Abstract:The purposes of this study were to examine technical details in deriving and maintaining rabbit embryonic stem (rES) cell lines and to analyze their characteristics. When STO cells were used as feeder cells, no rES cell lines were established using either intact blastocysts or inner cell masses (ICMs). On the mouse embryonic fibroblasts (MEF) feeder, rES cell lines were efficiently (24%) derived. Addition of leukemia inhibitory factor (LIF) to the cells cultured on the MEF feeders further increased the derivation efficiency (57%) of rES cells. The fact that LIF induced serine-phosphorylation of STAT3 suggested LIF-dependent maintenance of rES cells. Most of the rES cell lines expressed AP, SSEA-4, Oct4, TRA-1-60, and TRA-1-81. Western blot or RT-PCR analysis also confirmed the expression of Oct4, Nanog, and Sox2. When induced to form EBs in vitro or injected to the severe combined immunodeficiency (SCID) mice, the rES cells generated embryoid bodies (EBs) and teratomas with three germ layers expressing the marker genes including MAP2, Desmin, and GATA4, respectively. In conclusion, rabbit ES cell lines can be efficiently established using our current protocols with LIF supplement. These ES cells express pluripotent stem cell markers and retain their capability to differentiate into different tissue cells. Furthermore, rES cells depend on LIF for self-renewal, likely via the JAK-STAT pathway.
Derivation, Characterization and Differentiation of a New Human Embryonic Stem Cell Line from a Chinese Hatched Blastocyst Assisted by a Non-Contact Laser System

Abstract:Currently worldwide attention has focused on the derivation of human embryonic stem cells (hESCs) for future therapeutic medicine. However, the majority of existing hESCs are directly or indirectly exposed to non-human materials during their derivation and/or propagation, which greatly restrict their therapeutic potential. Besides the efforts to improve culture systems, the derivation procedure, especially blastocyst manipulation, needs to be optimized. We adopted a non-contact laser-assisted hatching system in combination with sequential culture process to obtain hatched blastocysts as materials for hESC derivation, and derived a hESC line ZJUhES-1 of a Chinese population without exposure to any non-human materials during blastocyst manipulation. ZJUhES-1 satisfies the criteria of pluripotent hESCs: typically morphological characteristics; the expression of alkaline phosphatase, human telomerase reverse transcriptase and multiple hESC-specific markers including SSEA-3, SSEA-4, TRA-1-60, TRA-1-81, OCT-4, Nanog, Rex-1, Sox-2, UTF-1, Connexins 43 and 45, TERF-1 and TERF-2, Glut-1, BCRP-1/ABCG-2, GDF3, LIN28, FGF4, Thy-1, Cripto1/TDGF1, AC133 as well as SMAD1/2/3/5; extended proliferative capacity; maintenance of a stable male karyotype after long-term cultivation; and robust multiple-lineage developmental potentials both in vivo and in vitro. Moreover, ZJUhES-1 has distinct identity revealed from DNA fingerprinting. Our xeno-free blastocyst manipulation procedure may promote the progression toward clinical-grade hESC derivation.
Generation and Characterisation of Rabbit Monoclonal Antibodies Against the Native Cell Surface Antigens of Embryonic Stem Cells.

Abstract:Embryonic stem (ES) cells are potent resources for cell therapy, and monoclonal antibodies (mAbs) against native cell surface markers of ES cells could be useful tools for therapeutic applications. Here, we report the development of a feasible approach, which could be used in mass production, for experimentally producing rabbit mAbs against native cell surface antigens on the cell surface. Two of the 14 mAbs, which were selected at random, could be bound to the cell surface antigens of mES cells. The immunocytochemistry (ICC) and Western blot results showed that mAb 39 recognises conformational epitopes. The target antigen of mAb 39 was then successfully purified using an improved immunoprecipitation approach in which mAb was bounded to intact mES cells before the cells were lysed. The LC-LTQ mass spectrum analysis showed that the target antigen of mAb 39 was Glut3. This result was further confirmed by Western blot using commercially available antibodies against Glut3. Further experiments showed that mAb 39 exhibited an antiproliferative effect on mES cells. We also found that Glut3 was differentially expressed among the mES cell population as detected by flow cytometry.
Development of a rabbit monoclonal antibody group against Smads and immunocytochemical study of human and mouse embryonic stem cells.

