Influence of valsartan on vascular remodeling in mice with vascular injury

Abstract:Objective To observe the influences of valsartan on intima and tunica media in mice with vascular injury.Methods Male C57BL/6 wild mice(8 weeks old,n=80) were randomly divided into operation group,treatment group,sham-operation group and control group(each n=20).The operation group was treated intragastrically with distilled water(1 mL/time,qd) after femoral artery operation with polyethylene tube around,and treatment group,intragastrically valsartan(10 mg/kg,qd).In sham-operation group femoral artery was liberated without polyethylene tube around and intragastrical medicinal.The control group was not given operation and intragastrical medicinal.On the 14th day,all mice were sacrificed and injured femoral artery was taken from tube place(from corresponding place in control group).The changes of vascular morphology were observed with light microscope after HE staining.The areas of intima and tunica media of new vessels were measured by using NIH analysis software.Results The tunica media was not thickened in all groups and the difference was not statistically significant compared with that before treatment(P>0.05).The intima was not thickened in sham-operation group and control group and the difference was not statistically significant compared with that before treatment(P>0.05).The intima was thickened and intima area was enlarged in operation group and treatment group,and the thickening was the most significant in operation group but the difference was not statistically significant compared with treatment group(P>0.05).The difference in intima area compared between operation group or sham-operation group and control group was statistically significant(P<0.05).Conclusion The short-term administration of valsartan can inhibit the intima thickness of injured vessels and vascular remodeling to some extent。
