Abstract:Oxidative stress has been implicated in various pathological processes, including skin tumourigenesis. Cutaneum carcinoma is commonly responsible for considerable morbidity and mortality, and treatments have not progressed substantially in recent years. Alternative strategies, such as chemoprevention, are being considered. In this study, we investigated the chemomodulatory potential of lycopene against 9,10-dimethylbenz[a]anthracene (DMBA)/12-O-tetradecanoylphorbol-13-acetate (TPA)-induced oxidative stress and skin carcinogenesis in female ICR mice. Pretreatment with lycopene at various doses significantly delayed tumour formation and growth. These treatments markedly reduced the tumour incidence and tumour volume. Moreover, lycopene inhibited the formation of reactive oxygen species and malondialdehyde, prevented the loss of glutathione, and affected the activities of a battery of oxidant enzymes in the skin of mice. Furthermore, mice that were administered lycopene exhibited higher levels of translocation of nuclear-factor-erythroid-2-related factor 2 into the nucleus compared with the vehicle-treated and model mice. Collectively, these results indicated that lycopene exerts a protective effect against DMBA/TPA-induced cutaneum carcinoma through antioxidant defence.

Abstract:Carotenoids is a family of nutrient and bioactive components in vegetable and fruits.Recently considerable epidemical studies found that the vegetable and fruits containing carotenoids could reduce the risk of some chronic diseases.The bioactivities of carotenoids except for the sources of VA are focused on again,especially the common antioxidant activity of all the carotenoids,the preventing of prostate cancer of lycopene,the protecting of eyes and skin and the preventing of osteoporosis.The paper briefly introduced the structure and occurrence in food and human body,emphasized especially the new researches of bioactivities,and discussed the problems and possible researching fields.

Abstract:Lycopene is an important pigment with an alkene skeleton from Lycopersicon esculentum, which is also obtained from some red fruits and vegetables. Lycopene is used in the food field with rich functions and serves in the medical field with multiple clinical values because it has dual functions of both medicine and food. It was found that lycopene was mainly isolated by solvent extraction, ultrasonic-assisted extraction, supercritical fluid extraction, high-intensity pulsed electric field-assisted extraction, enzymatic-assisted extraction, and microwave-assisted extraction. Meanwhile, it was also obtained via 2 synthetic pathways: chemical synthesis and biosynthesis. Pharmacological studies revealed that lycopene has anti-oxidant, hypolipidemic, anti-cancer, immunity-enhancing, hepatoprotective, hypoglycemic, cardiovascular-protective, anti-inflammatory, neuroprotective, and osteoporosis-inhibiting effects. The application of lycopene mainly includes food processing, animal breeding, and medical cosmetology fields. It is hoped that this review will provide some useful information and guidance for future study and exploitation of lycopene.

Abstract:Lycopene, a potential bioactive agent, is a non-pro-vitamin A carotenoid recognized as a potent antioxidant. It is extracted from plants like tomatoes, watermelons, red carrots and papayas and has remarkable health benefits. A significant amount of research has been assisted to date to establish the anticancer activity of lycopene. Our review enhances information about the promising anticancer potential of this compound. The biological activity of lycopene has been described in several studies in regard to pancreatic, breast, prostate, liver, gastric, ovarian, kidney, skin, intestine, brain and spinal cord cancers. Lycopene resists cancer by inhibition of apoptosis, induction of cell proliferation, cell invasion, cell cycle development, metastasis and angiogenesis. The mechanisms of anticancer action of lycopene are attributed to the management of certain signal transduction pathways, such as modulation of insulin-like growth factors system, PI3K/Akt pathway, modification of important gene expression, inhibit the activity of sex steroid hormones, and the conversation of mitochondrial behavior. Hence, this review focuses on current knowledge of sources, extraction techniques, and chemistry of lycopene, as well as the prospective mechanisms of action related with its anticancer activity. Also, it summarizes the background information about lycopene and the most current research with consideration to its aspect in treating several types of cancer together with future directions.

