The Relationship Between Expression of Interferon Alpha and Interleukin 18 in Peripheral Blood Mononuclear Cells and Interferon Response in Patients with Chronic Hepatitis C

Abstract:Objective To detect the expression of interferon alpha (IFN-α) and interleukin 18 (IL-18) in peripheral blood mononuclear cells (PBMCs) of patients with chronic hepatitis C virus (HCV) infection, and analyze the cellular immune status of patients with chronic hepatitis C(CHC). Methods PBMCs of CHC patients were cultured, and then detected their antiviral biological activity stimulated with high concentration PolyIC by viral protection assay. IL-18 mRNA expression from PBMCs stimulated with LPS was measured by reverse transcription-polymerase chain reaction (RT-PCR). Results The data indicated interferon level was kept lower from PolyIC induced PBMCs of CHC than from normal controls. Responder (n=12) enhanced activity of IFN-α from PBMCs stimulated with PolyIC after IFN-α2b therapy. However, non-responder (n=26) continuously kept IFN-α at low level from PBMCs after IFN-α2b therapy. As for the expression of IL-18 mRNA in PBMCs stimulated with LPS, the expression of responder and non-responder were remarkably lower than that of normal control group (n=20) before IFN-α2b therapy (P<0.01) However, after IFN-α2b therapy, the level of IL-18 mRNA of responder was sharply increased and that of non-responder was kept low . There was a positive correlation between level of IL-18 mRNA and IFN activity (r=0.873, P<0.01). Conclusions The cellular immune function of patients with CHC is impaired. The expression of IFN-α gene of non-responder of CHC patients was inhibited. The IFN-α activity and level of IL-18 mRNA from PBMC5 could be indicators for hepatitis C prognosis after IFN-α therapy for CHC patients.
