Abstract:An improved sparse representation algorithm is proposed for reducing speckle noise in optical coherence tomography (OCT) images. To reduce the dictionary training time, the orthogonal matching pursuit algorithm (OMP) is replaced by a piecewise orthogonal matching pursuit algorithm (ST-OMP), and a threshold standard is set to train multiple atoms simultaneously to improve the tracking efficiency. Normal, diabetic macular edema, and fundus neovascularization membrane retina OCT images were tested using the proposed algorithm and other denoising algorithms. The results show that the proposed algorithm exhibits an excellent denoising ability. The details and edge information in the OCT images were well preserved by the proposed algorithm. Meanwhile, the image processing time was reduced by half with the proposed algorithm compared with the traditional K-SVD algorithm.
Enhancing Sensitivity of High Resolution Optical Coherence Tomography Using an Optional Spectrally Encoded Extended Source (conference Presentation)

Abstract:High-resolution optical coherence tomography (OCT) is of critical importance to disease diagnosis because it is capable of providing detailed microstructural information of the biological tissues. However, a compromise usually has to be made between its spatial resolutions and sensitivity due to the suboptimal spectral response of the system components, such as the linear camera, the dispersion grating, and the focusing lenses, etc. In this study, we demonstrate an OCT system that achieves both high spatial resolutions and enhanced sensitivity through utilizing a spectrally encoded source. The system achieves a lateral resolution of 3.1 μm and an axial resolution of 2.3 μm in air; when with a simple dispersive prism placed in the infinity space of the sample arm optics, the illumination beam on the sample is transformed into a line source with a visual angle of 10.3 mrad. Such an extended source technique allows a ~4 times larger maximum permissible exposure (MPE) than its point source counterpart, which thus improves the system sensitivity by ~6dB. In addition, the dispersive prism can be conveniently switched to a reflector. Such flexibility helps increase the penetration depth of the system without increasing the complexity of the current point source devices. We conducted experiments to characterize the system’s imaging capability using the human fingertip in vivo and the swine eye optic never disc ex vivo. The higher penetration depth of such a system over the conventional point source OCT system is also demonstrated in these two tissues.
Contrast Enhancement of Spectral Domain Optical Coherence Tomography Using Spectrum Correction

Abstract:In this paper, we propose spectrum correction method to enhance image contrast of spectral domain optical coherence tomography (SD-OCT). The proposed method firstly reshapes the light source to a Hann-shape spectral light by digital processing and then strengthens the coherence numerical value in the main lobe of the axial point spread function(PSF). Experimental results acquired from fresh rat corneas and fixed human aortic atherosclerosis tissues show that the proposed method improves image definition and clarity, and consequently provides better detailed microstructural information than the existing method. This ability of improving image quality is obtained without any hardware change, making our method a cost-effective alternative to competing with hardware approaches.

Abstract:The depth reflectivity profile of Fourier domain optical coherence tomography (FD-OCT) is estimated from the inverse Fourier transform of the spectral interference signals (interferograms). As a result, the axial resolution is fundamentally limited by the coherence length of the light source. We demonstrate that using the autoregressive spectral estimation technique instead of the inverse Fourier transform, to analyze the spectral interferograms can improve the axial resolution. We name this method spectral estimation OCT (SE-OCT). SE-OCT breaks the coherence length limitation and improves the axial resolution by a factor of up to 4.7 compared with FD-OCT. Furthermore, SE-OCT provides complete sidelobe suppression in the depth point-spread function, further improving the image quality. We demonstrate that these technical advances enables clear identification of corneal endothelium anatomical details ex vivo that cannot be identified using the corresponding FD-OCT. Given that SE-OCT can be implemented in the FD-OCT devices without any hardware changes, the new capabilities provided by SE-OCT are likely to offer immediate improvements to the diagnosis and management of diseases based on OCT imaging. Abstract:Deconvolution methods are not feasible in spectral domain optical coherence tomography (SD-OCT) system due to the linear space variant property of its axial point spread function (PSF). In order to improve the axial resolution and enhance the imaging quality of the SD-OCT system, a Lucy-Richardson algorithm based deconvolution method is implemented with the numerically compensated A-scan signals and the effective axial PSF in this paper. The main factors decreasing the value of PSF along the axial direction and blurring the images, as well as the deconvolution process, are theoretical studied. With the developed SD-OCT system, the axial modulation function of the PSF is retrieved by curve fitting of the measured peak values of the PSF at different axial positions. After compensating the A-scan signal with the modulation function, deconvolution is performed with the measured effective axial PSF. In vivo imaging on typical samples is conducted and reconstruction is done based on the deconvolution method. The comparing results demonstrate that the proposed deconvolution method not only enhances the axial resolution but also improves the depth dependent sensitivity fall-off in SD-OCT system.
Contrast Improvement for Swept Source Optical Coherence Tomography Image of Sub-Surface Tissue

