Abstract:The mouse is an excellent model for investigations of stroke and neural injury. However, there is a paucity of long term functional outcome measurements for the mouse. We, therefore, developed a sensorimotor functional test (corner test) and applied this test to a model of focal cerebral ischemia in the mouse. Male C57/6J mice (n=20) were subjected to embolic middle cerebral artery (MCA) occlusion. Reduction of cerebral blood flow (CBF) was measured by perfusion weighted MRI at 1 h after ischemia. The corner test, which is sensitive to chronic sensorimotor and postural symmetries, a general neurological test battery, and a foot fault test were performed between 2 and 90 days after ischemia. Infarct volume was measured at 90 days after ischemia. Multivariable analysis revealed that the corner test was highly predictive for infarct volume measured at 90 days after stroke, with R(2) values ranging from 0.73 to 0.93. The foot-fault test and neurological score did not detect chronic behavioral impairments. A significant (P<0.001) correlation between the infarct volume and the corner test was detected at 90 days after mild focal cerebral ischemia, whereas, there was no correlation between the infarct volume and neurological score or foot-fault. The data demonstrate that the corner test is a sensitive and objective test, which can be applied to evaluate long term functional outcome after stroke in the mouse.

Abstract:The purpose of this study was to develop a quantitative and objective method for evaluating neurological deficits in mice with focal cerebral ischemia. After middle cerebral artery occlusion (MCAO), the neurological deficits were evaluated 24 h later. We measured the mean angles, dominant angles and turns in a hanged test in which the mice were sticked on the wall, and the holding angles in an inclined plane test as well, Then we determined the cerebral infarct volumes, neuron density in hippocampus, cortex and subcortical areas 24 h after MCAO. The correlations among infarct volume, neuron density and neurological deficits were analyzed. We also compared the quantitative method with two typical complex methods of behavioral assessment. The effect of [pranlukast, 4-oxo-8-[p-(4-phenylbutyloxy) benzoylamino]-2-(tetrazol-5-yl)-4H-1-benzopyran hemihydrate] (ONO-1078), a neuroprotective agent, on ischemic injury was observed using this method. We found that the variables measured by both quantitative and typical behavioral methods significantly changed in the ischemic mice, and correlated with the infarct volumes and neuron densities. The quantitative variables well correlated with those of typical behavioral assessment, too. ONO-1078 inhibited ischemic injury and reduced the total scores of quantitative assessment. Thus, the quantitative method we developed is useful in evaluating neurological deficits of focal cerebral ischemia with the advantages of objectivity, quantification, simplicity and non-invasion, and can be used in the evaluation of neuroprotective effects of drugs.

Abstract:Multi-frequency electrical impedance tomography (MFEIT) reconstructs the image of conductivity inside the human body based on the dependence of tissue conductivity on frequency. As there exist differences in the conductivity over frequency between blood, ischemic cortical tissue and normal cortical tissue, MFEIT has potential application in the detection of acute stroke. However, because the conductivity distribution of the human head is highly inhomogeneous and the conductivities of normal head tissue and stroke lesion tissue both change with frequency, the anomaly and normal head tissues are often mixed together in the reconstructed image, which makes it difficult to discern the anomaly. Here we present a spectral decomposition frequency-difference (SD-FD) imaging algorithm in an attempt to address this issue: firstly, we reconstruct so-called EIT spectral images according to the conductivity spectra of tissues; secondly, we obtain the EIT image of the anomaly from the spectral images by using independent component analysis. The results show that the proposed algorithm is capable of detecting the anomaly in a numerical head phantom, as well as in a realistic human head tank with frequency-dependent and heterogeneous conductivities distribution. The proposed SD-FD algorithm may support MFEIT use for human stroke imaging in the future.

Abstract:Objective To comparatively study on the application of four kinds of neurological function rating systems including forelimb placing test,Longa 5 scores,Ludmila Belayev 12 scores,and Elevated Body Swing Test at the period of chronic recovery on focal cer -ebral ischemic models in rats .Methods SD rats were randomly divided into MCAO ( middle cerebral artery occlusion ) group and Sham group:suture method was used in MCAO ,reperfusion after 90 minutes.Immediately Pulled out suture while plugging to middle cerebral artery in Sham group.To assess and analyze the outcomes by forelimb placing test ,Longa 5 scores,12 scores,and EBST neurological function rating systems in two groups at 2h,1d,3d,7d,10d,14d,17d,21d,24d,28d.Results The analysis of scores at different time point between the two groups showed:no statistic significance existed at 7d,10d,14d,17d,21d,24d,28d in Longa,no statistic significance existed at 21d,24d,28d in EBST.Conclusion Forelimb placing test and 12 scores neurological function rating systems can well evaluate the neurological condition of rats in chronic convalescence ,Longa 5 scores and EBST not effectively assess the neurological condition of rats in chronic convalescence .

