Abstract:A new "Meshed-Bag Gathered-Bunch" technology for the solid-phase synthesis of chemical libraries was developed. Using such technology, we synthesized muramyl dipeptide mimetics including derivatives at the N- and C-terminus, cyclic muramyl dipeptide mimetics, muramyl dipeptide and Tuftsin's analogue conjugates. The advantages of such a method include ease of manufacture, low unit cost of production, the physical encoding method, and the compatibility with both parallel and "split-mix" approaches.

Abstract:Herein, we report a novel method for highly efficient and stereoselective construction of bioactive 2-amino-2-deoxy-1,3-dithioglucosides as N -acetyl muramyl dipeptide analogues via organocatalyzed relay reactions with 3- O -acetyl-2-nitroglucal as the glycosyl donor. The method enjoys mild reaction conditions, efficient one-pot relay reaction, good to excellent yield, and broad substrate scope.

Abstract:Disulfide-constrained peptides (DCPs) have gained increased attention as a drug modality due to their exceptional stability and combined advantages of large biologics and small molecules. Chemical synthesis, although widely used to produce DCPs, is associated with high cost, both economically and environmentally. To reduce the dependence on solid phase peptide synthesis and the negative environmental footprint associated with it, we present a highly versatile, low-cost, and environmentally friendly bioproduction platform to generate DCPs and their conjugates as well as chemically modified or isotope-labeled DCPs. Using the DCP against the E3 ubiquitin ligase Zinc and Ring Finger 3, MK1-3.6.10, as a model peptide, we have demonstrated the use of bacterial expression, combined with Ser ligation or transglutaminase-mediated XTEN ligation, to produce multivalent MK1-3.6.10 and MK1-3.6.10 with N-terminal functional groups. We have also developed a bioproduction method for the site-specific incorporation of unnatural amino acids into recombinant DCPs by the amber codon suppression system. Lastly, we produced 15N/13C-labeled MK1-3.6.10 with high yield and assessed the performance of a semiautomated resonance assignment workflow that could be used to accelerate binding studies and structural characterization of DCPs. This study provides a proof of concept to generate functionalized DCPs using bioproduction, providing a potential solution to alleviate the reliance on hazardous chemicals, reduce the cost, and expedite the timeline for DCP discovery.

Abstract:An environmentally friendly solution-phase synthesis has been developed for producing trans-5-amino penta-3-enoic acid (trans-δ-Apa), along with several Nα- and C-terminal protected derivatives and its dipeptide. This method utilises low temperatures, minimal power, and short coupling times. Microwave-assisted synthesis demonstrates the production of non-proteogenic delta-amino acids and peptides with high purity, better yield, and scalability. The reaction is suitable for a wide range of carboxylic acid-derived amines and has been demonstrated on a gram-scale synthesis. The process involves running five cycles of ion-exchange resin, followed by regeneration. The replacement of central amide bonds by (E)-alkene in a δ-amino acid proves to be a promising approach for mimicking peptide backbone geometry in a dipeptide of α-amino acids. Introducing an (E) Cβ=C alkene unit enhances conformational limitation for δ-amino acid residues, resulting in peptidomimetics with conformational constraints. Successful implementation of this efficient synthetic approach has resulted in amino-terminal protected, carboxy-terminal protected, and both Nα-terminal and C-terminal protected δ-amino acids with good yields. HBTU/HOBt/DIEA coupling yields the best results for dipeptides, while o-NosylOXY/DIEA exhibits the next best coupling efficiency. Additionally, the choice of solvent has been optimised for better yields. Microwave dielectric heating significantly reduces reaction times, offering an instant, cost-effective alternative with moderate to good yields under mild conditions and a greener method than conventional thermal heating methods.

Abstract:To achieve the synthesis of head-to-tail cyclic peptides via the liquid-phase on-support cyclization and synergistic self-cleavage strategy, 4,4’-bis(diphenylphosphinyloxyl) diphenyl ketoxime (BDKO) and 4-diphenyl phospholoxy benzyl alcohol (DPBA) were designed and prepared as small-molecular supports for greener peptide synthesis. During liquid-phase peptide synthesis on BDKO (or DPBA) support for peptide chain extension by Boc protocol, the last amino acid at the N-terminus of the peptide chain ends with a Fmoc protected amino acid. With the removal of the Fmoc group, head-to-tail cyclization and self-cleavage of the peptide chain synchronously occurred on BDKO (or DPBA) support to release the target cyclic peptide and the original support itself. They can be easily separated and purified, and the support can be recycled for reusing directly. The total synthesis of naturally occurring cyclic peptides such as Mortiamide A-E, Gramicidin S and cyclo(Phe-Pro) on BDKO (or DPBA) support in a resin-free manner was successfully demonstrated.

