The Cleavage of an in Vitro Transcript of the Protease Cathepsin B by Miniature Ribozymes

Abstract:Objective To detect the cleavage of an in vitro transcript of the protease cathepsin B protease by miniature ribozymes.Methods The secondary structure of cathepsin B mRNA was analyzed with the program RNAdraw.Ribozyme cleavage sites were chosen in accordance with the principles of miniature ribozyme design.Specific antisense sequences of miniature ribozymes were synthesized.The ribozyme was cloned into the Giardia canis viral vector to construct a recombinant vector flanked with long or short cathepsin B antisense RNA on each arm(CRzS or CRzL).Two control groups were established: PCB without the inserted ribozyme and GRzL flanked with GAPDH antisense RNA.The recombinant vector was linearized to detect the cleavage of cathepsin B mRNA.The cleavage of an in vitro transcript of cathepsin B by the miniature ribozyme was then analyzed with real-time quantitative RT-PCR.Results The two ribozymes CRzS and CRzL cleaved cathepsin B mRNA in vitro at a rate of 75.42% and 83.14%,respectively,according to real-time quantitative RT-PCR.In contrast,the PCB had cleavage activity of 23.0% and GRzL had cleavage activity of 0.02%.Conclusion The miniature ribozyme cleaved an in vitro transcript of cathepsin B,laying the foundation for inhibition of the protease cathepsin B in the body.
