Analysis of Soil Bacterial Diversity and Effective Control of Mango Anthracnose

Abstract:This study employed metagenomic sequencing technology to analyze the differences in diversity and abundance of soil bacterial from healthy and stunted mango. And the relationship between soil bacterial diversity around the roots of healthy and poorly growing mango and soil pH, organic matter, total nitrogen, total phosphorus and total potassium was elucidated through joint analysis with soil physical and chemical properties. A total of 32 healthy and stunted mango roots and soil samples were collected from Leye County, Baise City, China. Soil DNA was extracted and high-throughput sequencing was performed. In addition, so as to reduce the use of chemical fertilizers and pesticides, anthrax pathogen from mango and rhizosphere bacteria antagonized anthrax pathogens from healthy soil samples were screened and identified. The sequencing results showed that there were differences in bacterial diversity between healthy and unhealthy mango soils. The combined analysis of microbial diversity and soil physical and chemical properties found that Candidate, Acidobacterium, Pelagibius, Luteitalea, Blastocatellia and Sphingobium were significantly positively correlated with pH, Ktedonobacterales, Paraburkholderia, Streptosporangiales and Corynebacteriales were significantly negatively correlated. Burkholderia and Paraburkholderia showed a significant positive correlation with total nitrogen. Candidate, Pelagibius and Luteitalea showed a significant positive correlation. Solibacteraceae, Candidatus and Acidobacteriia showed a significant positive correlation with organic matter, while Sphingobium, Catenulisporales, Leclercia and Pelomonas showed a significant negative correlation. Acidobacteriia, Terriglobus and Sphingopyxis were significantly positively correlated with soil total phosphorus, yet Chloroflexales, Thermales, Chthoniobacterales and Candidate were significantly negatively correlated. Acidimicrobiales, Xanthomonadales and Rhodanobacter were positively correlated with total potassium in soil, but Verrucomicrobia was negatively correlated. In this study, the pathogen M1 was isolated from the anthrax spot of the infected mango. And M1 could infect mango fruits and lead to mango anthracnose. Leclercia adecarboxylata MHA-2-F1 strain was screened from mango rhizosphere soil to inhibit mango anthracrum. It also had five growth-promoting functions, including phosphorus solubilizing, calcium dissolving, IAA producing, siderophore-producing. MHA-2-F1 can not only effectively inhibit the infection of mangoes by Colletotrichum gloeosporioides, but also improving plant performances regarding number of roots and plant wet weight. The results providing insights for future research and the development of innovative and sustainable agricultural practices.
