Abstract:Objectve To evaluate the therapeutic efficacy of transplanting bone marrow mesenchymal stem cells(BMMSCs) that increase the amount of proteoglycan and collagen typeⅡcontent in early degenerating intervertebral discs.Methods An in vivo model was established to investigate the feasibility of marrow mesenchymal stem cells that cultured in vitro and marked with SPIO retention,and survived in disc space.In 2,4,6,8 weeks,immunohistochemical staining was used to determine the change of collagen type Ⅱcontent;spectrophotometry to determine the change of amount of proteoglycan with phlorglucinol.Magnetic resonance scanning was also performed to trace the implanted cells in discs.Result BMMSCs could maintain viability and proliferate in the rabbit intervertebral disc.The amount of proteoglycan and collagen type Ⅱcontent of the intervertebral in matrix synthesis in the experiment group was increased in 8 weeks.No changes were found in the model group.Conclusion Transplanted mesenchymal stem cells can delay or even prevent the degeneration of the intervertebral disc.The transplanted cells can migrate in the discs.
