Large-Conductance Calcium-Activated K+ Channels Are Involved in Edhf-Mediated Relaxations in the Rat Renal Artery.

Abstract:The aim of the present study was to investigate the long-term effect of menopause (bilateral ovariectomy) on endothelial function in isolated aortic rings of spontaneously hypertensive rats (SHR).Methods: Female SHR aged 13 weeks were randomly divided into two groups: ovariectomized females (OVX fSHR, n=10) and shamovariectomizcd females (SHAM t'SHR, n=10).After that, rats were manteined in standard conditions (constant tcmperature; 12-hour dark/light cycle; standard rat chow and water ad libitum) for 24 weeks.At 37 weeks of age, the vascular reactivity in aortic rings was studied.The systolic blood pressure (SBP) was estimated during wceks 12 and 37 by the tail-cuff method.Results: Bilatcral ovariectomy did not significantly increase SBP levels compared with their respective basal values and with those of shamoperated animals.In studies designed to assess the basal release of nitric oxide (NO), we found that vasoconstriction induced by L-NAME (10 "6-l0 "4 M) was greater in aortic rings from SHAM fSHR than in those obtained from OVX fSHR (p<0,05).Concentration-related relaxant responses to SOD (0.l-100 units/mL) were also significantly greater in sham-ovariectomized group as compared to ovariectomized rats (p<0,05).In contrast to the differences seen in basal NO release, muscarinic receptor-mediated release of NO was no altered by ovariectomy, as deduced from ACh concentration-responses curves (10-9-10 "5 M).Nevertheless, both groups show dccreased ACh-induced relaxations associated with hypcrtension as expected.Responses to the exogenous NO donor SNP were identical in OVX and SHAM fSHR, ruling out differences in smooth muscle reactivity to NO. Conclusion: In onr model, both stimulated NO release, associated with hypertension, and basal NO release, associated with menopause, are impaired.Accordingly, decreased NO release from the vasculature may account in large part for the increased cardiovascular risk of hypertensivc postmenopaasal women.
