Abstract:Three chalcone derivatives were synthesized by the reaction of acetophenone with dimethylamino benzaldehyde, vanillin, diphenyl formaldehyde, respectively. The results showed that: A yield of 80.4% was obtained by the reaction of acetophenone and dimethylamino benzaldehyde in ethanol as solvent, 10% NaOH solution as catalyst at room temperature. A yield of 25.0% was obtained by the reaction of acetophenone and vanillin in ethanol as solvent, a mixed glacial acetic acid and hydrochloric acid as the catalyst, adding 10% KOH solution for dehydration by heating after the reaction at room temperature. A yield of 76.3% was obtained by the reaction of acetophenone and diphenyl formaldehyde in ethanol as solvent, 10% NaOH solution as catalyst at room temperature. Acid was used as the appropriate catalyst when chalcone derivatives were synthesized by aldehyde with hydroxy.
Synthesis of 1-Chloro-2,5-dimethoxybenzene

Abstract:1-Chloro-2,5-dimethoxybenzene was prepared from p-benzoquinone through chlorination and methylation with total yield of 78%. p-Benzoquinone was firstly converted catalytically to 1-chloro-2,5-dihydroxybenzene in the binary system of dichloroethane and hydrochloric acid at 35℃ with yield of 89%, then the final product was synthesized by methylation of obtained 1-chloro-2,5-dihydroxybenzene with dimethyl sulfate in the presence of sodium hydroxide. The intermediate 1-chloro-2,5-dihydroxybenzene and the obtained title compound 1-chloro-2,5-dimethoxybenzene were characterized by IR and GC-MS respectively. The results showed that their chemical structures were correct and the purity of title compound was over 98%.

Abstract:The chalcone was synthesized by the Claisen-Schmidt condensation of acetophenone with aromatic aldehydes using the phase transfer catalyst polyethylene glycol (PEG). The influence of some factors,such as the quantity of the catalyst, the molecular weight of PEG and the type of the solvent,on the product yield was investigated. At the optimum experimental conditions,the yield can be 80%.

Abstract:Several 3,3'-(1,4-phenylene)bis(1,5-diones) and their chalcone precursors have been prepared in good to excellent yield via aldol addition and Michael addition starting from 3-acetyl-2,5-dimethylfuran or 3-acetyl-2,5-dimethyl-thiophene with terephthalaldehyde in the presence of appropriate base NaOH or lithium diisopropylamide. The kind and amount of alkali played a critical role in improving the reaction rates and yields of the products.

Abstract:A new intermediate containing dihydrochalcone moiety, i.e., bis(dihydrochalcone) diphenyl ether (BHCDPE), was first synthesized by the Friedel-Crafts alkylation reaction of chalcone and diphenyl ether (DPE). Subsequently, a series of novel poly(aryl ether ketone) (PAEK) polymers were prepared by further Friedel-Crafts polycondensation of BHCDPE and DPE with isophthaloyl dichloride (IPC). It was observed that BHCDPE exhibited strorkg reactivity during Friedel-Crafts polycondensation, i.e., one non-conjugated gamma-carbonyl would not decrease the Friedel-Crafts acylation's reactivity of corresponding benzene rings, such that the dihydrochalcone moiety could be successfully incorporated into the backbone of PAEK to form novel PAEKs with dihydrochalcone moiety. The synthesis route was a continuous procedure and displayed inexpensive and commercially available of the raw materials, mild reactive conditions, as well as high yields due to the absence of complicated process of intermediate treatment. The polymers' structures were characterized by FT-IR, H-1-NMR and XRD analysis. Moreover, obtained polymers maintained good thermal properties, and incorporation of a dihydrochalcone moiety significantly improved polymer solubility in organic solvents, which was advantageous for further functionalization, processing and interactions with other substances. Thus, the designed synthesis route was appropriate to obtain novel soluble PAEKs containing dihydrochalcone moieties, which were expected as new potential candidates for more useful materials in various fields.
Alkali-catalyzed Synthesis of 4-Methyl-2'-hydroxyl-4'-methoxychalcone and Its Crystal Structure