Abstract:\We report here the development of a group of rabbit monoclonal antibodies against Smad1, Smad2, Smad3, and Smad5, and the immunocytochemistry (ICC) staining of human embryonic stem cells (hESC) and mouse embryonic stem cells (mESC). Eight New Zealand rabbits were immunized with synthesized peptides linked to KLH, and splenocytes from these rabbits were fused with rabbit immortal B cell 240E-W2. Resulting hybridomas producing anti-Smad monoclonal antibodies were screened by enzyme-linked immunosorbent assay (ELISA) with BSA-linked peptides. Clones were chosen for antibody production based on their activities in Western blotting and on paraffin-embedded human tissues, and the capacity of the antibodies in immunocytochemistry was demonstrated. Using these antibodies, we performed ICC staining on routinely cultured human and mouse embryonic stem cells, and showed that both cell types strongly express these genes. We propose that both hESCs and mESCs have the ability to transduce signals from both BMPs and TGFb/Activin.
Efficient Derivation of Human Embryonic Stem Cell Lines from Discarded Embryos Through Increases in the Concentration of Basic Fibroblast Growth Factor

Abstract:We describe the derivation and characterization of three novel human embryonic stem (hES) cell lines (YT1, YT2, YT3). One hES line (YT1) was obtained from six discarded blastocysts in a culture medium supplemented with 12 ng/ml basic fibroblast growth factor (bFGF), and two lines (YT2, YT3) were obtained from three discarded blastocysts in the same medium but supplemented with 16 ng/ml bFGF. These cell lines were derived by partial or whole embryo culture followed by further expansion after manual dissection of the passaged cells. These cells were passaged continuously for more than 6 or 8 months and possessed all of the typical features of pluripotent hES cell lines, such as typical morphological characteristics and the expression of hES-specific markers (TRA-1-60, TRA-1-81, SSEA-4, SSEA-3, alkaline phosphatase, Oct4, Nanog) and pluripotency-related genes (Oct4, Nanog, TDGF1, Sox2, EBAF, Thy-1, FGF4, Rex1). The lines maintained normal karyotypes after long-term cultivation. The karyotype of YT1 and YT3 was 46, XX, and that of YT2 was 46, XY. Pluripotency was confirmed by in vitro and in vivo differentiation, and genetic identity was demonstrated by DNA fingerprinting. Our results indicate that higher concentrations of bFGF at the early culture stage support efficient the hES cell derivation.

Abstract:OBJECTIVE To explore the methods of culture, identification and label of embryonic rat neural stem cells. METHOD The cells isolated from fetal rat hippocampus were identified with nestin immunocytochemical fluorescent staining. The cellular multiplication was observed by immunocytochemical fluorescence co-label after accession of BrDU. The neural stem cells (NSCs) were marked with fluorescent dye, bisbenzimide (Hoechest33342) and induced to differentiate. The differentiated cells were detected with Neuron Specific Enolase (NSE) and Glial Fibrillary Acidic Protein (GFAP) immunocytochemical fluorescent staining respectively. RESULT Nest-like clusters of neural stem cells were obtained in suspension and the cells could be differentiated into neurons and astrocytes which maintaining the main characteristics of NSCs after 8 passages of culture. The label efficiency of cells with Hoechest33342 was 97% and no attenuation of fluorescent brightness was observed after 8 passages of culture. The cellular fluorescence was observed in the NSCs and the differentiated cells. CONCLUSION The cells from embryonic rat hippocampus possessed the abilities of division, multiplication and self-renew, which were believed to be the main characteristics of NSCs of the central nervous system. The cells could be efficiently labeled with fluorescent dye and could be used as donor cells in experimental research on NSCs transplantation.

