Activation of Akt/c-Myc Signaling Pathway Suppressing the Apoptosis of Gastric Cancer Cells

Abstract:Background and purpose:Activation of Akt signaling pathway may be able to decrease the chemotherapy sensitivity of gastric cancer cells,but the mechanism was still obscure.Our aim was to elucidate the underlying mechanism,the correlations between Akt/c-Myc signaling pathway and apoptosis in human gastric cancer cell line SGC7901 and BGC823.Methods:Both SGC7901 and BGC823 were exposed to etoposide with or without Wortmannin(a special inhibitor of PI3’K/Akt pathway) .Cell cycle distribution and cell apoptosis analysis were performed by flow cytometry and Hoechst 33258 staining,respectively.Cytotoxicity was assessed by determining cell survival with 3-(4,5-dimethyl thiazol-2-yl) -2,5-diphenyl tetrazolium bromide(MTT) .Akt activities were detected using nonradioactive immunoprecipitation(IP) -kinase assay.Western blotting was exploited to evaluate the phosphorylation levels of AktSer473 and the protein expressions of c-Myc.The mRNA expressions of c-Myc were detected by reverse transcription polymerase chain reaction(RT-PCR) .Results:Etoposide caused a time and dose-dependent reduction of cell viability of SGC7901 and BGC823.Wortmannin signifi cantly enhanced the cell inhibitory effi ciency of etoposide.The cancer cells treated with both etoposide and Wortmannin showed signifi cantly higher level of apoptotic phenotype than those treated with etoposide only.Phosphorylation levels of Akt,Akt activities and expressions of c-Myc were signifi cantly induced by etoposide in a time-dependent manner.Wortmannin suppressed the up-regulation of c-Myc protein expressions induced by etoposide but had no affect on the expressions of c-Myc mRNA.Conclusion:Chemotherapy agent activated Akt/c-Myc signaling pathway,which decreased the chemotherapy sensitivity of gastric cancer cells SGC7901 and BGC823 via up-regulating the protein expression of c-Myc.This might be one of the mechanisms of gastric cancer chemoresistance.
