Abstract:Objective: To examine the association between polymorphisms of cytochrome P450 Ⅱ E1(CYP2E1) gene and Paraoxonase 2 (PON2148) gene on the low birthweight.Methods:Using standard questionnaire, the 231 singleton live born infant-mother pairs (cases:65 pairs; controls:166 pairs)from 1998-1999 at Anqing City in Anhui Province were investigated by trained field workers with epidemiologic case-control method.Results:Both CYP2E1 and PON2148 polymorphisms were not significantly associated with low birthweight, respectively, after adjustment for major confounders including maternal age, education, parity, passive smoking, infant gestational age and sex. They were significant respectively( CYP2E1, OR=2.4, 95%CI=1.17-4.89; PON2148, OR=2.16, 95%CI=1.06-4.41). When CYP2E1 and PON2148 polymorphisms were considered jointly, with CYP2E1 homozygous wild-type and PON2148 homozygous wild-type as reference, the low birthweight was significantly associated with CYP2E1 variant/mutation and PON2148 variant/mutation among women(OR=4.77, 95%CI=1.63-13.95), But we did not observe the gene interactions in relation to low birthweight. Conclusion: Our data suggest a genetic influence on low birthweight in this population, It is suggestied that there is no synergism of gene CYP2E1 and gene PON2148.
[Analysis of Association Between Gene Polymorphisms of Microsomal Epoxide Hydrolase (EPHX1) and Infant Birthweight].

Abstract:We investigated association between genetic polymorphisms of EPHX1 in mother and infant birthweight. Data of 342 female workers were collected in textile mill. A total of 342 mothers were genotyped for the His139Arg polymorphism of EPHX1 by a polymerase chain reaction-restriction fragment length polymorphism assay. Using multiple linear regression models, we estimated the adjusted association between polymorphisms of EPHX1 and birthweight, with adjusted for potential confounders. We found that polymorphisms of EPHX1 were closely associated with reduced birthweight (beta +/- SE = -149 g +/- 56, P = 0.0083) after adjustment for major cofounders. In subgroup of passive smoking analysis, the polymorphisms of EPHX1 were highly associted with birthweight for those with passive smoking (beta +/- SE = -234 g +/- 88, P = 0.0088); The significant association of EPHX1 polymorphisms with reduced birthweight were showed for those with working stress (beta +/- SE = -157 g +/- 59, P = 0.0079). Our data provide polymorphisms of EPHX1 in mothers were significant association with birthweight of their infant, and showed gene-environmental interaction in relation to birthweight.

Abstract:GSTT1 (absent) was significantly associated with reduced birth length, but MSP1, GSTM1 polymorphisms and organic solvents exposure were not significantly associated with reduced birth length, respectively. After adjustment for major confounders including maternal age, education, parity, occupation, passive smoking, pre-pregnant BMI, infatn gestational age and sex, GSTT1, GSTM1 and organic solvents exposure were significantly associated with reduced birth length, respectively. Moreover, in comparison of birth lengths in eight subgroups defined by organic solvent status (nonexposure/exposure) and maternal genotype for MSP1 (AA, Aa/aa) and GSTM1 (present, absent), in the presence of organic solvents^ exposure, the birth length was a clear gradient of adverse effects. However, in the absence of organic solvents^ exposure, the birth lengths did not differ significantly by maternal genotype. The similar results were observed in the group of organic solvent status (no, yes), MSP1 (AA, Aa/aa) and GSTT1 (present, absent).
Polymorphism in Maternal HLA-DRB5 is Associated with the Risk of Preeclampsia in Chinese Population

