H2 O2 is required for abscisic acid-regulated maize primary root growth at low water potentials

Abstract:Maize(Zea mays L.'Nongda 108',from Nanjing Agricultural University,China)was used in this experiment and 25% PEG 6000 was used to simulate low water potentials(-1.6 MPa).The objective of this experiment was to determine whether H2O2 is involved in abscisic acid(ABA)maintained primary root growth at low water potentials.At low water potentials,seedlings were pretreated with the ABA inhibitors fluridone(FLU,10 μmol·L-1)and tungstate(Tun,1 mmol·L-1),both of them largely inhibited the primary root elongation,and the addition of 100 μmol·L-1 ABA almost fully prevented the inhibition of root elongation.Transmission electron microscopy was used to find that a little amount of H2O2 accumulated in the maize primary root at low water potentials.Using the scavengers of H2O2 to pretreat the seedlings and then exposing them to low water potentials,the primary root elongation was largely inhibited.In summary,these results show that ABA maintains maize primary root growth at low water potentials and H2O2 is involved in this process.If all the H2O2 is removed at low water potentials,the elongation of the primary root will be greatly inhibited.