Abstract:Lycopene has been widely applied in the food and medical fields due to having antioxidant, anticancer, and anti-inflammatory properties. Traditional lycopene production processes including natural extraction and chemical synthesis cannot meet the growing market demand and the pursuit for natural products of consumers. Microbial synthesis of lycopene has offered a promising alternative owing to its unique advantages. Industrial yeasts can produce lycopene by introducing heterologous synthetic pathways, which have become a reliable process to replace plants for lycopene production. In order to increase the lycopene production in industrial yeast, some strategies such as increasing the supply of precursors, looking for the optimal combination of lycopene synthesis genes, modularizing the enzymes in the metabolic pathway, increasing the content of cofactors, reducing product toxicity to cells, and optimizing fermentation methods make industrial yeast an effective strain for producing lycopene. This review mainly summarizes the research progress of lycopene produced by several industrial yeasts, analyzes the factors affecting the lycopene synthesis in the metabolic pathways, and summarizes and prospects the methods and strategies used to achieve high yields of lycopene in yeast to help with future research.

Abstract:Carotenoids are tetraterpene compounds acting as precursors to vitamin A, with functions that include protecting eyesight, enhancing immunity, promoting cell growth and differentiation, and providing antioxidative benefits. Lycopene, β-carotene, and astaxanthin are particularly critical for health and have diverse applications in food, health products, and medicine. However, natural carotenoids are encased within cell structures, necessitating mechanical methods to disrupt the cell wall for their extraction and purification—a process often influenced by environmental conditions. Thus, improving the efficiency of carotenoid extraction from natural resources is of great interest. This review delves into the research progress made on the extraction processes, structures, and biological functions of carotenoids, focusing on lycopene, β-carotene, and astaxanthin. Traditional extraction methods primarily involve organic solvent-assisted mechanical crushing. With deeper research and technological advancements, more environmentally friendly solvents, advanced machinery, and suitable methods are being employed to enhance the extraction and purification of carotenoids. These improvements have significantly increased extraction efficiency, reduced preparation time, and lowered production costs, laying the groundwork for new carotenoid product developments.

Abstract:Carotenoids are one of the most important groups of natural food colorants beneficial for human health. Carotenoids exist widely in higher plants, alga, a few microorganisms and some animals, while biosynthetic pathways and compositions of carotenoids accumulated vary greatly. Through optimization of culture conditions, application of genetic transformation and hydrolases-assisted extraction, production of carotenoids was greatly improved and the cost reduced, facilitating the wider consumption of natural carotenoids as healthy food colorants.

Abstract:Lycopene, a popular antioxidant, exists abundantly in nature in all-E form, but the Z-lycopene has better solubility and absorption characteristics. Lycopene is labile to light, thermal and oxygen. Emerging technological approaches are developed to improve the stability of lycopene during production and processing. This review presents the degradation mechanism of lycopene. A comprehensive evaluation has made by comparing the advantages and disadvantages of different extraction and isomerization methods. Encapsulation of lycopene in different delivery systems can further improve its stability. The process and formulation influence factors, as well as the advantages and disadvantages of lycopene encapsulation delivery systems are summarized. In the future, it is necessary to scientifically design low-cost and high-efficiency lycopene production methods.The stable and high bioavailable lycopene encapsulation systems need to be designed. Further study on the chemical stability mechanism of lycopene, the toxicity of biological materials, the safety of lycopene preparations also need to be evaluated.

Abstract:Sesquiterpenes and tetraterpenes are classes of plant-derived natural products with antineoplastic effects. While plant extraction of the sesquiterpene, germacrene A, and the tetraterpene, lycopene suffers supply chain deficits and poor yields, chemical synthesis has difficulties in separating stereoisomers. This review highlights cutting-edge developments in producing germacrene A and lycopene from microbial cell factories. We then summarize the antineoplastic properties of β-elemene (a thermal product from germacrene A), sesquiterpene lactones (metabolic products from germacrene A), and lycopene. We also elaborate on strategies to optimize microbial-based germacrene A and lycopene production.

Abstract:Background: Lycopene is a natural red compound with potent antioxidant activity that can be utilized both as pigment and as a raw material in functional food, and so possesses good commercial prospects. The biosynthetic pathway has already been documented, which provides the foundation for lycopene production using biotechnology. Aim of review: Although lycopene production has begun to take shape, there is still an urgent need to alleviate the yield of lycopene. Progress in this area can provide useful reference for metabolic engineering of lycopene production utilizing multiple approaches. Key scientific concepts of review: Using conventional microbial fermentation approaches, biotechnologists have enhanced the yield of lycopene by selecting suitable host strains, utilizing various additives, and optimizing culture conditions. With the development of modern biotechnology, genetic engineering, protein engineering, and metabolic engineering have been applied for lycopene production. Extraction from natural plants is the main way for lycopene production at present. Based on the molecular mechanism of lycopene accumulation, the production of lycopene by plant bioreactor through genetic engineering has a good prospect. Here we summarized common strategies for optimizing lycopene production engineering from a biotechnology perspective, which are mainly carried out by microbial cultivation. We reviewed the challenges and limitations of this approach, summarized the critical aspects, and provided suggestions with the aim of potential future breakthroughs for lycopene production in plants.
Lycopene in protection against obesity and diabetes: A mechanistic review