Abstract:Embryonic stem (ES) cells are pluripotent stem cells derived from the inner cell mass of the blastocyst. ES cell surface molecules are important for the identification, labeling, sorting, quality control and functional studies of ES cells. Currently, knowledge of ES surface molecules is limited. To identify new surface molecules, we generated a panel of rabbit monoclonal antibodies (rMabs) against mouse ES (mES) cells. We identified three monoclonal antibodies that interact with molecules on the mES cell surface and found that the expression of their respective antigens decreased upon mES cell differentiation. The antigen of the rMab ZjuESrMab29 was identified as granulocyte macrophage colony-stimulating factor receptor α (GM-CSFR α) by liquid chromatography–tandem mass spectrometry (LC–MS/MS). This study demonstrated that rabbit monoclonal antibody production via whole-cell immunization could be a practical method for the discovery of stem cell surface antigens.

Abstract:Mouse embryonic stem cells (mESCs) have played a key role in the newly emerging fields of stem cell research. The traditional derivation and culture of mESCs have been based on the use of mouse embryonic fibroblasts (MEFs) treated with exogenous leukemia inhibitory factor (LIF). However, the rapid senescence of MEFs, coupled with the high cost of LIF, has significantly hampered the widespread use of mESCs in stem cell research. Thus, we present a novel exogenous LIF-free culture system for general mESCs applications, comprising fibroblast-like cells derived from the rabbit spleen (RSFs). We demonstrated that mESCs cultured on RSFs (mESCs-RSFs) maintained all mESC features after prolonged LIF-free culture, including alkaline phosphatase, cell surface markers (SSEA-1), molecular markers (OCT-4, NANOG, TERT, REX-1), karyotype, and pluripotency. The high expression level of both LIF and WNT3A in the RSFs may account for their ability to maintain mESCs without exogenous LIF. Moreover, this exogenous LIF-free culture system was verified to be of microbiological quality through analysis with electron transmission microscopy.
EX VIVO ISOLATION AND EXPANSION OF RABBIT MESENCHYMAL STEM CELLS IN DEFINED SERUM-FREE MEDIA

Abstract:Introduction: Mesenchymal stem cells (MSCs) derived from rabbit bone marrow are routinely investigated in cardiovascular and orthopedic models for regenerative medicine. However, classical media used for generating MSCs are typically augmented with ill-defined supplements, such as fetal bovine serum, which increases safety concerns as well as inconsistency in MSC generation. We tested three commercial serum-free media for the generation and expansion of rabbit MSCs (rbMSCs). Materials and methods: RbMSCs were separated by adherent tissue culture and cultured in serum-free or serum-containing media. Cultured cells were characterized by phase contrast microscopy, cell-proliferation, flow cytometry, and qPCR analyses. Results: MesenCult (R) XF medium supported the generation and expansion of rbMSCs, whereas StemPro (R) hMSC and BD Mosaic hMSC media did not propagate freshly isolated rbMSCs. At passage 5, total cell yields for serum-containing and MesenCult (R) XF media were 4.0 +/- 0.16 x 108 and 1.9 +/- 0.28 x 109, and cumulative population doublings were 11.5 +/- 0.12 and 13.08 +/- 0.19, respectively. Colony formation efficiencies in MesenCult (R) XF were 21 to 26%, whereas those in serum-containing medium were 14 to 17%. Flow cytometry showed that rbMSCs in MesenCult (R) XF were positive for CD29, CD44 and CD73, and negative for CD34 and CD45. After corresponding differentiation, rbMSCs from MesenCult (R) XF could differentiate into adipogenic, osteogenic and chondrogenic lineages. qPCR analysis demonstrated that cells cultured in serum-free media had higher differentiation potentials than those cultured in serum-containing media. Discussion: rbMSCs were efficiently generated and expanded in MesenCult (R) XF, while maintaining the necessary characteristics attributed to MSCs for potential therapeutic use.
Research on the High Efficient Methodology of Establishment of 129s1/svimj Murine Embryonic Stem Cells.

