Effect of spatial learning capacity and emotional behavior on kindled rats by pentylenetetrazole

Abstract:Objective Pentylenetetrazole(PTZ)is a chemical kindling agent used to examine the efficacy of potential anticonvulsants in rats.However,the extent to which PTZ mimics post-seizure symptoms of epilepsy has not been thoroughly examined.This study assessed whether PTZ-induced seizures produce cognitive and emotional deficits that mimic those observed in many epileptic patients.Methods Twenty-four male Sprague-Dawley rats were divided randomly into 2 groups:①PTZ group(n=16),in which rats were injected intraperitoneally with PTZ(35mg·kg-1·48h-1 in 0.9% saline);②the control group(n=8),in which rats received intraperitoneal injections of the 0.9% saline.Results There were no significant different in the time taken to swim to the hidden platform to escape between rats given PTZ and rats in control.But rats given PTZ spent less time in the quadrant where the platform was previously positioned(P<0.01),and exhibited emotional abnormalities in the forced swim test,the systematic handling test,and the open-field exploratory maze(P<0.01),similar to what was observed in human epilepsy.Conclusion Investigating the consequences of PTZ kindling in a Morris water maze and the open-field,we can find significant change on cognitive capacities and emotional consequences of rats given PTZ.These results indicate that PTZ kindling provides an useful model of post-seizure dysfunction,which can serve as a screen for potential treatments for those cognitive,emotional deficits that resemble those symptoms observed in human epilepsy.
