Changes in the expression of liver tight junction protein in dextran sulfate sodium induced ulcerative colitis rat model

Abstract:Objective To investigate the expression of liver tight junction,liver function and its histopathological change in experimental ulcerative colitis.Methods Sprague-Dawley(SD) rats were randomly divided into two groups,experimental group fed with 3% dextran sulfate sodium(DSS) in the drinking water for six days and the control group.Histological examination was performed in liver and colon.The liver function was assessed by serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(AP),and total bilirubin(Tbili) level.Real time RT-PCR was used to detect the expression of seven proteins including occludin,ZO-1,claudin1,claudin2,claudin3 and claudin.Results Compared to control group,DSS treated rats showed weight lose(P<0.05),shorter colon length(P<0.01),and prominent colitis in distal colon characterized by dilations of gland crypt,ulcer,and inflammatory cell infiltration.The inflammation was the most severe at the 14th day,and then tended to subside.DSS group had increased serum ALT and decreased liver weight(P<0.01).Although inflammatory cell infiltration and focal injury in the liver were found in some DSS-treated rats,they were not significantly different from control group.Compared with control group,DSS treated rats had lower expression of ZO-1,claudin1,claudin2 mRNA(P<0.01),but higher expression of claudin7 mRNA(P<0.05).Conclusions A six-day treatment of 3% DSS in drinking water followed by 14 days of tap wate induced a colitis model in SD rats which was characterized by acute injury and slow recovery.In addition,DSStreated rats showed increased ALT and changes of expression of tight junction protein,suggesting that DSS-induced ulcerative colitis rat had liver cell injury and impaired liver barrier function.