Abstract:The high quality embryos of 129S1/SvImJ strain mouse were chosen after superovulation, and directly seeded in the irradiated frozen-thaw mouse embryonic fibroblast cells. Compared with the low concentration long time digestion, the high concentration short time digestion was adopted in the study, and 4 lines of mouse embryonic stem cells were successfully established by trypsinization of 0.25%trypsin-0.04%EDTA, which is the high concentration short time digestion method. The ratio of establishment cell lines is 36.3%, and the cell strains exhibited the typical ES cell characteristic identified by different ways including the AKP and karyotype assay, the differentiation potential in vivo and in vitro, and the examination of specific surface markers expression of Oct-4 and Nanog. The ES cell lines can be made scalable culture and used as an ideal experimental material for the further research.

Abstract:Embryonic stem cells were isolated from rabbit blastocysts derived from fertilization (conventional rbES cells), parthenogenesis (pES cells) and nuclear transfer (ntES cells), and propagated in a serum-free culture system. Rabbit ES (rbES) cells proliferated for a prolonged time in an undifferentiated state and maintained a normal karyotype. These cells grew in a monolayer with a high nuclear/cytoplasm ratio and contained a high level of alkaline phosphate activity. In addition, rbES cells expressed the pluripotent marker Oct-4, as well as EBAF2, FGF4, TDGF1, but not antigens recognized by antibodies against SSEA-1, SSEA-3, SSEA-4, TRA-1-10 and TRA-1-81. All 3 types of ES cells formed embryoid bodies and generated teratoma that contained tissue types of all three germ layers. rbES cells exhibited a high cloning efficiency, were genetically modified readily and were used as nuclear donors to generate a viable rabbit through somatic cell nuclear transfer. In combination with genetic engineering, the ES cell technology should facilitate the creation of new rabbit lines.

Abstract:Mammalian stem cells are invaluable research resources for the study of cell and embryonic development as well as practical tools for use in the production of genetically engineered animals and further therapeutics. It is important that we further our knowledge and understanding of a variety of stem cells from several different animal species before trials in humans commence. Here we describe methods for establishing rabbit embryonic stem (rES) cell lines with indefinite proliferation potential. rES cells attain maximum proliferation potential when cultured at a feeder cell density of one-sixth of that of full confluency. Higher and lower densities of feeder cells induced ES cell differentiation or division arrest. Fibroblast growth factor (FGF)2 can maintain the undifferentiated status of rES cells; however leukemia inhibitory factor (LIF) is dispensable. Under optimized conditions, rES cells could be passaged by trypsinization 50 times. This culture system enabled efficient gene transduction and clonal expansion from single cells. rES cells grew as flat monolayer cell colonies, as reported for monkey and human ES cells, and expressed pluripotency markers. Embryoid bodies and teratomas formed readily in vitro and in vivo, respectively. Characterization of ES cells from different species is important for establishing common features of pluripotency. We have demonstrated the similarity of ES cells between rabbit and humans. These cell lines could be applied directly using gene-targeting techniques, or in combination with induced pluripotent stem cells. Thus, rES cells are a suitable model for studying human transplantation therapy and disease treatments.

Abstract:Studies of cultured human embryos have provided insights into human peri-implantation embryogenesis but are limited by ethical and technical constraints. Here we assembled a 3D integrated embryo model (IEM) using naive human embryonic stem cells (hESCs) and hESC-derived proliferating trophoblasts. By comparing phenotypic cell fate and comprehensive single-cell transcriptome of IEMs with 3D-cultured human embryos, we reveal that IEMs with embryonic and extra-embryonic cell types mimic developmental landmarks and 3D morphological features of natural human post -implantation embryos, including pluripotent state transitions, epithelial-mesenchymal transition, anterior-posterior axis formation, amniotic cavity, yolk sac, bilaminar embryonic disc, extra-embryonic mesenchyme, primitive streak and primordial germ cells. Using IEMs, we unravel composition and gene expression patterns of embryonic lineages, and that trophoblasts secret WNT, BMP and TGFβ/Activin signals to instruct post-implantation embryogenesis . Our findings show that IEMs provide a scalable and tractable model for decoding regulatory mechanisms of human peri-implantation embryogenesis.
Optimization of culture condition on human embryonic stem cell line H1