Abstract:The synthesis method of chalcone was investigated under the assistance of ultrasonic irradiation with alkali as the catalyst.A novel paeonol derivative named 4-methyl-2'-hydroxyl-4'-methoxychalcone was synthesized in alkaline medium under the assistance of ultrasonic irradiation,the reaction of paeonol and 4-methylbenzaldehyde was by ClaisenSchmidt condensation reaction at room temperature,its yield reached 71.57％,its structure existing 1,3-diphenyl acrylic ketone was characterized by UV,IR,MS,1 H NMR,13 C NMR and 135° DEPT.The crystal structure of 4-methyl-2'-hydroxyl-4'-methoxychalcone was also characterized by X-ray structure analysis,its crystal belonged to space group P2(1) / n in triclinic crystallographic system,and its cell parameters:a =1.1288(10) nm,b =0.6916(6) nm,c =2.0509(4) nm,α =90.0(8) °,β =109.8(8) °,γ =90(8) °,V=1.4056 (5) nm3,Dc =1.272 mg/cm3.The novel paeonol derivative existing 1,3-diphenyl acrylic ketone in the molecular structure was synthesized through the reaction of paeonol and 4-methylbenzaldehyde under the assistance of ultrasonic irradiation with alkali as the catalyst.

Abstract:A serial of photosensitive diamines containing chalcone moiety was prepared using the materials aldehyde and ketone through the reaction of adol condensation. The diamines were named as 1-(3-aminophenyl)-3-(4-aminophenyl)-2-propen-1-one and 1-(3-aminophenyl)-3-{4-(3-(3-aminophenyl)-3-oxo-1-propenyl) phenyl}-2-pro-pen-1-one. Best conditions for synthetic process were attained by the optimization of experiments as follows: diamines were synthesized from aldehyde and ketone dissolved in alcohol,adding sodium hydroxide as catalyst with continuous stirring for 3h at 0℃ with total yield over 86% and the purity were 99.92% and 99.91% respectively by the analysis of HPLC. The method had the advantages of cheapness, high yield and easy operation. Elemental analysis, HNMR and IR characterized the target compounds.

Abstract:The paper has reported that commerical NaOH efficently catalyzes Michael reactions of chalcones (1) with acetophenone (2) under solvent-free conditions. And a series of 1,5-di aryldiketones can be obtained in 71-90% yields. All the product structures were characterized satisfactorily by IR. (HNMR)-H-1. MS spectroscopy and elemental analysis. The procedure is performed by grinding the components with an agate mortar and a pestile under solvent-free at room temperature or preheating at 45 degrees C less than 1.3 hours. The result indicated that the aromatic systems are more active than the ferrocenyl analogous.

Abstract:A structurally novel 1,3-benzodioxole-bearing indole-based chalcone,namely(E)-3-(benzo[d][1,3]dioxol-5-yl)-1-(2-chloro-1-(4-chlorobenzyl)-1H-indol-3-yl) prop-2-en-1-one(3) was synthesized,involving the ultrasound-assisted or solvent-free Claisen-Schmidt condensation reaction of 3-acetyl-2-chloro-1-(4-chlorobenzyl) indole and piperonaldehyde. The ultrasound-assisted Claisen-Schmidt condensation reaction was carried out using 1,4-dioxane as solvent and KOH as catalyst at room temperature to give the product 3 in 78%yield. Alternatively,the reaction could also be conducted under solvent-free condition by physical grinding in a mortar and pestle to obtain 3 in comparable yield of 75%. The two procedures offered easy access to 1,3-benzodioxole-bearing indole-based chalcone in short reaction time and good yield under mild conditions.

Abstract:An ionic liquid (IL) catalyzed solvent-free process was developed for the direct synthesis of chalcone and its derivatives by using substituted acetophenones and benzaldehydes via aldol reaction under mild conditions. A series of acidic and basic ILs were selected and screened. The influences of cations and reaction conditions on product yield and selectivity were systematically investigated. The [Bmim]OH was identified as the optimal IL, with the highest yield and selectivity reaching up to 96.7% and 100%, respectively. A reaction mechanism-based kinetic model was established and regressed with experimental data, revealing the β-Hydroxylketone dehydrolysis with activation barrier of 37.8 kJ·mol-1 was observed as the rate-controlling step.

