Effects of IgY, coptis chinensis and radix pseudostellariae on gastric mucous membrane in mice with helicobacter pylori infection

Abstract:AIM: To observe the morphological changes of gastric mucous membrane (GMM) before and after being treated by IgY, coptis chinensis (CC) and radix pseudostellariae in mice with helicobacter pylori (HP) infection. METHODS: The experiment was conducted in the Department of Immunology, Jiangxi Medical University from September 2003 to March 2004. Thirty-six mice of Kunming species were randomly divided into 6 groups with the number ranged from 1 to 6∶ IgY group, CC group, radix pseudostellariae group, colloidal bismuth subcitrate (CBS) group, saline group and control group. Except the control group, rats in other 5 groups were used to establish HP infection models (GMM samples were isolated to culture, perform the morphological examination and urease test as infectious index of experimental animals, and the successful model was positive after isolated culture or for the later 2 items). The control group adopted the normal mice without HP infection. At 4 weeks after modeling, mice were treated with intragastric administrations of 20 g/L IgY, 180 g/L radix pseudostellariae, 60 g/L CC, 1.8 g/L CBS, 0.5 mL saline and 0.5 distilled water in the corresponding group respectively, once a day for 5 continuous days. Four weeks after administrations, all the mice were executed to observe the general and pathological changes of GMM. Referring to Sydney system, all the mice were evaluated for GMM affection levels and ranked by 4 grades in accordance with inflammatory level, inflammatory activity, atrophic gastritis, intestinal metaplasia and gastritis locus, as well as scored correspondingly (0=none, 3=severe). GMM affection was assessed with the summation of 5 items, and 6 mice of each group were scored respectively. RESULTS: Totally 36 mice were involved in the result analysis without any loss.(1)HP infectious rate of GMM in mice: The GMM of mice at 4 and 8 weeks after infection were positive in smear-microscopy, urease test and Warthin-Starry stain. The infectious rate of HP was 100%.(2)Comparison of inflammation scores: The analysis of variance indicated that there were significant differences between saline group and other groups (F=3.98, P < 0.05). The paired comparison by SNK method demonstrated that, there was obvious promoting effect of IgY on the turnover of HP-induced inflammation, compared with saline group (Six mice were scored as 2, 3, 5, 3, 6, 2; 10, 8, 11, 7, 7, 5, P < 0.05). But the effect was higher in CC group and radix pseudostellariae group than in IgY group (Six mice were scored as 4, 6, 3, 3, 4, 9; 5, 6, 6, 4, 6, 8). CONCLUSION: Oral administration of IgY is an effective method that can control the GMM inflammation induced by HP, and the interventional effect is superior to those of coptis chinensis and radix pseudostellariae.