Abstract:AbstractBackground:Preeclampsia is an important clinical syndrome occurring during pregnancy. It shows genetic disposition, and the genetic risk has large ethnic heterogeneity. The study was designed to investigate the genetic risk of preeclampsia in Chinese pregnancies, and to apply it in early screening of the disease.Methods:We performed a genome-wide association study to screen candidate risk loci associated with preeclampsia in Chinese people, and validated them with an independent cohort of enlarged size. We also trained prediction models using the genotypes of newly identified risk loci to screen the pregnancies with high preeclampsia risk.Results:A segment in chromosome 6 coveringHLA-DQB1,HLA-DRB5and other immune-related genes shows the most significant association, and three loci inHLA-DRB5were confirmed with an enlarged validation cohort. One of the validated loci, rs147440497, forms an amino acid change by the nucleotide polymorphism, which further causes a conformational change in the antigen-binding domain of HLA-DRB5 protein. With the genotypes of risk genetic loci and other demographic features, a machine-learning model was trained, which can predict Chinese preeclampsia pregnancies accurately, with a cross-validated recalling rate of 0.63 at a false positive rate of 8%.Conclusion:We identified a novel gene from maternal genome,HLA-DRB5, the polymorphism in which is associated with preeclampsia. The genotypes of risk SNP loci can also be used for prediction of preeclampsia risk in Chinese population accurately.

Abstract:Objectives: The aim of this study was to explore the association of vitamin D (VitD) levels during pregnancy and its metabolic pathway genes with the risk for preterm birth (PTB) among pregnant women in southeast China. Methods: This study was conducted in Zhoushan Maternal and Child Health Hospital, Zhejiang, from August 2011 to May 2018. Plasma 25-hydroxyvitamin vitamin D [25(OH)D] levels in three trimesters and single-nucleotide morphisms in the VitD metabolic pathway were measured. Relevant information was collected using questionnaires and an electronic medical recorder system. Multiple statistical methods including linear regression, logistic regression, and crossover analysis were applied. Results: The prospective cohort study included 3465 pregnant women, of which 202 were PTB (week of gestation at delivery: 33.38 +/- 4.05), accounting for 5.8%. After adjusting for potential confounders, VitD sufficiency (>= 30 ng/ mL) in the second and third trimesters was associated with longer gestational age at delivery compared with VitD deficiency (<20 ng/mL). However, no significant association was found between VitD with the risk for PTB. rs7041, rs10210408, and rs2228171 were associated with gestational week and the risk for PTB. Significant associations were found of rs10210408, rs2209314, rs1155563, rs2544381 and the status of VitD in the second and third trimester with the gestational week. We also found that rs7041 and VitD in the second trimester might exert interaction on gestational week and the risk for PTB (P-inter = 0.038; P-inter = 0.019); rs16846876 and VitD in the second trimester might exert interaction on gestational week (P-inter = 0.024); rs4334089 and VitD in the third trimester might exert interaction on gestational week (P-inter = 0.024). Similar results were found when we tested pregnant women's plasma 25(OH)D in the first and second trimesters. Conclusions: Women with VitD deficiency were associated with shorter gestational weeks. Single-nucleotide morphisms in VitD metabolic pathway genes were significantly associated with gestation week and the risk for PTB, mainly in vitamin D -binding protein (GC) and low-density lipoprotein-related protein 2 (LRP2) genes. Additionally, maternal VitD with GC gene and maternal VitD with vitamin D receptor (VDR) gene might exert interactions on the risk for PTB. (C) 2021 Elsevier Inc. All rights reserved.

Abstract:Objective: To examine the interaction effects of prenatal exposed to environmental tobacco smoke (ETS) and genotypes of cytochrome P4501A1 (CYP1A1), glutathione S-transferases (GSTs) on the risk of full-term low birth weight (FT-LBW). Study design: We conducted a case-control study among pregnant women at two Women and Children's Hospitals in Guangdong, China (n = 910). Information was collected through interview, medical records review, and blood lab tests. Maternal selfreport and serum cotinine concentration were combined to define prenatal exposed to ETS. Logistic regression approach was applied for statistical analysis. Results: Our results showed that regardless of genotypes, prenatal exposed to ETS significantly increased the risk of FT-LBW. Then, two-way interactions showed increased prevalence of FT-LBW in prenatal exposed to ETS mothers with the CYP1A1 variant genotype (MspI "CC"), or with GSTT1-null genotype. Furthermore, three-way interactions showed that women with CYP1A1 variant (MspI "TC" or BsrDI "AG") genotypes and GSTT1 "null" genotype had higher risk to give birth of FT-LBW. Additionally, among nonexposed ETS mothers, genotype did not independently confer adverse effects on FT-LBW. Conclusions: Our results revealed that prenatal exposed to ETS is independently associated with FT-LBW while gene polymorphisms of CYP1A1 and GSTs merely play modified roles in this process. This study extends understanding of three-way interaction, and stresses the need to tobacco control toward pregnant women for better pregnant outcomes.

