Effects of superparamagnetic iron oxide nanoparticles labeling on ability to survive of retinal stem cells in vitro

Abstract:AIM:To evaluate the effect of superparamagnetic iron oxide (SPIO) nanoparticles labeling on the ability to survive of retinal stem cells (RSCs) in vitro and to provide a possible way of magnetic resonance imaging (MRI) to trace the implanted labeled cells in retina.METHODS: RSCs were prepared from the retina of embryonic 19-day SD rats and labeled with different concentration of SPIO (Fe3+5.6mg/L, 11.2mg/L, 16.8mg/L, 22.4mg/L). Prussian blue staining and electron microscope were used to evaluate the labeling efficiency of SPIO labeled RSCs. Cell proliferation and apoptosis were quantified by fluorescence activated cell sorter (FACS), MTT and Tunel. RESULTS: More than 90% of the cells were positively labeled and iron nano particles were found in their endochylema by the TEM when SPIO was at the concentration of 11.2-16.8mg/L in the medium. Labeling rate was below 60% when SPIO was under 5.6mg/L. Dead cells increased when SPIO was above 22.4mg/L in the medium. MRI showed remarkable low-signal intensity changes in part of the rat retina transplanted with labeled RSCs by subretinal injection.CONCLUSION: The efficiency of labeling is high and SPIO labeling does not affect the ability to survive of RSCs in vitro when SPIO is at concentration of 11.2-16.8mg/L in medium. SPIO can be used to label RSCs in vitro and MRI plays a role in tracking SPIO-labeled RSCs after transplantation by subretinal injection.