Abstract:Swept source optical coherence tomography (SSOCT) is an attractive biological imaging technology due to its advantages of simple setup and high imaging speed. As the light intensity attenuated rapidly in high scattering biological tissues, the contrast of OCT image will drop with depth. In this paper a new method was introduced to compensate the attenuation of imaging contrast in SSOCT. The interference signal was divided into two channels of analog to digital converter (ADC) with a splitting ratio of 1:5. The higher level signal in one channel was used to reconstruct deeper structure of tissue and the lower level signal in the other channel was used to reconstruct surface structure of tissue. Low-frequency signals in one channel were filtered by a high pass filter and then combined with the signal in the other channel to obtain a high contrast image in both surface and deep area of tissue. Human finger and porcine airway imaging obtained with the system show that the contrast of SSOCT images can be improved in deeper region of tissue.

Abstract:An inherent compromise must be made between transverse resolution and depth of focus (DOF) in spectral domain optical coherence tomography (SD-OCT). Thus far, OCT has not been capable of providing a sufficient DOF to stably acquire cellular-resolution images. We previously reported a novel technique named multiple aperture synthesis (MAS) to extend the DOF in high-resolution OCT [Optica4, 701 (2017)]. In this technique, the illumination beam is scanned across the objective lens pupil plane by being steered at the pinhole using a custom-made microcylindrical lens. Images captured via multiple distinctive apertures were digitally refocused, which is similar to synthetic aperture radar. In this study, we applied this technique for the first time to image both a homemade microparticle sample and biological tissue. The results demonstrated the feasibility and efficacy of high-resolution biological tissue imaging with a dramatic DOF extension.

Abstract:We developed spectral domain optical coherence tomography using a dual-channel spectrometer for complex conjugate artifacts (CCA) suppression. We used a three-line charge coupled device to simultaneously detect two interferometric spectra with 2 pi/3 phase difference. The complex interferometric signal was reconstructed by trigonometric manipulation of two real interferometric spectra, and then full-range images were obtained by use of inverse Fourier transform. Artifacts at direct current (DC) and the ghost remnant of the CCA are common issues with the previously reported two-spectrometer method because the slight mismatching between two spectral detection channels had strong negative effects on CCA suppression and appeared to be the limiting factor on system performance. This novel dual-channel spectrometer uses the same spectrometer optics for the two spectral detection channels and, therefore, achieves better matching between two spectral detection channels and consequently better performance in CCA suppression as compared with the dual spectrometer solution. Fullrange imaging with CCA suppression up to similar to 25 dB was demonstrated when imaging an attenuated reflector. The efficacy of both CCA and DC suppressions also was validated by imaging the anterior segment of a rat eye ex vivo. The quality of CCA-suppressed images was significantly improved with regard to those obtained with the dual-spectrometer design. (C) 2017 Optical Society of America
Dynamic Optical Clearing Effect of Tissue Impregnated with Hyperosmotic Agents and Studied with Optical Coherence Tomography

Abstract:The depth of light penetration into highly scattering tissues can be improved by the application of biocompatible and osmotically active chemical agents. We compare the dynamics of optical clearing of tissue by the topical application of glycerol and dimethyl sulfoxide (DMSO) using optical coherence tomography (OCT). It is demonstrated experimentally that both agents can largely improve the OCT imaging depth for porcine stomach tissue. During a period of approximately 20 to 30 min after the application of glycerol image contrast is also enhanced. This enhancement disappears over time. Such enhancement of image contrast is not observed with DMSO. Glycerol causes a higher degree of dehydration of the tissue than DMSO does. We suggest that these phenomena are caused by a two-stage diffusion of the chemicals. The first stage of diffusion is from the top tissue to the intercellular space, and the second is into the cell matrix. During the first stage, the imaging contrast could be improved by dehydration.