Abstract:This study investigated the association between polymorphisms in cytochrome P450 genes (Cyp1A1MspI, CYP1A1HincII, and CYP2E1) and low birth weight. Between July 1999 and June 2002, we conducted a study using infant-parents triads in Anqing, China. The analyses included the families of 248 normal birth weight, full-term infants, and 248 low-birth-weight infants. Genotyping was performed for the polymorphisms of cytochrome P450 genes using standard techniques. We used log-linear modeling to analyze the association of CYP1A1 and CYP2E1 gene polymorphisms with the risk of low birth weight. In the analysis of children's genotypes, the relative risk was 1.92 (95 % confidence interval: 1.05, 3.52, p = 0.034) for CYP1A1Mspl C/C6235 compared with CYP1A1 MspI T/T6235. In the analysis of mothers' genotypes, an association was also seen for maternal CYP1A1MspI C/C6235 compared with CYP1A1MspI T/T6235 (relative risk: 1.68, 95 % confidence interval: 1.06, 2.68, p = 0.029). We did not observe a joint effect between mother's and children's genotypes. Analysis of control triads suggests Mendelian transmissions of the variant alleles of CYP1A1MspI, CYP1A1HincII, and CYP2E1. In conclusion, both infant and maternal CYP1A1MspI C/C6235 genotypes were associated with increased risk of low birth weight in our study population. Abstract:Objective: To investigate the influences of aryl hydrocarbon (AHR), cytochrome P450 (CYP1A1), epoxide hydrolase 1 (EPHX1), and glutathione S-transferase P1 (GSTP1) genetic polymorphisms on small-for-gestational-age (SGA) infants. Methods: This nested case-control study (126 cases and 381 controls) was based on a prospective cohort study in Shanxi Province, China. We collected the general information of subjects using questionnaire and identified their single nucleotide polymorphisms by the MassARRAY genotyping platform. Results: The polymorphisms of CYP1A1 (rs4646421 and rs4646903) and EPHX1 (rs1051740) were significantly associated with SGA. Neonates of women with EPHX1 (rs1051740) and GSTP1 (rs1695) variant alleles were at a significantly increased risk of SGA compared with the reference group (OR ?=? 5.26; 95% CI, 1.08?25.66), as were neonates of women with CYP1A1 (rs4646903) and EPHX1 (rs1051740) variant alleles (OR ?=? 7.11; 95% CI, 1.55?32.62). The results of strata analysis by AHR (rs2282883 and rs17137566) showed that the associations between the polymorphisms of CYP1A1 (rs4646421 and rs4646903) EPHX1 (rs1051740), GSTP1 (rs1695) and SGA were of significance in women with variant heterozygous or homozygous genotype. Conclusions: CYP1A1 (rs4646421 and rs4646903), EPHX1 (rs1051740), and GSTP1 (rs1695) genetic variances might increase the risk of SGA. AHR (rs2282883 and rs17137566) resulted in estimated effects varying across strata on CYP1A1 (rs4646421 and rs4646903), EPHX1 (rs1051740), and GSTP1 (rs1695).