Abstract:Objective:To establish the optimal culture condition for H1 line(46,XY,NIH Code WA01) according to medium and feeder cells and analyze their characteristics,and to improve clone survival after frozen-thawed.Methods: H1 as cultured on HFF-1(human fetal fibroblast,SCRC-1041,ATCC) or MEF feeders(mouse embryonic fibroblast,MEF) derived from ICR mice embryos.The basic medium was DMEM/F12 or Knock-outTM DMEM(K-DMEM).The experimental groups included MEF+DMEM/F12,MEF+K-DMEM,HFF+DMEM/F12 and HFF+K-DMEM.Alkaline phosphatase activity,immunofluorescence,RT-PCR and Giemsa staining were used to detect the biological characteristics of H1.Results: The morphology of H1 clones in MEF+DMEM/F12 group was regular and remained undifferentiated state and grew significantly fast.However,differentiation occured in clones in the MEF+K-DMEM group at day 4 after passages.The clones became flatten and differentiated in the HFF+DMEM/F12 and HFF+K-DMEM groups at day 3 after passages.In MEF+DMEM/F12 group H1 remained normal karyotype and expressed the markers for pluripotent cells,including positive expression for Oct4,SSEA-4,TRA-1-60,TRA-1-81,alkaline phosphatase activity,and negative for SSEA-1.Conclusion: The culture system differs substantially in different hES cell lines although they appear similar in the undifferentiated state.MEF+DMEM/F12 is sufficient for self-renewal and proliferation of H1.HFF-1 does not support H1 growth.

Abstract:In recent years, considerable attention has been paid to chicken embryonic stem cells (ESCs) studies in relation to extensive applications in gene therapy and regenerative medicine. However, the approaches used are still immature. In this study, we showed that the chicken ESCs clones with a clear border can express alkaline phosphatase and marker proteins such as SSEA-1, SOX2, and OCT4 stably. In addition, culture medium containing 10mol/L of vitamin C (VC) could significantly promote the proliferation of ESCs cells. Moreover, ESCs transfected with p:enhanced green fluorescent protein (pEGFP)-hTERT could be subcultured more than tenth generations in culture medium containing exogenous factors (mLIF+bFGF+hSCF) and VC, and these ESCs clone could still be regenerated following cryopreservation. Quantitative real-time polymerase chain reaction results showed that there was no significant difference between SSEA-1, SOX2, and OCT4 expression during ESCs immortalization and that the tenth generation of ESCs was still able to express marker proteins SSEA-1, SOX2, and OCT4. Our results showed that an immobilized system for ESCs was established, and the ESCs were cultured in vitro maintaining their pluripotency.

Objective: To analyze the morphological characteristics and colony forming capacity of early cultured rabbit adipose tissues-derived mesenchymal stem cells in vitro. Design, Time and Setting: The cytological in vitro study was performed at the Institute of Ophthalmology, and Central Laboratory, Eye and ENT Hospital, Fudan University from January to March 2008. Materials: Nine female New Zealand rabbits aged three months were obtained from the Shanghai Yingenyang Rabbit Room. Methods: Adipose tissue was harvested from the nape fat pad of the New Zealand rabbits following anesthesia. Primary cultured cells were established using type I collagenase and cell cultures were maintained with DMEM containing 0.1 volume fraction of fetal bovine serum. Cells were passaged when 80% was confluent. The second, third and fourth passages of cells were utilized for the study. Main Outcome Measures: The morphology of adipose tissues-derived mesenchymal stem cells was observed using inverted microscope. Cell phenotype was detected utilizing flow cytometry. The cloning capacity was determined. Results: Adipose tissues-derived mesenchymal stem cells were fibroblast-like and proliferated rapidly in vitro at the second, third and fourth passage. Flow cytometry showed that adipose tissues-derived mesenchymal stem cells expressed CD29 and PCNA. The colony-fonrang rates were respectively (8.0±0.6)%, (6.7±0.4)% and (4.6±0.5)% (F=12.18, P < 0.05). Conclusion: Rabbit adipose tissues-derived mesenchymal stem cells at early stage proliferate rapidly in vitro. The colony forming capacity is gradually decreased obviously with increased passage number.