Abstract:Objective To discuss neurological behavior in focal cerebral ischemia in rats.Methods Cerebral ischemia was achieved by occluding the middle cerebral artery with an intraluminal suture technique in rats, neurological deficit score evaluation and infarct volume estimate was used.Results Up till 24 hours,there are significant difference in rats which used Nagaoka?Bederson?zea-Longa and Zhang evaluation(P0.05), but there is no significant difference in rats which used Zausinger evaluation. Moreover, there is highly significant difference in infarct volume of rats(P0.01)and only Bederson evaluation is correlative to infarct volume in 24 hours(r=0.91613,P0.05).Conclusion A new neurological behavior method representing cerebral ischemia will be suggested because available evaluations don't reflect pathophysiologic status of cerebral ischemia exactly.
Using MicroPET Imaging in Quantitative Verification of the Acupuncture Effect in Ischemia Stroke Treatment

Abstract:Acupuncture has been indispensable in Chinese medicine. However, its function still remains elusive. This paper studies the effect of acupuncture in ischemia stroke treatment using the Sprague Dawley rat animal model. We induced focal cerebral ischemia in rats using the middle cerebral artery occlusion (MCAO) procedure. For each rat in the real acupuncture group ( n = 63), the sham acupoint treatment group ( n = 62) and the blank control group ( n = 30), we acquired 3-D fluorodeoxyglucose-microPET images at baseline, after MCAO and after treatment, respectively. Then, we measured the changes of the injury-volume in the right hemisphere of these rats. The measurements showed that real acupuncture slightly reduced the injury-volume, sham acupoint treatment increased the injury-volume and blank control had no obvious effect in reducing the injury-volume. Statistical tests also confirmed that acupuncture was more effective than random stimulus in improving the metabolic recovery after stroke.

Abstract:While acupuncture has survived several thousand years’ evolution of medical practice, its function still remains as a myth from the view point of modern medicine. Our goal in this paper is to quantitatively understand the function of acupuncture in ischemia stroke treatment. We carried out a comparative study using the Sprague Dawley rat animal model. We induced the focal cerebral ischemia in the rats using the middle cerebral artery occlusion (MCAO) procedure. For each rat from the real acupuncture group (n = 40), sham acupoint treatment group (n = 54), and blank control group (n = 16), we acquired 3-D FDG-microPET images at baseline, after MCAO, and after treatment (i.e., real acupuncture, sham acupoint treatment, or resting according to the group assignment), respectively. After verifying that the injured area is in the right hemisphere of the cerebral cortex in the brain by using magnetic resonance imaging(MRI) and triphenyl tetrazolium cchloride (TTC)-staining, we directly compared the glucose metabolism in the right hemisphere of each rat. We carried out t-test and permutation test on the image data. Both tests demonstrated that acupuncture had a more positive effect than non-acupoint stimulus and blank control (P < 0.025) in increasing the glucose metabolic level in the stroke-injured area in the brain, while there was no statistically significant difference between non-acupoint stimulus and blank control (P>0.15). The immediate positive effect of acupuncture over sham acupoint treatment and blank control is verified using our experiments. The long-term benefit of acupuncture needs to be further studied.
Force Decoding of Caudal Forelimb Area and Rostral Forelimb Area in Chronic Stroke Rats

Abstract:Brain machine interfaces (BMIs) used for movement restoration primarily rely on studies of motor decoding. It has been proved that local field potentials (LFPs) from primary motor cortex and premotor cortex of normal rodents could be used for decoding motor signals. However, few studies have explored the decoding performance of these brain areas under motor cortex damage. In this work, we focus on force decoding performance of LFPs spectrum from both ipsilesional caudal forelimb area (CFA) and rostral forelimb area (RFA) of rodents with ischemia over CFA. After three months of ischemia induced by photothrombosis over CFA, the power of high-frequency bands (>120 Hz) from both CFA and RFA can decode force signals by Kalman filters. The fair performance of CFA indicates motor reorganization over penumbra. Further exploration of RFA decoding ability proves that at least four electrodes of RFA should be used on decoding and electrodes far from CFA of stroke rats could achieve almost as good results as those close to CFA of normal rats, which indicates the motor remapping. Experimental results show the long-term stability of PM LFPs decoding performance of stroke rats as the trained Kalman model could be used to accurately decode force some days later which provides a possibility for online decoding system. In conclusion, our work shows that even under CFA ischemia, high-frequency power of LFPs from RFA is still able to accurately decode force signals and has long stability, which provides the possibility of BMIs for motor function reconstruction of chronic stroke patients.
Establishment of a pMCAO model in SD rats and screening for behavioral indicators suitable for long-term monitoring