Abstract:BACKGROUND:Prenatal disinfection by-products (DBPs) exposure is linked with adverse birth outcomes. Genetic susceptibility to DBP metabolism may modify the exposure-outcome associations.OBJECT:To investigate whether CYP2E1 and GSTZ1 genetic polymorphisms modified the associations of prenatal DBP exposures with adverse birth outcomes.METHODS:Two biomarkers of DBP exposures including trihalomethanes (THMs) in blood and trichloroacetic acid (TCAA) in urine were determined among 426 pregnant women from a Chinese cohort study. CYP2E1 (rs2031920, rs3813867, and rs915906) and GSTZ1 (rs7975) polymorphisms in cord blood were genotyped. Statistical interactions between prenatal DBP exposures and newborns CYP2E1 and GSTZ1 polymorphisms on birth outcomes (birth weight, birth length, and gestational age) were examined by multivariable linear regression with adjustment for potential confounders.RESULTS:We found that newborns CYP2E1 genetic polymorphisms (rs2031920 and rs3813867) modified the associations of maternal blood THMs or urinary TCAA levels with birth outcomes. However, these interactions were nonsignificant after Bonferroni correction for multiple comparisons, except for the interaction between maternal blood BrTHMs [sum of dibromochloromethane (DBCM), bromodichloromethane (BDCM), and bromoform (TBM)] and newborns CYP2E1 gene rs2031920 polymorphisms on birth weight (P for interaction = 0.003).CONCLUSION:Newborns genetic variations of CYP2E1 rs2031920 may modify the impacts of prenatal BrTHM exposure on birth weight. This finding needs to be further confirmed.
Low Organic Solvent Exposure and Combined Maternal-Infant Gene Polymorphisms Affect Gestational Age.

Abstract:Objective: Little information is available on the associations of combined maternal-infant genetic susceptibility, environmental exposures and reproductive outcomes, especially in the Chinese population. The aim of this study was to investigate whether the polymorphisms of combined maternal-infant metabolic genes, CYP1A1 HincII, CYP1A1 MspI, GSTT1 and GSTM1 affect the association of maternal exposure to organic solvents with gestational age.Methods: 1113 mother-infant pairs were enrolled from the Beijing Yanshan Petrochemical Corporation between June 1997 and June 2002, of which 546 mothers were exposed to organic solvents and 567 were not. Multiple linear regression models were used to estimate the combined maternal-infant gene effects, and to characterise combined maternal-infant genetic susceptibility to organic solvents in relation to gestation.Results: Organic solvent exposure was significantly related to shortened gestation (-1.2 weeks, 95% CI-1.6 to -0.9). Additionally, combined maternal-infant genotypes including Ile/Ile462-Ile/Ile462 (-0.6 weeks, 95% CI-0.9 to -0.4) in CYP1A1 HincII and absent absent in GSTT1 (-0.4 weeks, 95% CI-0.9 to -0.3) were significantly associated with shorter gestation. When considering both organic solvent exposure and combined maternal-infant genotypes, the largest associations were found among exposed women with the absent-absent genotype (-1.5 weeks, 95% CI-1.8 to -1.2) in GSTT1 and the Ile/Ile462-Ile/Ile462 genotype (-1.5 weeks, 95% CI-1.8 to -1.2) in CYP1A1 HincII, suggesting that combined genotypes would modify the effect of exposure to organic solvents on gestation.Conclusions: This study demonstrates the role of combined maternal-infant genotypes in modifying the adverse effects of organic solvent exposure on gestation, and the maternal-infant interaction of four genes was determined.
Association Between Small Heterodimer Partner Gene and High Birth Weight Infants

Abstract:Objective To evaluate the relationship of small heterodimer partner(SHP) gene and birth weight in China. Methods A cohort study of 191 normal pregnant women was conducted. Both maternal and cord blood samples were collected. PCR-RFLP was used to detect the polymorphism of SNP-rs7504 of SHP. Results (1) The frequency of both neonatal and maternal C allele and (TC+CC) genotype increased significantly with birth weight (P=0.004, OR=3.168; P=0.005, OR=3.315; P=0.013, OR=2.495; P=0.013, OR=2.495). (2) The babies were heavier if they were C allele carrier. The average increase of birth weight was 246.3 g comparing the neonates with TC+CC genotype with those with TT genotype [(3658.7±400.94)g vs (3412.4±444.4)g, P=0.005]. The average birth weight of those maternal C allele carriers was 210.3 g heavier than those non-C allele carriers[(3628.9±405.5) g vs (3418.6±449.0 g]. (3) The fetal C allele was associated with maternal weight in pregnancy, prepregnant BMI, paternal height and weight. Women with C allele were heavier and had higher BMI without statistical significance comparing with those non-C allele carriers. Neither neonatal nor maternal SHP gene was associated with blood glucose and insulin level. (4) Multiple factors analysis showed that birth weight was related to maternal height, weight gain during pregnancy, prepregnant BMI, maternal and cord blood insulin level. After adjustment, the neonatal birth weight remained significantly correlated with cord blood SHP (P=0.0354), but not with maternal SHP gene (P=0.0711). Conclusions SHP gene is associated with newborns birth weight and may affect fetal growth.
A Genome-wide Association Analysis in Four Populations Reveals Strong Genetic Heterogeneity For Birth Weight.

