Effects of Infection of Epiphyte Caused Fruit Rot on Defense Mechanism in Pear Fruit Callus

Abstract:Using the pear fruit callus system,we determined the cell membrane permeability,MDA content and the activities of the defensive enzymes of the pear fruit callus inoculated by two kinds of fungi(Botryosphaeria berengriana f.sp.piricola and Monilinia fructigena Honcy),try to analyze the relationship between the defensive enzyme activities and disease process and their roles in disease resistance at the cellular level.The results showed that there was similar trend of increasing relative conductivity rate and MDA content of the callus infected by two kinds of fungi,but the change was smaller in callus infected by B.berengriana f.sp.piricola than in callus infected by M.fructigena Honcy.Within 120 h after inoculation,the activities of peroxidase(POD),superoxide dismutase(SOD),catalase(CAT),polyphenol oxidase(PPO) and phenylalanine ammonia-lyase(PAL) showed similar trend,increased at first and followed by a decrease.The peak of defensive enzyme activities appeared at different time points,but the enzyme activity peaks were higher in callus infected by B.berengriana f.sp.piricola than in callus infected by M.fructigena Honcy.It showed that defensive enzymes played important roles in the physiological defense response to the infection by the fungi.There is a positive correlation between the changes of the defensive enzymes and the resistance of callus to rotting.
