Abstract:To develop a RP-HPLC method for the determination of dioscin in purple Dioscorea alata Linn.After being extracted by refluxing with 70% MeOH for 1 hour,samples were achieved and separated by DiamonsilTM C18 column(250mm×4.6mm,5μm)with methanol-water(84∶16)as mobile phase.The flow rate was 1mL·min-1.The column temperature was 30℃.The UV detector was set at 206nm.Dioscin was well-separated.The linear calibration curve was obtained in the concentration range of 0.65~65μg·mL-1(r=0.9999),the average recovery of dioscin was 100.6%.The contents of dioscin in purple Dioscorea alata Linn.of different habitats was in larger difference.The method was simple,accurate and reproducible.It was also proposed to be used for the quality control of dioscin in Dioscorea opposite Thunb and Dioscorea alata Linn.