Abstract:Low or high birth weight is one of the main causes for neonatal morbidity and mortality. They are also associated with adulthood chronic illness. Birth weight is a complex trait which is affected by baby's genes, maternal environments as well as the complex interactions between them. To understand the genetic basis of birth weight, we reanalyzed a genome-wide association study data set which consists of four populations, namely Thai, Afro-Caribbean, European, and Hispanic population with regular linear models. In addition to fit the data with parametric linear models, we fitted the data with a nonparametric varying-coefficient model to identify variants that are nonlinearly modulated by mother's condition to affect birth weight. For this purpose, we used baby's cord glucose level as the mother's environmental variable. At the 10(-5) genome-wide threshold, we identified 33 SNP variants in the Thai population, 26 SNPs in the Afro-Caribbean population, 18 SNPs in the European population, and 7 SNPs in the Hispanic population. Some of the variants are significantly modulated by baby's cord glucose level either linearly or nonlinearly, implying potential interactions between baby's gene and mother's glucose level to affect baby's birth weight. There is no overlap between variants identified in the four populations, indicating strong genetic heterogeneity of birth weight between the four ethnic groups. The findings of this study provide insights into the genetic basis of birth weight and reveal its genetic heterogeneity.
Correlation between Pax-9 genetic polymorphism and low birth weight infants

Abstract:Objective:To explore the correlation between Pax-9 genetic polymorphism and low birth weight infants.Methods:60 low birth weight infants and 70 normal birth weight infants were selected,the three gene locus (rs2073244,rs2073247,rs4904210) of Pax-9 gene were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP),and the genotypes and distribution frequency of alleles were compared.Results:There was significant difference in the total distribution frequency of Pax-9 gene in rs2073247 situs and rs4904210 situs (χ2=14.53 and 16.66,P<0.05), but there was no difference in rs2073244 situs (χ2=5.74,P>0.05). The distribution frequencies of AG genotype in rs2073244 situs, TT genotype in rs2073247 situs and CC genotype in rs4904210 situs in low birth weight infants group were significantly higher those in normal birth weight infants group (P<0.05),while the distribution frequencies of AA genotype in rs2073244 situs, TC genotype in rs2073247 situs and CG genotype in rs4904210 situs reversed (P<0.05).Conclusion:The distribution frequencies of rs2073247 situs,rs4904210 situs and rs2073244 situs in Pax-9 gene affect the incidence of low birth weight infants.

Abstract:Objective To investigate whether genetic polymorphism in the intestinal fatty acid binding protein(IFABP) gene is associated with low birth mass in neonates.Methods Polymerase chain reaction-restriction fragment length polymorphism was used to detect the polymorphism in 86 low birth mass neonates and 96 normal ones.Results The A54T site frequencies of AA,TT,AT genotypes of IFABP gene were 24.4%,5.8%,69.8% in disease group while those were 20.8%,41.7%,37.5% in normal controls,the differences were significantly seen in the distribution frequency between two groups(P=0.000).Conclusion The combination between genetic polymorphisms in the IFABP gene in neonates is significantly associated with neonatal low birth mass;the frequency of encoding 54Ala/ Thr polymorphism in IFABP gene is related with low birth mass.