Abstract:Objective: This study aimed to establish a permanent middle cerebral artery occlusion (pMCAO) model in rats to simulate the pathological process of stroke patients with no reperfusion. And screen highly sensitive items that could be used to detect long-term behavioral abilities in rat of intraluminal suture models. Method: Established the pMCAO model then tested the rats for the bilateral asymmetry, modified neurological severity score, grid-walking, cylinder, rotating, and water maze test from week 1 to week 16. Results: The infarct volume of the model rats was stable (26.72% ±1.86%). The sensorimotor test of bilateral asymmetry, grid-walking, cylinder, and mNSS test showed significant differences from week 1 to week 16 after injury. The water maze test at week 16 showed significant differences in spatial exploration and learning ability between the two groups. We confirmed that there was no significant difference between MRI and TTC staining in detecting the degree of brain injury, which facilitated the diversity of subsequent detection methods. We also confirmed that at multiple time points, grid, cylinder and water maze test were significantly positively correlated with rat brain infarct volume. Conclusion: They are suitable for the long-term observation of behaviors in the sequela stage of stroke in rat.

Quantitative Analysis of Focal Ischemic Cerebral Infarction in Mice

Abstract:AIM:To confirm the action of the light transmission method in evaluating focal ischemic cerebral infarction on persistent focal cerebral ischemia in mice. METHODS:Persistent focal cerebral ischemia was induced by middle cerebral artery occlusion (MCAO). Bederson's neurological scores, climbing board and hanging test were performed 24 h after ischemia, and infarct volume, brain hemisphere area, neuron density of cortex and subcortex were measured with computer-assisted imaging. Pranlukast ( 0.1 mg·kg -1) or nimodipine ( 0.4 mg·kg -1) were injected ip once daily for 3 days and to 1 h before MCAO assess the neuroprotective effect. RESULTS:The infarct volumes measured by light transmission closely correlated with that measured by TTC staining and neuron densities. The infarct volumes measured by light transmission well correlated with the neurological scores measured by integrated graded approach, too. Both pranlukast and nimodipine significantly attenuated infarct volumes and the ratio of ischemic/non-ischemic hemispheres, and reduced neurological deficits and neuron death. CONCLUSION:Light transmission and integrated graded approach can be used not only for qualitative analysis of focal cerebral ischemia, but also for evaluating the neuroprotective effect of drugs.
Similar cognitive deficits in mice and humans in the chronic phase post-stroke identified using the touchscreen-based paired-associate learning task

Abstract:Abstract For many chronic stroke survivors, persisting cognitive dysfunction leads to significantly reduced quality of life. Translation of promising therapeutic strategies aimed at improving cognitive function is hampered by existing, disparate cognitive assessments in animals and humans. In this study, we assessed post-stroke cognitive function using a comparable touchscreen-based paired-associate learning task in a cross-sectional population of chronic stroke survivors (≥ 5 months post-stroke, n = 70), age-matched controls (n = 70), and in mice generated from a C57BL/6 mouse photothrombotic stroke model (at six months post-stroke). Cognitive performance of stroke survivors was analysed using linear regression adjusting for age, gender, diabetes, systolic blood pressure and waist circumference. Stroke survivors made significantly fewer correct choices across all tasks compared with controls. Similar cognitive impairment was observed in the mice post-stroke with fewer correct choices compared to shams. These results highlight the feasibility and potential value of analogous modelling of clinically meaningful cognitive impairments in chronic stroke survivors and in mice in chronic phase after stroke. Implementation of validated, parallel cross-species test platforms for cognitive assessment offer the potential of delivering a more useful framework for evaluating therapies aimed at improving long-term cognitive function post-stroke.

