Detection of Ingredients in Yinchenhao Decoction by the Method of RP-HPLC Gredient Elution

Abstract:Objective: To establish an optimal method to analyze “Yinchenhao decoction” by RP-HPLC. Methods: Lichrospher C18 column (4.6 mm×25 cm, 5μm ) with temperature at 25 ℃ was used with a mobile phase of A (0.5 ‰ phosphoric acid ) ,B(methanol with 0.5 ‰ phosphoric acid), used a non-linear gradient of 90 % to 40 %.A phase in 50 minutes and from 40 % to 28.5 % in the followed 5 minutes mixed with B phase. The elution was performed at the flow rate of 1 m l / min. UV detection wavelength was 254 nm, the injection volume was 5μL . Results: The ingredients in Yinchenhao decoction were well seperated by gredient elution. Conclusion: The ingredients in Yinchenhao decoction were well presented in the chromatographic detection via this method, and the method can be applied for the study of the fingerprints of Yinchenhao decoction.
