The effect of intratumoral implanted ~(125)I seeds on tumor growth and apoptosis-associated protein expression of Lewis Lung Cancer

Abstract:Objective To study the influence of 125I seeds, which were intratumoral implanted,on tumor growth and apoptosis-associated protein of Caspase-9 and Caspase-3 expression on Lewis Lung Cancer. Methods Lewis tumor were established by injecting intradermally 1×107 tumor cells (in 0.2ml of saline) into the flank of KM mice. When the tumor reached 0.20-2.5cm2,the tumor-bearing mice were divided into 3 groups (contal,5Gy and 10Gy). Then,the 125I seeds were implanted intratumoraly in the mice of 5Gy and 10Gy. Subsequently, the tumor diameters were measured and tumor growth curves were described. On day, tumors were each excised and Caspase-9 and Casepas-3 expression were detected by the method of immunocytochemistry. Results Before the seeds being implanted, the tumor volunmers of the 3 groups were almost quivalency and there were no significant differences (p>0.05). However, the tumor volume were markedly smaller in the group of 1.0mCi on day 4 (5Gy) and 9(10Gy) than that of contral group.In the group of 0.5mCi, the tumor size kept invariably on the day 9(5Gy), while on the day 19, they were smaller than that of contral group (p<0.05). As for the expression of Caspase-9, Casepas-3, they were evidently highly expressed in irradiated tumor and correlated with the tumor size (p<0.05 or p<0.01). Conclusions Low-dose irradiation treatment could markedly inhibited the tumor growth in vivo and this may be due to the fact of high expression of Caspase-9and Casepas-3, which could increase the tumor cells apoptosis.