Trap channal test:a novel method to evaluate the neurological functions of cerebraljschemic rats

Abstract:Objective To evaluate a novel,convenient and sensitive gauging system suitable to cerebral ischemia animal model--trap channel test.Method The cerebral ischemia models were made by ligating the middle cerebral artery(MCA)of Sprague-Dawley rats.The ischemic injury was confirmed by TTC staining.The neurological functions of the ischemic rats were evaluated with 5-score system,trap channel test and somatosensory evoked potential(SEP)and motor evoked potential(MEP)on the 2nd day,1st week,2nd week and 4th week after surgery.Results TTC staining demonstrated an infarctive area in the cortex of MCA distribution.5-score system could confirm the cortex injury,but could not be used to distinguish the subtle recoveries of the neurological functions.In the trap channel test,the Sum of bilateral extremities scores showed a peak on the 2nd day and 1st week after surgery,and decreased after the 1st week;the difference of the scores between injured and intact extremities showed a peak on the 1st week after surgery,and decreased after the 1st week.All components of SEP disappeared within the first week after the surgery,while manifested a recovery trend after 2w post-surgery,however,the MEP showed a paradox,i.e.the amplitude of MEP of the injured lateral was higher than the counter-part of intact lateral,and there was no significant differentiation between bilateral MEP latency.Conclusions Trap channel test is a convenient and sensitive gauging system to assess the neurological functions of the ischemic rat models,and can be beneficial for the experimental studies of cerebral ischemia.

Abstract:Abstract Ischemic strokes cause devastating brain damage and functional deficits with few treatments available. Previous studies have shown that the ischemia-hypoxia rapidly induces clinically similar thrombosis and neuronal loss, but any resulting behavioral changes are largely unknown. The goal of this study was to evaluate motor and cognitive deficits in adult HI mice. Following a previously established procedure, HI mouse models were induced by first ligating the right common carotid artery and followed by hypoxia. Histological data showed significant long-term neuronal losses and reactive glial cells in the ipsilateral striatum and hippocampus of the HI mice. Whereas the open field test and the rotarod test could not reliably distinguish between the sham and HI mice, in the tapered beam and wire-hanging tests, the HI mice showed short-term and long-term deficits, as evidenced by the increased number of foot faults and decreased hanging time respectively. In cognitive tests, the HI mice swam longer distances and needed more time to find the platform in the Morris water maze test and showed shorter freezing time in fear contextual tests after fear training. In conclusion, this study demonstrates that adult HI mice have motor and cognitive deficits and could be useful models for preclinical stroke research.

Refined movement analysis in the staircase test reveals differential motor deficits in mouse models of stroke

Abstract:Accurate assessment of post-stroke deficits is crucial in translational research. Recent advances in machine learning offer precise quantification of rodent motor behavior post-stroke, yet detecting lesion-specific upper extremity deficits remains unclear. Employing proximal middle cerebral artery occlusion (MCAO) and cortical photothrombosis (PT) in mice, we assessed post-stroke impairments via the Staircase test. Lesion locations were identified using 7 T-MRI. Machine learning was applied to reconstruct forepaw kinematic trajectories and feature analysis was achieved with MouseReach, a new data-processing toolbox. Lesion reconstructions pinpointed ischemic centers in the striatum (MCAO) and sensorimotor cortex (PT). Pellet retrieval alterations were observed, but were unrelated to overall stroke volume. Instead, forepaw slips and relative reaching success correlated with increasing cortical lesion size in both models. Striatal lesion size after MCAO was associated with prolonged reach durations that occurred with delayed symptom onset. Further analysis on the impact of selective serotonin reuptake inhibitors in the PT model revealed no clear treatment effects but replicated strong effect sizes of slips for post-stroke deficit detection. In summary, refined movement analysis unveiled specific deficits in two widely-used mouse stroke models, emphasizing the value of deep behavioral profiling in preclinical stroke research to enhance model validity for clinical translation.

Abstract:To evaluate the application of locomotor activity test in functional injury after global cerebral ischemia (GCI) in C57BL/6 mice.GCI was induced by bilateral carotid arteries occlusion for 30 min in C57BL/6 mice. Mice were divided into sham group, GCI group and minocycline group. Saline or minocycline (45 mg/kg) was i.p. injected once daily for 6 d after ischemia. At Day 6 after ischemia, locomotor activity was recorded for 1 h in open field test. Total distance, central distance, central distance ratio, periphery distance, periphery distance ratio, central time and periphery time were used to evaluate the behavior characteristics of locomotor activity in C57BL/6 mice after ischemia. The survival neuron density was detected by Nissl staining in hippocampus, cortex and striatum.Compared with sham group, total distance, central distance and central time increased and periphery time decreased in C57BL/6 mice after GCI (Ps<0.05). However, minocycline significantly reduced the central distance and central time and increased the periphery time (Ps<0.05). Neurons were damaged in hippocampus, cortex and striatum after GCI, which manifested by decreased neurons and the most serious damage in hippocampal CA1 region. Minocycline significantly improved the neuron appearance and increased the neuron number in hippocampus and striatum (P<0.001 or P<0.05).Locomotor activity in open field test can objectively evaluate the behavior injury after GCI in mice. Central distance and central time can be used as indexes of quantitative assessment.
Light transmission measurement of focal ischemic cerebral infarction in mice