Abstract:Lycopene, a natural pigment that mainly exists in the mature fruit of tomatoes, has gained increasing attention due to its protective effects against obesity and diabetes. The aim of this review is to summarize the potential mechanisms in which lycopene exerts protection against obesity and diabetes, along with highlighting its bioavailability, synthesis and safety. Literature sources used in this review were from the PubMed Database, China Knowledge Resource Integrated Database, China Science and Technology Journal Database, National Science and Technology Library, Wanfang Data, and the Web of Science. For the inquiries, keywords such as lycopene, properties, synthesis, diabetes, obesity, and safety were used in various combinations. About 200 articles and reviews were evaluated. Lycopene exhibits anti-obesity and anti-diabetic activities in different organs and/or tissues, including adipose tissue, liver, kidney, pancreas, brain, ovaries, intestine, and eyes. The underlying mechanism may be attributed to its anti-oxidant and anti-inflammatory properties and through its ability to regulate of AGE/RAGE, JNK/MAPK, PI3K/Akt, SIRT1/FoxO1/PPARγ signaling pathways and AchE activity. The epidemiological investigations support that lycopene consumption may contribute to lowering the risk of obesity and diabetes. The cis-isomers of lycopene are more bioavailable and better absorbed than trans-lycopene, and mainly distribute in liver and adipose tissue. Lycopene exhibits a good margin of safety and can be obtained by plant extraction, chemical synthesis and microbial fermentation. In summary, lycopene consumption beneficially contributes to protecting against diabetes and obesity in animal studies and epidemiological investigations, which supports the potential of this compound as a preventive/therapeutic agent against these disorders. Well-designed, prospective clinical studies are warranted to evaluate the potential therapeutic effect of lycopene against common metabolic diseases.

Abstract:Background: Lycopene, one of the carotenoids, was increasingly attractive thanks to the significant nutritional and pharmaceutical value, causing the numerous demands of non-chemically synthesized lycopene. However, the insufficient lycopene production led to the high price. One potential way to ameliorate the situation was to elevate the extraction efficiency of lycopene by emerging technologies such as ultrasound-assisted extraction, microwave-assisted extraction. However, there is no comprehensive evaluation of these technologies. Therefore, this review is to comprehensively evaluate these technologies. Scope and approach: The market investigations indicated the huge demands of lycopene. Rising challenges spawned many new assisted technologies to weaken the hindrances in lycopene extraction. This review mainly discussed the emerging technologies for natural lycopene extraction. In addition, the evaluation of various technologies was performed, including the influences on isomerization and degradation, economic analysis and sustainability analysis. Key findings and conclusions: Hindrances of lycopene extraction from cells were illustrated. Besides, almost all the treatments could elevate the efficiency to different extents, which offered various ideas and pathways for lycopene extraction. And the summarization of advantages and disadvantages indicated the promising application potential. Many technologies could affect the isomerization and degradation of lycopene, indicating the necessity to control the extraction conditions. Statistical analysis presented comprehensive comparisons of key indicators (temperature, time, L/S and yield), and supported further evaluation. The economic analysis and sustainability analysis indicated that the cost and environmental effects varied with methods. In the future, methods with low cost, high efficiency and environment-friendly effects will be preferred.

Abstract:Lupeol is a natural triterpenoid that widely exists in edible fruits and vegetables, and medicinal plants. In the last decade, a plethora of studies on the pharmacological activities of lupeol have been conducted and have demonstrated that lupeol possesses an extensive range of pharmacological activities such as anticancer, antioxidant, anti-inflammatory, and antimicrobial activities. Pharmacokinetic studies have indicated that absorption of lupeol by animals was rapid despite its nonpolar characteristics, and lupeol belongs to class II BCS (biopharmaceutics classification system) compounds. Moreover, the bioactivities of some isolated or synthesized lupeol derivatives have been investigated, and these results showed that, with modification to C-3 or C-19, some derivatives exhibit stronger activities, e.g., antiprotozoal or anticancer activity. This review aims to summarize the advances in pharmacological and pharmacokinetic studies of lupeol in the last decade with an emphasis on its anticancer and anti-inflammatory activities, as well as the research progress of lupeol derivatives thus far, to provide researchers with the latest information, point out the limitations of relevant research at the current stage and the aspects that should be strengthened in future research.

