Smoking before Surgery Deeply Affects the Native MMPs/TIMPs Gene Expression in Saphenous Vein of Patients undergoing CABG

Abstract:Objective Smoking is a major risk factor for cardiovascular disease.Native matrix metallopeptidase and inhibitor(MMPs/TIMPs) milieu may significantly impact the tendency of vessel pathologic remodeling.However,studies on MMPs/TIMPs in saphenous vein(SV) of smokers are limited.We investigated the effects smoking status on native MMPs/TIMPs expression in SV.Methods One hundred and ten patients undergoing CABG were divided into 3 groups:non-,heavy smokers,and long-term quitters group.MMP-2,MMP-9,TIMP-2 and TIMP-3 mRNA levels were analyzed by real-time RT-PCR.Protein levels were evaluated by immunocytochemistry and western-blot.Results Compared with the non-smokers,MMP-2 and MMP-9 expression were significantly increased than the other two groups,respectively,P < 0.05.In contrast to MMPs response,TIMP-2 and TIMP-3 expression was significantly decreased,respectively,P < 0.05.Conclusion This study suggests that smoking status has deep effects on native MMPs/TIMPs gene expressions in SV before CABG.The imbalanced MMPs/TIMPs gene expression has been considered to disturb the constitution of excellular matrix in vein graft.The dysregulated MMPs/TIMPs gene expression may recover after smoking cessation.Even so,the recovery is not desirable.