Abstract:The imaging penetration depth of an optical coherence tomography (OCT) system is limited by the dynamic range of the system. In a common case that signals exceed the dynamic range of a Fourier domain OCT (FDOCT) system, saturation artifacts degrade the image quality. In this chapter, we demonstrate some new costeffective techniques to improve the dynamic range of a swept-source OCT (SSOCT) system. For example, one method is based on a dual-channel detection technique to enhance the dynamic range by reconstructing the saturated signals due to strong reflection of the sample surface. Another method utilizes a tunable high-pass filter to compensate the attenuation of light signal in deep tissue. It was demonstrated that these techniques can improve the dynamic range of an SSOCT system by more than 10 dB with a low bit-depth analog-to-digital converter.

Abstract:SIGNIFICANCE:Optical coherence tomography (OCT) has been widely used in clinical studies. However, the image quality of OCT decreases with increasing imaging depth since the light is rapidly attenuated in biological tissues.AIM:We present a compensation approach to preserve weak high-frequency signals from deep structures and compress the dynamic range of the detected signal for superior analog-to-digital conversion and image display capability.APPROACH:A homemade frequency gain compensation amplifier is designed and fabricated to amplify the electrical signal from a balanced photodetector and compensate for the signal attenuation in swept-source OCT (SSOCT).RESULTS:It is demonstrated in imaging various objects that this cost-efficient technique effectively enhances the contrast of the deep tissue image.CONCLUSIONS:A frequency gain compensation amplifier is designed and used to compress the dynamic range of the electrical signal detected by the photodetector of an SSOCT system, which enables weak signals from deep structures to be acquired by the ADC and displayed with enhanced local contrast.

Abstract:In this work, we used a short coherence digital holographic microscopy system to demonstrate cross-talk noise suppression and imaging performance enhancement by optical clearing. Performance of the system on both phantom and in vitro porcine skin tissues before and after the treatment of 70% v./v. glycerol-saline solution was investigated. Our results showed that optical clearing effectively inhibits the cross-talk noise and improves the image quality in the deep of the in vitro porcine skin tissues. The imaging depth was increased by about 30% after topical application of the glycerol-saline solution for 30 min.

Abstract:Optical clearing can effectively control the optical properties of in tissue using the optical immersing technique by hyperosmotic agents and be paid more and more attention in the biomedical and optical field.This mothed could reduce the number of the multiple scattered photons,increase the depth of the optical imaging,and get a better contrast grade of images.We measured the change of the optical properties of the tissue of the different depth during the optical clearing process using optical coherence tomography(OCT).We applied the gradient of the OCT signal algorithm to analyze the temporal and spatial distribution of the scattering coefficient during the clearing process.It was demonstrated experimentally that optical clearing could effectively improve the OCT imaging depth.During the process of the optical clearing,the scattering coefficient,the range and the time and the trend of the change of the scattering coefficient changed with the different depth,which was related to the tissue microstructure,the clearing effect due to the microstructure and the diffuse process of the agent.The results could be used to understand the process and the mechanism of the optical clearing.

Abstract:A deconvolution method for image enhancement suitable for spectral domain optical coherence tomography (SD-OCT) is proposed. The method includes Wiener filtering for spectral deconvolution and successive homomorphic filtering for spatial deconvolution, aiming to minimize blurring effect of axial point spread function (PSF) and its depth dependence. Tunable factor in Wiener filtering is optimized for tradeoff between compensation ratio and signal-to-noise ratio (SNR). In vivo OCT imaging of a fresh shrimp is done and image reconstructions are performed. Comparing results demonstrates the proposed method suppresses the coherent noises in OCT image while compensating sensitivity fall-off with increased averaged SNR and recovers structural information at deep depths. Contrast-to-noise ratio (CNR) and averaged SNR in the reconstructed image based on the proposed method in contrast to those without deconvolution are improved by 3.4 dB and 4.6 dB, respectively. Abstract:The growing need to understand the molecular mechanisms of diseases has prompted the revolution in molecular imaging techniques along with nanomedicine development. Conventional optical coherence tomography (OCT) is a low-cost in vivo imaging modality that provides unique high spatial and temporal resolution anatomic images but little molecular information. However, given the widespread adoption of OCT in research and clinical practice, its robust molecular imaging extensions are strongly desired to combine with anatomical images. A range of relevant approaches has been reported already. In this article, we review the recent advances of molecular contrast OCT imaging techniques, the corresponding contrast agents, especially the nanoparticle-based ones, and their applications. We also summarize the properties, design criteria, merit, and demerit of those contrast agents. In the end, the prospects and challenges for further research and development in this field are outlined.
Feasibility and Safety of Very-Low Contrast Combined Ringer's Solution in Optical Coherence Tomography Imaging