Abstract:OBJECTIVE: To evaluate the feasibility of light transmission to measure focal cerebral ischemia in mice. METHODS: Persistent focal cerebral ischemia was induced by middle cerebral artey occlusion (MCAO) in mice. The brain were removed 24 h after MCAO and coronally dissected into 1 mm sections. Using a stereomicroscope, the brain section was illuminated with a halogen lamp and computerized images were stored. Next the brain sections were stained for 30 minutes with 0.5% TTC (2, 3, 5-triphenylterzolim chloride) at 37 degrees C. Using an image analyzer (AnalyPower 1.0), the infarct volumes obtained by light transmittance and TTC staining were calculated. Integrated gray scales of sections of both hemispheres were calculated by Photoshop 5.0. RESULTS: A close correlation existed between cerebral infarct volume measured by light transmission and TTC staining (r=0.81). The mean gray scales measured by both techniques of the ischemic hemispheres as well as those of the cortex, subcortex and hippocampus were siginificantly higher than those of non-ischemic hemispheres and of control mouse hemispheres (P <0.001). Further there were no significant difference between the two hemispheres of control mice and between hemispheres of control mice and non-ischemic hemispheres of the MCAO mice. CONCLUSION: Light transmission can be used for qualitative analysis of focal cerebral ischemia.

Abstract:In the treatment of ischemic cerebral stroke (ICS), most conventional treatments, including carotid endarterectomy and carotid artery stenting, may cause cerebral ischemia-reperfusion injury (CIRI). For treated ICS patients, changes in cerebral blood flow are directly related to brain function. At present, computed tomography perfusion, dynamic susceptibility contrast-enhanced perfusion weighted imaging and magnetic resonance arterial spin labeling perfusion imaging are used to monitor cerebral blood flow, but they still have some limitations. Our study aimed to monitor the changes in cerebral cortical blood flow by laser speckle contrast imaging (LSCI) in CIRI model mice and to propose a new method for predicting outcomes after CIRI. C57BL/6 N mice were used to establish a mouse CIRI model based on a modified thread-occlusion method and divided into a good outcome group and a poor outcome group according to survival within 7 days. The cerebral cortical blood flow of the area supplied by the left middle cerebral artery was monitored by LSCI at baseline (before modeling), 1 h after ischemia, immediately after reperfusion and 24 h after reperfusion. Then, the brains of the mice were removed immediately and stained with hematoxylin and eosin to observe the pathological changes in brain neurons.

An Improved Quantitative Method for Evaluation of Ischemic Injury and Neuroprotection in Mouse Brain Slices

Abstract:OBJECTIVE:To establish a simpler and more accurate method for evaluating in vitro ischemic injury and neuroprotective effects of drugs through improving experimental instrument and quantitative index in mouse brain slices.METHODS:An incubation instrument was developed and its application tested. 2,3,5-triphenyltetrazolium chloride (TTC) was used as a substrate to biosynthesize formazan standard in mouse brain slices, and formazan was isolated, purified and identified. Ischemic injury of mouse brain slices was induced by oxygen/glucose deprivation (OGD), the produced formazan from TTC in the cortex and striatum was measured at 490 nm spectrophotometrically. Edaravone and ONO-1078 were added into the incubation medium to observe their neuroprotective effects.RESULT:The incubation instrument worked well for incubating brain slices and obtaining stable results efficiently. Standard formazan was biosynthesized and purified with a purity of 99.3%, and showed a linear range of 0.05 - 1 mg/ml in absorbance at 490 nm (r=0.9997). OGD decreased formazan production in the cortex and striatum in a duration-dependent manner. Edaravone (0.01 to 1 micromol/L) recovered OGD-induced decrease of formazan production, but ONO-1078 showed no effect.CONCLUSION:The incubation instrument and quantitative measurement of formazan developed in this study are efficient,accurate and simple for evaluating ischemic injury and neuroprotection,which can be used in screening of neuroprotective drugs in vitro.
