Preparation of Capsules Filled with Nicotinic Acid Sustained-release Particles and Simvastatin Particles

Abstract:The nicotinic acid sustained-release particles and simvastatin particles prepared respectively by extruded method were filled into the capsules according to their specifications.Each capsule contained 250 mg nicotinic acid and 10 mg simvastatin.The release behavior of nicotinic acid and the dissolution behavior of simvastatin from the product were similar with the Simcor(reference tablets).The results of pharmacokinetics in Beagle dogs showed that the main parameters of nicotinamide of the product and reference tablets were as follows: cmax(15.8±6.0) and(13.2±3.2)mg/L,tmax(22±9) and(22±9)h,t1/2(5.3±1.7) and(6.5±5.1)h,AUC0→48 h(429.7±118.9) and(359.1±101.3)mg.h.L-1;the parameters of simvastatin were as follows: cmax(8.6±0.6) and(8.2±0.8) g/L,tmax(2.7±0.3) and(2.5±0.0)h,t1/2(1.4± 0.1) and(2.0±0.9)h,AUC0→48 h(42.7±3.6) and(41.8±4.3) g.h.L-1.The results indicated that the two preparations were bioequivalent.