Abstract:Background Optical coherence tomography (OCT) is an important modality used in coronary intervention. However, OCT requires a high amount of contrast media, limiting its extensive application in clinical practice. This study compared OCT images of coronary lesions obtained using contrast media and very-low contrast combined Ringer's solution (VLCCR) in patients with acute coronary syndrome (ACS). Methods Thirty ACS patients with a total of 36 native lesions and stenoses from 70 to 90% were included in this study. Two kinds of flushing media (a contrast medium and VLCCR) were used in succession in a random order for OCT image pullback of each lesion. VLCCR method is using low volume contrast (4–5 ml) injected into the guiding catheter previously combination with injector infused Ringer's solution instead of pure contrast medium. The safety of procedure was evaluated by recording the patients ‘symptoms, changes of ECG, blood pressure and heart rate. OCT images were analyzed to determine the image clarity. Lumen area and diameter were also measured and the consistency between the two media was compared. Results OCT procedure using either contrast or VLCCR did not show any peri-procedural adverse events. There was no difference in changes of blood pressure and heart rate in both procedures, however, VLCCR procedure showed less procedure-related symptoms and ECG changes. We found that the percentage of clear image frame was equivalent between the contrast and VLCCR media (98.0 vs. 96.9%, P = 0.90). We also observed a high degree of similarity between the different lesion phenotypes of ACS for both media. There was a linear correlation of the phenotypes obtained with these two different methods, and a significant correlation was observed between measurements obtained with contrast and VLCCR without correction for the refractive index of VLCCR (correlation coefficients ranged between 0.829 and 0.948). Conclusions OCT imaging using VLCCR for blood clearance is feasible and safe and provides similar imaging quality compared to OCT imaging obtained using radiographic contrast media for ACS patients.

Abstract:Utilizing the ultra-fast laser or superluminescence diode (SLD) as the light source, optical coherence tomography (OCT) is a new tomographic imaging technique. It has great applying foreground in many fields such as medicine, biology, material and so on. We founded an OCT system and applied it in biological imaging successfully. Furthermore, we found a potential great application for this technique in measuring the inner structure of bedded samples such as paint films.The experiment in OCT has made prompt development, while only a little work has been done an the theoretical analysis. Some experimental results, thus, can't be explained clearly. In this paper, we use the Monte-Carlo simulation method to analyze OCT image. The differences between single scattered (or least scattered) light and multiply scattered light and its influence to the quality of OCT image are discussed in detail.To show an example, our simulating calculation object is a conduit immersing in a high scattering medium that can be regarded as the approximation of blood vessel. After programming and calculating we get the theoretical result as the form of pseudo color image. The images of conduit immersing in the high scattering medium are clearly seen. Comparing with the experimental imaging of vein they are accordant basically.All above prove the Monte-Carlo simulation method correct and effective in theoretical research of OCT. Many significant results, which are very helpful for experimental research, are draw through this method.
Imaging depth adaptive resolution enhancement for optical coherence tomography via deep neural network with external attention

Abstract:Abstract Optical coherence tomography (OCT) is a promising non-invasive imaging technique that owns many biomedical applications. In this paper, a deep neural network is proposed for enhancing the spatial resolution of OCT en face images. Different from the previous reports, the proposed can recover high-resolution en face images from low-resolution en face images at arbitrary imaging depth. This kind of imaging depth adaptive resolution enhancement is achieved through an external attention mechanism, which takes advantage of morphological similarity between the arbitrary-depth and full-depth en face images. Firstly, the deep feature maps are extracted by a feature extraction network from the arbitrary-depth and full-depth en face images. Secondly, the morphological similarity between the deep feature maps is extracted and utilized to emphasize the features strongly correlated to the vessel structures by using the external attention network. Finally, the SR image is recovered from the enhanced feature map through an up-sampling network. The proposed network is tested on a clinical skin OCT data set and an open-access retinal OCT dataset. The results show that the proposed external attention mechanism can suppress invalid features and enhance significant features in our tasks. For all tests, the proposed SR network outperformed the traditional image interpolation method, e.g. bi-cubic method, and the state-of-the-art image super-resolution networks, e.g. enhanced deep super-resolution network, residual channel attention network, and second-order attention network.