Abstract:OBJECTIVE:To report the derivation and characterization of a new human embryonic stem cell (hESC) line NJGLLhES1.METHODS:From the inner cell mass of frozen-thawed human embryos and with ICR mouse embryonic fibroblasts as the feeder layer, we established a new human embryonic stem cell line, which was named NJGLLhES1. We detected the karyotype of the cell line, determined the expressions of alkaline phosphatase, the specific cell surface antigens SSEA-3, SSEA-4, TRA-1-60, TRA-1-81 and the marker gene Oct-4, and examined the formation of embryoids and teratomas.RESULTS:NJGLLhES1 was maintained for over 1 year in vitro, with the morphological characteristics of hESC, a normal karyotype, positive expressions of alkaline phosphatase and specific cell marker genes, and the potential of forming embryoids and teratomas.CONCLUSION:A new human embryonic stem cell line NJGLLhES1 was successfully established, which remains karyotypically and phenotypically stable, undifferentiated and capable of self-renewal and pluripotential differentiation.
Derivation and characterization of novel nonhuman primate embryonic stem cell lines from in vitro-fertilized baboon preimplantation embryos

Abstract:The development of nonhuman primate (NHP) embryonic stem cell (ESC) models holds great promise for cell-mediated treatment of debilitating diseases and to address numerous unanswered questions regarding the therapeutic efficacy of ESCs while supplanting ethical considerations involved with human studies. Here we report successful establishment and characterization of 3 novel baboon (Papio cynocephalus) ESC lines from the inner cell mass of intracytoplasmic sperm injection-derived blastocysts. Embryos were cultured in an improved baboon embryo in vitro culture protocol. The inner cell mass of blastocyst was laser-dissected and plated on mouse embryonic fibroblast feeder cell monolayer in the NHP ESC culture medium. Three cell lines with characteristic ESC morphology have been cultured through an extended period (>14 months), with 2 male cell lines (UT-1 and -2) and 1 female cell line (UT-3) displaying normal baboon karyotypes. Reverse transcription-polymerase chain reaction analysis confirmed that all 3 lines express primate ESC pluripotency markers, including OCT-4, NANOG, SOX-2, TERT, TDGF, LEFTYA, and REX-1. All 3 lines demonstrated positive immunocytochemical staining for OCT-4, stage-specific embryonic antigen-3, stage-specific embryonic antigen-4, TRA-1-60, and TRA-1-81. Baboon ESCs injected into NOD/SCID mice formed teratomas with all 3 germ layers. In addition, embryoid body-like spherical structures were derived and initial outgrowth was observed when embedded into extracellular matrix Matrigel. The ESC lines established in this NHP model have the potential to extend our knowledge in the fields of developmental biology, regenerative medicine, and future applications, including preclinical safety assessment of in vivo stem cell therapy.

The morphological characterization of adipos derived stromal cells at early stage was described. Expressions of CD29 and Proliferating Cell Nuclear Antigen were analyzed by flow cytometry. The Cloning capacity was determined using a ring cloning procedure. Data was statisticallly analyzed by software SPSS 16.0.RESULTS: ADSCs were successfully obtained and maintained which appeared fibroblast-like and proliferated rapidly in vitro. Flow cytometry showed that ADSCs expressed CD29 and PCNA. The colone-forming rate of passage 2 was (8.0 +/- 0.6)%, while they were (6.7 +/- 0.4)% and (4.6 +/- 0.5)% at passage 3 and passage 4, respectively. Statistical analysis indicated that the significant difference among the ADSCs at early stage.CONCLUSION: Rabbit ADSCs at early stage can selfrenew and proliferate rapidly in vitro. The colony formation capacity was gradually decreased obviously with passage number increased.
