Study on the Effect of HLA-G on Trophoblast Cell Multiplication

Abstract:Objective:To investigate that whether the human leukocyte antigen-G(HLA-G) gene could regulate trophoblast cell multiplication.Methods:This experiment was divided into three groups:the purpose siRNA group with titer of 5 ×108pfu/ml adenovirus infection liquid transfected JEG-3 cells,the non-specific siRNA group with titer of 109 pfu/ml adenovirus infection liquid transfected JEG-3 cells,and the control group did not produce siRNA.Using RNA interference technology to down-regulate the HLA-G gene expression in JEG-3,the non-radioactive cytotoxicity of the cells(the 490nm OD value) was determined.The results of the staining experiments and the colorimetric experiment were analyzed.Results:①After the experiment,the cells of the purpose siRNA group and the non-specific siRNA group were all stained in blue,but no stain of the cells of the control group;②There was no significant difference of OD490nm value between non-specific siRNA group and the control group(q=3.15,P 0.05);But the purpose of siRNA group was statistically significant difference than the non-specific siRNA group and the control group(q= 3.92,P0.05;q = 5.58,P0.01).Conclusions:This study reveals an important non-immune function for HLA-G in controlling trophoblast cell multiplication.