Abstract:Nutritional antioxidants derived from phytochemicals have been shown to have multifaceted health benefits via the pleiotropic molecular mechanisms. Lycopene is one such aliphatic hydrocarbon carotenoid with no beta-ionone and imparts red coloration to watermelon, tomatoes, and papayas. It is a potent antioxidant and free radical scavenger owing to the presence of 13 carbon–carbon double bonds, out of which 11 are conjugated and the remaining 2 are non-conjugated. It has many biological effects on different types of cancers like prostate, breast, skin, and oral cancer by targeting the Keap1-NF-kB, Keap1-Nrf 2 and PI3K/AKT/m-TOR signalling pathway. In addition to cancer, lycopene also has the ability to modify the gut microbiome, and also work against SARS-CoV-2. This review focuses on the sources, isomers, metabolites, therapeutic targets, mechanistic action, nanoformulations, pharmacokinetics, completed and ongoing clinical trials. In addition to above we are adding the potential ability of lycopene in obesity, infertility, metabolic disorder, gut microbiome, and COVID-19 treatment.

Abstract:The lamellarins, a group of natural pyrrole alkaloids with interesting anticancer activities, were firstly isolated from a prosobranch mollus Lamellaria sp.. Due to their multidrug resistance (MDR) reversal in some cancer cell lines, total synthesis and structure modifications of lamellarins to provide drug candidates for the treatment of cancer have become one of the most promising project in the area of novel anticancer drug discovery. In this article, the latest results and progresses regarding the synthesis and modification of lamellarins are reviewed.

Abstract:Lycopene is a dark red carotenoid belonging to C40 terpenoids and is widely found in a variety of plants, especially ripe red fruits and vegetables. Lycopene has been shown to reduce the risk of prostate cancer, other cancers, and cardiovascular disease. It is one of the most widely used carotenoids in the healthcare product market. Currently, commercially available lycopene is mainly extracted from tomatoes. However, production of lycopene from plants is costly and environmentally unfriendly. To date, there have been many reports on the biosynthesis of lycopene by microorganisms, providing another route for lycopene production. This review discusses the lycopene biosynthetic pathway and natural and engineered lycopene-accumulating microorganisms, as well as their production of lycopene. The effects of different metabolic engineering strategies on lycopene accumulation are also considered. Abstract:Carotenoid pigments, particularly β-carotene and lycopene, are consumed in human foodstuffs and play a vital role in maintaining health. β-carotene is known to quench singlet oxygen and can have strong antioxidant activity. As such, it was proposed that β-carotene might reduce the risk of cancer. Epidemiological studies found inverse relationships between cancer risk and β-carotene intake or blood levels. However, clinical trials failed to support those findings and β-carotene supplementation actually increased lung cancer incidence in male smokers. Early experimental animal studies found dietary β-carotene inhibited UV-induced skin cancers. Later studies found that β-carotene supplementation exacerbated UV-carcinogenic expression. The discrepancies of these results were related to the type of diet the animals consumed. Lycopene has been associated with reduced risk of lethal stage prostate cancer. Other carotenoids, e.g., lutein and zeaxanthin, play a vital role in visual health. Numerous studies of molecular mechanisms to explain the carotenoids’ mode of action have centered on singlet oxygen, as well as radical reactions. In cellular systems, singlet oxygen quenching by carotenoids has been reported but is more complex than in organic solvents. In dietary β-carotene supplement studies, damaging pro-oxidant reactivity can also arise. Reasons for this switch are likely due to the properties of the carotenoid radicals themselves. Understanding singlet oxygen reactions and the anti-/pro-oxidant roles of carotenoids are of importance to photosynthesis, vision and cancer.

Abstract:Efficient and mild synthetic routes for bioactive natural product derivatives are of current interest for drug discovery. Herein, on the basis of the pharmacophore hybrid strategy, we report a two-step protocol to obtain a series of structurally novel oleanolic acid (OA)-dithiocarbamate conjugates in mild conditions with high yields. Moreover, biological evaluations indicated that representative compound 3e exhibited the most potent and broad-spectrum antiproliferative effects against Panc1, A549, Hep3B, Huh-7, HT-29, and Hela cells with low cytotoxicity on normal cells. In terms of the IC50 values, these OA-dithiocarbamate conjugates were up to 30-fold more potent than the natural product OA. These compounds may be promising hit compounds for the development of novel anti-cancer drugs.