Abstract:Herein, we aimed to study four sets/25 derivatives (3a-f , 4a-e , 4g , 5a-e , 5h , 6a-f and 6i) of simple chalcone's optical and electrochemical properties by altering the chemistry in the chalcone skeleton. The design of chalcone derivatives for the current study was made by creating donor–acceptor-donor/acceptor (D-A-D/A) or push–pull effect in the chalcone backbone. Based on the designed structures, the desired compounds were synthesised by Claisen-Schmidt condensation reaction from four aromatic ketones (1a-d) and seven aromatic aldehydes (2a-i) with 75–94 % yield. Our research interest is to investigate the tunable optical behaviour of these chalcones by varying the combinations of aldehydes and ketones. By doing so, the π-electrons push–pull effect experienced in the chalcone skeleton may vary by default. The theoretical studies of a few selected chalcone derivatives were performed to validate the planned hypothesis using Gaussian 09 W software. All the optimised structures were visualised using GaussView 5.0. DFT (Density Functional Theory) with the B3LYP method and 6–31 + g(d) basis set were used to explain the structure of the selected chalcones via optimisation and FMOs (Frontier Molecular Orbitals). In CHCl 3 solvent, the effects of the absorption and emission results elucidate the effects of the substituents (electron donating group/EDG or electron withdrawing group/EWG) in the aldehyde ring on the optical properties of the chalcones. The compound's emission colour position in the Commission Internationale de l'Eclairage (CIE) chromaticity colour space guarantees its ability to emit visible light in different colour ranges. Among the four chalcone sets, the chalcones 6a-f and 6i exhibited better photophysical results in the CHCl 3 solvent. Hence, the solid-state photophysical characteristics of the same set of chalcones were investigated and recorded in the present work. The chalcone 6d displayed the highest quantum yield of 41.28 %, and 6c had the maximum fluorescence lifetime of 24.5 ns. The chalcone family members are already proven candidates for various applications in multidisciplinary fields; the results produced here may aid future researchers in finding more applications by utilising the recorded properties.

Synthesis, electronic, and spectral properties of novel geranylated chalcone derivatives: a theoretical and experimental study

Abstract:Novel chalcone derivatives with different substituents attached to A and B-rings: hydroxyl, methoxyl, geranyl, and prenyl groups were synthesized. The obtained compounds were characterized by NMR, HRMS, UV-Vis, IR, and MS. The theoretical analysis was carried out in all the compounds using density functional theory (DFT) with the B3LYP, PBE0, and M06-2X functionals in combination with the 6-311G(d,p) Pople-type basis set. The excited state properties were calculated by time dependent density functional theory (TD-DFT) using the same methodology applied for the ground state properties. The calculated vertical absorption wavelengths (λmax) in gas phase and in ethanol as a solvent are consistent with the experimental ones, being the TD-DFT:B3LYP/6-311G(d,p) and PCM-TD-DFT:PBE0/6-311G(d,p) the best methodologies for these calculations with good approximation to the experimental values. The calculated reorganization energies indicated that, the four chalcone derivatives present an electron transfer character due to the smaller registered values. From these parameters it is proposed that these show an n-type semiconductor character. The localization of the frontier orbitals (HOMO and LUMO) shows that only the compound containing a hydroxyl group on the A-ring displays a marked delocalization favoring the charge-transfer process in this system. The HOMO-LUMO gap energies indicate that the inclusion of different donor groups in the rings does not improve the obtained values for this property. Abstract:Aims: In this paper, we report on the synthesis and liquid crystalline properties of some low molecular weight bis-chalcone compounds derived from acetone, cyclopentanone and cyclohexanone mesogenic cores. Background: Structurally bis-chalcones belong to a broader family of chalcone compounds. Chalcone is a compound that consists of two aromatic rings linked by an unsaturated α, β-ketone. Objective: Liquid crystalline chalcones are prepared by aliphatic chain substituents on two aromatic rings. Chalcones are well studied for their mesomorphic properties. Compared to a large number of chalcone based LCs reported, only a few articles have been published on the mesomorphic properties of bis-chalcone compounds. The target compounds of the present study varied not only in their central core but also in number and position of terminal aliphatic chain substitution-a key structural unit in deciding the liquid crystalline properties of a compound. Methods: All target compounds were synthesized in good yield by base catalyzed Claisen-Schmidt condensation reaction. Molecular structures were confirmed by FT-IR, 1H NMR, 13C NMR, and mass spectroscopic methods. Liquid crystalline property of these compounds was evaluated using polarizing optical microscopy and differential scanning calorimetry. Results: Although none of the acetone based compounds exhibited mesomorphism, cyclopentanone and cyclohexanone based compounds with octyloxy chain at para position on either side of the dibenzylidine ring stabilized liquid crystalline smectic (SmA and SmC) and nematic (N) phases. The observed structure-liquid crystalline property relationship was explained by structural analysis of molecules using DFT calculations. Considering the inherent photoluminescence nature of the chalcone moiety, a preliminary study was carried out on a selected compound to reveal its fluorescence property. Conclusion: Our study brings about an important structure-liquid crystalline property relationship in a relatively unexplored class of bis-chalcone liquid crystals.

