Effect of External Factors on Formation and Growth of Free-living Conchosporangia in Porphyra yezoensis

Abstract:In this paper,effects of external factors of different photoperiod(8L: 16d,12L: 12d and 16L: 8d),temperature(15℃,20℃,25℃)and light intensity(29,42,57,72 and 86 μmol m-2s-1)on the free-living conchosporangia formation from free-living conchocelis filament cuttings and growth in Porphyra yezoensis HB,line were studied.When free-living conchocelis were cultured at 25℃ and 57μmol m-2·s-1 for 30 days,the formation rates of free-living conchosporangia in short photoperiod(8L: 16d)groups were the highest,as high as 92.5%.When they were cultured for more than 50 days,formation rates reached up to 100%.Cultured at 57μmol m-2·s-1 and 8L:16d for 30 days,the formation rates of free-living conchosporangia in 57μmol m-2s-1 light intensity groups,were 92.2%.If light intensity was too high or too low,the conchosporangia formation would be inhibited.With 57μmol m-2·s-1 and 8L:16d,formation rates of free-living conchosporangia at 25℃ groups was the highest,and free-living conchosporangia rate would decrease with temperatures decreasing.At 57μmol m-2·s-1,12 L:12d and 25℃,daily relative growth rate of free-living conchosporangia reached as high as 54.43%.The study data would lay an important foundation for further development of new breeding technology used for free-living conchosporangia.