Abstract:A great hurdle in the production of synthetic polypeptides lies in the access of N -carboxyanhydrides (NCA) monomers, which requires dry solvents, Schlenk line/gloveboxe, and protection of side-chain functional groups. Here we report a robust method for preparing unprotected NCA monomers in air and under moisture. The method employs epoxy compounds as ultra-fast scavengers of hydrogen chloride to allow assisted ring-closure and prevent NCA from acid-catalyzed decomposition under moist conditions. The broad scope and functional group tolerance of the method are demonstrated by the facile synthesis of over 30 different α/β-amino acid NCAs, including many otherwise inaccessible compounds with reactive functional groups, at high yield, high purity, and up to decagram scales. The utility of the method and the unprotected NCAs is demonstrated by the facile synthesis of two water-soluble polypeptides that are promising candidates for drug delivery and protein modification. Overall, our strategy holds great potential for facilitating the synthesis of NCA and expanding the industrial application of synthetic polypeptides.

Abstract:A general and efficient method for the synthesis of azabicyclo[4,3,0]nonanone and azabicyclo[5,3,0]clecanone amino acid derivatives was developed with the palladium-catalyzed coupling of intermediates 9 and 10 with BocNH(2) and Boc(2)NH and the following reduction of the C-C double bond by hydrogenation as the key steps. The exploration of the synthesis of C-6-substituted dipeptide mimetics from 9 and 10 using Suzuki coupling as the key reaction has also been performed.

Abstract:3,4-Dihydroxyphenylalanine (DOPA) derivatives inspired by mussels have attracted broad interest in functional coatings of biomaterials and biomedical devices. Fmoc-DOPA(acetonide)-OH is the key precursor for solid-phase synthesis of adhesive mussel proteins and peptides. However, existing synthesis methods of Fmoc-DOPA(acetonide)-OH were tedious and costly which greatly hindered its practical application. Herein, a simple two-step strategy for preparing Fmoc-DOPA(acetonide)-OH is reported, which is a simple and cost-effective synthesis method with broad application prospects.

Abstract:N(alpha)-Methylamino acid containing peptides exhibit interesting therapeutic profiles and are increasingly recognized as potentially useful therapeutics. Unfortunately, their synthesis is hampered by the high price and unavaibility of many N(alpha)-methylamino acids. An efficient and practical preparation of N(alpha)-methyl-N(alpha)-(o-nitrobenzenesulfonyl)-alpha-amino acids without extensive purification is described. The procedure is based on the well-known N-alkylation of N(alpha)-arylsulfonylamino esters which was improved by using dimethyl sulfate and DBU as base. Ester cleavage is efficiently achieved by using an S(N)2-type saponification with lithium iodide, avoiding racemization observed with lithium hydroxide hydrolysis. Compatibility of the synthesized N(alpha)-methylamino acids with Fmoc solid-phase peptide synthesis is demonstrated by using normal coupling conditions to efficiently prepare N-methyl dipeptides. The described procedure allows the preparation of N(alpha)-methylamino acids in a very short period of time and a rapid synthesis of N-methyl peptides using Fmoc solid-phase peptide synthesis.

Abstract:Synthetic polypeptides, commonly prepared by the ring-opening polymerization (ROP) of amino acid N-carboxyanhydrides (NCA), are a family of biomimetic materials with vast biomedical applications. A great hurdle in the pro-duction of synthetic polypeptides is the synthesis of NCA, which requires ultra-dry solvents, Schlenk line/gloveboxes, and the protection of sidechain functional groups. Herein, we report a robust and scalable new method for the production of unpro-tected NCA monomers in air and under moisture. The method employs propylene oxide or epichlorohydrin as an inexpensive and ultra-fast scavenger of hydrogen chloride to prevent NCA from acid-catalyzed decomposition under moist conditions. The broad scope and outstanding functional group tolerance of the method are demonstrated by the successful synthesis of more than 30 different NCAs, including many otherwise inaccessible compounds with reactive functional groups (e.g. hy-droxyl, thiol, and carboxylic acid), at high yield and up to ten-gram scale. The scope of the method can be further extended to various α-hydroxyl acid O-carboxyanhydrides (OCA) and β-amino acid NCAs (βNCA). Given these merits, our strategy holds great potential for revolutionizing the synthesis of NCA and polypeptides, and dramatically expanding the industrial application of synthetic polypeptides

Abstract:Unnatural polar -aminoxy acid residue with proteingenous hydroxymethyl side chain, a building block of the peptidomimetic foldamer of -aminoxy peptide, was synthesized starting from natural amino acid L-serine. The starting material, L-serine, undergoes a reaction sequence to produce compound 1 in three steps: (1) the neighboring carboxyl group participates in diazotization/bromination to transform the amino group to a bromo group, (2) the C-terminal carboxyl group is protected, and (3) bromide is SN2-displaced by N-hydroxyl phthalimide to introduce a N-O bond. After several conventional deprotection/coupling reactions, compound 1 is easily transformed to an -aminoxy diamide, which can be widely used in peptidomimetics design.

