Abstract:Stem cells have both the ability to self-renew and to differentiate. These cells can be either totipotent, pluripotent, multipotent or unipotent based on their transdifferentiating potential. Originally, stem cells can be found in the zygote as totipotent cells with the ability of forming a fully functioning organism. Cells from the inner cells mass (ICM) of an embryo are pluripotent and can differentiate into the cells forming the three germ layers. To date, research is progressively growing, focusing on understanding how both the extrinsic signaling pathways as well as the chromatin related intrinsic factors regulate, not only, proper stem cell functions, but also identity and activity. The functio of the core transcriptional factors like, but not limited to, Oct4, Nanog and Sox2 defines the stemness and transdifferentiation potential of the stem cells. Several signal transduction pathways, including LIF-STAT pathway, Wnt pathway and TGF-β pathway, are crucial for understanding the regulation of the stem cell pluripotency by affecting the before mentioned core pluripotency factors. Chromatin remodeling, DNA methylation, and post-translational histone modifications represent the major epigenetic mechanisms known to cooperate in stem cells. Epigenetic alteration of the chromatin state of genes does not affect the DNA sequences. Collectively, transcription factors, signal transduction pathways and epigenetics define cell fate and maintain cell pluripotency.