Abstract:Objective:To explore the relationship between polymorphism of quinone oxidoreductase NQO1 gene and low birth weight of neonates.Methods:Case-control study was used to collect the data of mothers and neonates,PCR-RFLP method was applied to analyze NQOI genotype of mothers,logistic regression analysis was used to analyze the relationship between NQOI(C609T) genotype of mothers and low birth weight of neonates.Results:There was no significant difference in frequency of NQOI C609T genotype between observed value and expected value in the two groups(χ2=159,P0.05;χ2=0.12,P0.05),the distribution of NQO1 genotype in the two groups corresponded with Hardy-Weinberg law of genetic equilibrium;after adjusting the main confounding factors,there was a significant correlation between genotype of NQOI(C609T) mutation and low birth weight(OR=1.922,95%CI 1.205~3.066,P=0.005;OR=4.533,95%CI 1.732~11.867,P=0.002).Conclusion:There is significant correlation between polymorphism of NQO1 gene and low birth weight.
Gene-environment Interaction among GSTT1, PON2 Polymorphisms and Organic Solvents on Gestational Age in a Chinese Women Cohort

Abstract:To investigate interactions on gestational age among two environmental risk factors and four maternal genetic polymorphisms: organic solvents, passive smoking, CYP1A1 rs4646903 (MspI), EPHX1 rs2234922 (His139Arg), GSTT1 and PON2 rs12026 (Ala148Gly).A pregnant women cohort was conducted at Beijing Yanshan Petrochemical Corporation, and 1,097 mothers with live singleton births were included in analysis. Generalized Multifactor Dimensionality Reduction (GMDR) method was used to explore interactions among these factors with adjustment for important potential confounders. Multiple linear regression models were used to estimate the association of interaction with gestational age.A three-factor model of organic solvents, GSTT1 and PON2 rs12026 had the highest testing balanced accuracy (57.05 %) and best cross-validation consistency (10/10). Compared with organic solvents unexposed mothers with GSTT1 non-null genotype and PON2 rs12026 CC genotype, organic solvents exposed mothers with GSTT1 null genotype and PON2 rs12026 CG + GG genotype had the largest reduction in gestational age (-0.36 weeks, 95%CI: -0.70 to -0.02). The significant reductions in different groups were from 0.24 weeks to 0.36 weeks.Maternal genetic susceptibility GSTT1 and PON2 rs12026 could significantly modify the association of organic solvents with gestational age.

Abstract:Genetic factors significantly influence the concentration of metabolites in adults. Nevertheless, the genetic influence on neonatal metabolites remains uncertain. To bridge this gap, we employed genotype imputation techniques on large-scale low-pass genome data obtained from non-invasive prenatal testing. Subsequently, we conducted association studies on a total of 75 metabolic components in neonates. The study identified 19 previously reported associations and 11 novel associations between single-nucleotide polymorphisms and metabolic components. These associations were initially found in the discovery cohort (8,744 participants) and subsequently confirmed in a replication cohort (19,041 participants). The average heritability of metabolic components was estimated to be 76.2%, with a range of 69%-78.8%. These findings offer valuable insights into the genetic architecture of neonatal metabolism.
Passive Smoking, Metabolic Gene Polymorphisms, and Infant Birth Weight in A Prospective Cohort Study of Chinese Women

Abstract:The authors investigated whether polymorphisms in two maternal metabolic genes, cytochrome P-450 1A1 (CYP1A1) MspI and epoxide hydrolase 1 (EPHX1) Tyr113His, affect the association of maternal passive smoking with infant birth weight. The study was conducted in a cohort of 1,388 newly married mothers of liveborn singletons who worked in textile mills in Anqing, China, from 1996 to 2000. Multiple linear regression models were used to estimate the associations of passive smoking and genetic susceptibility with birth weight, with adjustment for important potential confounders. In the passive smoking group, there was a remarkable decrease in birth weight with the C/C6235 genotype (156.3 g, 95% confidence interval (CI): -283.6, -29.0) for CYP1A1 MspI and with Tyr/His113 (93.8 g, 95% CI: -188.6, -1.1) as compared with His/His113 (244.6 g, 95% CI: -491.0, -1.9) for EPHX1. When results were stratified by maternal genotype, passive smoking conferred a significantly negative effect in the EPHX1 Tyr/His113 group (103.5 g, 95% CI: -205.8, -9.2) and in the His/His113 group (687.3 g, 95% CI: -748.3, -178.3). The data further showed that there was a significant interaction between maternal passive smoking and maternal EPHX1 genotype for birth weight. The authors conclude that the CYP1A1 MspI and EPHX1 genotypes modified the association between maternal passive smoking and infant birth weight in this study, which is suggestive of possible gene-environment interaction.