Abstract:A novel series of unsymmetrical bis-quinolin-3-yl chalcones has been synthesized under visible light using a Claisen-Schmidt condensation reaction between the 2-(morpholine-piperidine-pyrrolidine-thiomorpholine) substituted quinoline-3-carbaldehyde and 1-(2-methyl-4-phenylquinolin-3-yl) ethan-1-one derivatives, conducted at room temperature in the presence of NaOH/EtOH. The structures of the synthesized compounds have been confirmed by NMR spectroscopy and high-resolution mass spectroscopy. The synthesized compounds exhibit λ max values ranging from 215 nm to 290 nm in non-polar to polar solvents, demonstrating positive solvatochromism. Compounds containing nitro group substituent exhibit a significant blue shift owing to their strong electron-withdrawing ability, which also contributes to fluorescence quenching. Geometrical optimization and the calculation of HOMO, LUMO, and energy gap are calculated by using the DFT/B3LYP/6-31G(d) method.

Abstract:Seven novel 3'-mono-substituted chalcone derivatives were synthesized from 2,4-dihydroxyacetophenone in 4 steps in 18-31% overall yield. Their structures were confirmed by H-1 NMR. The anticancer activity of these new target compounds (4 similar to 10) against human leukemia K562 cells were evaluated by MTT assay in vitro. This study leads to the identification of one highly active molecule, compounds 10 (IC50 = 5.60 mu M) against K562.

Abstract:Two chalcones were synthesized by the aldolic condensation of enolizable aromatic ketones with substituted benzaldehydes under Claisen-Schmidt reaction conditions and then treated with 2,4-dinitrophenylhydrazine to yield their corresponding hydrazones. The two (E,Z)-2,4-dinitrophenylhydrazone structures, namely (Z)-1-(2,4-dinitrophenyl)-2-[(E)-3-(4-methylphenyl)-1-phenylallylidene]hydrazine, C22H18N4O4, (H1), and (Z)-1-[(E)-3-(4-chlorophenyl)-1-(naphthalen-1-yl)allylidene]-2-(2,4-dinitrophenyl)hydrazine, C25H17ClN4O4, (H2), were isolated by recrystallization and characterized by FT-IR, UV-Vis, single-crystal and powder X-ray diffraction methods. The UV-Vis spectra of the hydrazones have been studied in two organic solvents of different polarity. It was found that (H2) has a molar extinction coefficient larger than 40000. Single-crystal X-ray diffraction analysis reveals that the molecular zigzag chains of (H1) and (H2) are interconnected through noncovalent contacts. A quantitative analysis of the intermolecular interactions in the crystal structures has been performed using Hirshfeld surface analysis. All the synthesized chalcones and hydrazones were evaluated for their antibacterial and antioxidant activities. Results indicate that the studied compounds show significant activity against Gram negative Escherichia coli strain and the chalcone 3-(4-methylphenyl)-1-phenylprop-2-en-1-one, (C1), was the most effective. In addition, only hydrazone (H1) displayed a moderate DPPH (2,2-diphenyl-1-picryl hydrazyl) scavenging efficiency.

Abstract:The intermediates including chalcone and 4-dimethylaminochalcone were synthesized from benzaldchyde, 4-dimethylamino-benzaldchyde and hypnone by Claisen-Schmidt reaction. And then using chalcone and 4-dimethylaminochalcone with thiosemicarbazide as raw material, two novel Schiff bases comprising thiosemicarbazone were synthesized by aldol condensation under the faintly acid condition. The compounds were characterized by elemental analysis, infrared, GC-MS and 1HNMR. The results show that in the infrared spectra of C=C and alkene C-H there are absorption peaks at 1644-1651, 3020-3034, 988-998 cm-1, respectively, indicating that the two Schiff bases are trans-isomerization. In the infrared spectra of C=S there are absorption peaks at 1219-1221 cm-1, lower than its characteristic frequency at 1230-1253 cm-1, and the chemical shifts of primary amine to low magnetic field occur greatly in reference to that of secondary amine, which indicates the molecules is combined with intermolecular hydrogen bonds and concludes that C=S exists mainly in the form of keto tautomer.
