bioreactor.Moreover,the article discusses the prospect of mammary gland bioreactor based on the conjunction of gene targeting and somatic cloning.

Abstract:The immense potency of bioactive molecules of human breast milk and importance of breastfeeding is known worldwide.Less attention has been paid on the cellular constituents and properties of breast milk.In the past decade,a research has been done by multidimensional approach to investigat the cells human milk.Technological advances have played an important role in this work,which has resulted in the breakthrough discovery of breast milk stem cells with self renewal and multilineage potentials.Regenerative medicine may benefit from utilizing these cells.Breast milk stem cells are transferred to the offspring during breastfeeding.This contributes substantially to infant health.This review summarizes the current knowledge on the potential of breast milk stem cells,with emphasis on their origin,properties and future applications.

Abstract:Application of high-throughput sequencing and screening help to detect the transcriptional and metabolic discrepancies in organs provided with various levels of nutrients. The influences of individual essential amino acid (EAA) administration on transcriptomic and metabolomic profilings of bovine mammary epithelial cells (BMECs) were systematically investigated. A RNA sequencing and liquid chromatography-tandem mass spectrometry generated a comprehensive comparison of transcriptomics, non-targeted metabolomics and targeted amino acids profilings of BMECs with individual EAA stimulation by turn. The sequencing data and raw LC-MS/MS data of samples were presented in the databases of Gene Expression Omnibus, MetaboLights and Figshare for efficient reuse, including exploring the divergences in metabolisms between different EAAs and screening valuable genes and metabolites regulating casein synthesis.