Abstract:The use of pre-prepared diaminodiacids has been established as an effective approach for the chemical synthesis of peptide disulfide bond mimics. A technical problem often encountered in the implementation of the diaminodiacids strategy is the use of heavy metal reagents to remove the side-chain protecting groups. In the present work, we reported the development of diaminodiacid that carry 4-(N-[1-(4,4dimethy1-2,6-dioxocyclo-hexylidene)-3-methylbutyl]amino)benzyl (Dmab) and 1-(4,4-dimethyl-2,6dioxocyclohex-1-ylidene)-3-methylbutyl (ivDde) protecting groups. This pair of protecting groups can be readily removed by mild hydrazinolysis during the solid-phase synthesis on resin. We demonstrated the use of Dmab/ivDde protected diaminodiacids in the successful synthesis of a disulfide surrogate of oxytocin. (C) 2017 Published by Elsevier Ltd.

Abstract:Polypeptides have been widely utilized in the fields of biomaterials and biomedicine.Ever since N-carboxyanhydride (NCA) was reported by Hermann Leuchs in 1906,ring-opening polymerization of NCAs has been extensively used to prepare polypeptides.Despite continuous innovations,it is still challenging to synthesize polypeptides in high molecular weight efficiently.To address this challenge,we developed KHMDS/NaHMDS initiated fast NCA polymerization that is also moisture tolerant,open-flask amenable and terminal tunable.This NCA polymerization was able to proceed in most common solvents and meet the solubility requirement of variable NCA monomers and corresponding polypeptides.KHMDS can initiate γ-benzyl-L-glutamate-N-carboxyanhydride (BLG NCA) polymerization in a reaction rate 92 times faster than does hexylamine and 80 times faster than does triethylamine.This NCA polymerization also demonstrated easy and fast synthesis of gram-scale long chain polypeptides in an open flask.

Abstract:Both sulfonopeptides and phosphonopeptides are important analogs of naturally occurring peptides and have been widely used as enzyme inhibitors and haptens for producing catalytic antibodies due to their tetrahedrally structural features. A series of hybrid sulfonophosphinodipeptides composing of taurines and 1-aminoalkylphosphinic acids were first and conveniently synthesized in satisfactory to good yields via a Mannich-type reaction of N -benzyloxycarbonylaminoalkanesulfonamides, aldehydes, and aryldichlorophosphines, and subsequent hydrolysis. The current method provides an efficient and direct synthesis of hybrid sulfonophosphinodipeptides.

Abstract:A series of dipeptide derivatives containing non coded ammo, acids, N-Boc-4-X-Phe-Ala-NENHPh (X = Cl, Br, I, NO(2)), were synthesized by using thermoase in organic solvents. The physical data were consistent with the same samples prepared by 3-(diethoxyphosphoryloxy)-1, 2, 3-benzotriazin-4 (3H)-one (DEPBT). Influence of different substituted groups of the noncoded ammo acids and different organic solvents on the enzymatic peptide synthesis was studied.

Abstract:A simple and efficient preparation of gramicidin S and its analogues is described. It involves solid-phase peptide synthesis and on-resin macrolactamization without side chain protection, affording cyclic products in high yield and high purity. The high specificity of the cyclization reaction was shown to originate in the formation of a pre-organized conformation of the linear biosynthetic precursor of gramicidin S. This facile method will provide convenient access to the analogues of the natural product for functional optimization to counter microbial resistance.
New synthetic route for D-prolinamide

Abstract:Aim To establish a new method for the synthesis of D-prolinamide.Method D-proline was obtained from the asymmetric transformafion of L-proline via the formation of(S)-tartaric acid salt by using butanal as catalyst.Then,D-prolinamide was synthesized by amino protection,esterification,amidation and deprotection.Results and conclusion The yield of final product was 78.4% and optical purity was 99.3%.Comparing with patent synthetic procedure for D-prolinamide,it is a new synthetic procedure with better yield and purity and suitable for industrial manufacture.

It was found that many problems occurred preparing amino acid derived N-acylsulfonamides with the commonly used coupling approach in our previous studies. In this paper, we report an efficient approach to synthesize various amino acids derived N-acylsulfonamides in high yields without any racemization.

Abstract:N alpha-Dithiasuccinoyl (Dts) amino acids needed for solid-phase peptide synthesis have been prepared in good yields and excellent purities by a new method that exploits the solubility properties of polyethylene glycol (PEG; bifunctional with average molecular weight 2000 was found to be optimal). Suitably side-chain protected amino acid derivatives are first reacted with a polymeric xanthate, following which the free alpha-carboxyl is blocked by silylation and the Dts heterocycle is elaborated in the same pot by reaction with chlorocarbonylsulfenyl chloride. Upon aqueous work-up, the polymeric carrier removes any urethane blocked amino acids which arise during the process. Exaggerated conditions were explored to prove the power of this functional purification approach, and mechanisms of formation of polymer-bound urethanes are proposed and supported by solution model studies. The preparation and characterization of the companion N-(iso-propyldithio)carbonyl derivative of proline is also presented.
