Establishment of a Piglet Model of Chronic Pancreatitis

Abstract:Objective To establish an animal model of piglet with chronic pancreatitis.Methods Twenty-eight piglets were included in this study.Six healthy piglets served as control.An upper midline laparotomy was performed to partially ligate the pancreatic duct(PD)to produce chronic pancreatitis in twenty-two piglets(CP group).Histopathological examination of the pancreas was performed in the control and the 22 experimental piglets after two to twenty weeks of PD ligation,respectively.CP was staged according to the severity of fibrosis in the pancreas.Results Four piglets were sacrificed after one to two weeks of duct ligation in the CP group.In the left 18 piglets,3 piglets had normal pancreas and 15 piglets had chronic obstructive pancreatitis,including 7 piglets with grade 1,4 piglets with grade 2,and 4 piglets with chronic pancreatitis grade 3.The successful rate of CP induction was 68.2%(15/22)in the piglets.The severity of CP was partially correlated to the duration after pancreatic duct ligattion(r=0.39,P0.05).Conclusion Chronic obstructive pancreatitis can be produced by partial ligation of the pancreatic duct in piglets,and its severity is partially correlated with the duration of pancreatic duct ligation.
