Protective effect and mechanism of neutrophil gelatinase-associated lipocalin on ischemia-reperfusion renal injury in rats

Abstract:Objective To investigate the protective effect and mechanism of neutrophil gelatinase-associated lipocalin (NGAL) on ischemia-reperfusion (I/R) renal injury in rats. Methods Thirty adult male Sprague-Dawley rats were randomly divided into three groups (n=10). The models of renal injury were creat by unilateral nephrectomy and 45-minute I/R. In the I/R+NGAL group, 50 μg NGAL was dissolved into 2 mL sterile water and administered by tail-vein injections at 30 mins before renal pedicle clamping, during renal pedicle clamping, and at 30 mins and 60 mins after renal pedicle clamping. Meanwhile, the same amount of normal saline was injected in the sham operation group and I/R+NS group. After 24-hour reperfusion, blood sample was harvested from inferior vena cava to measure the serum creatinine (sCr) and blood urea nitrogen (BUN). Renal tissue sections were stained with hematoxylin-eosin. The expressions of B cell lymphoma/leukemia-2 associated X protein (Bax) and activated cysteinyl aspartate-specific protease-3 (Caspase-3) were detected by immunohistochemistry. TUNEL meathod was used to examine the apoptosis of tubular cells. Results After 24-hours reperfusion, sCr and BUN were significantly higher than those in sham operation and I/R+NGAL groups (all P<0.05). The I/R+NGAL group displayed an attenuated histopathologic response as compared with I/R+NS group under light microscope. The expression rates of Bax and Caspase-3 were (3.72±0.48)% and (0.61±0.15)% in sham operation group and (7.44±1.64)% and (3.17±0.32)% in I/R+NGAL group, which were significantly lower than those in I/R+NS group (15.46±1.96% and 7.29±1.06%, P<0.05). The apoptosis cells were 1.80±0.84 per high power field (HPF) in sham operation group and 7.80±1.92 per HPF in I/R+NGAL group, which were significantly lower than that in I/R+NS group (15.40±3.05 per HPF, P<0.05). Conclusion Exogenous NGAL can inhibit the activation of Caspase-3 and reduce the expression of Bax. Thus it can protect kidney from I/R injury and provide new therapeutic targets for acute kidney injury.
