Abstract:Physical characterization of formulations in frozen and freeze-dried solid states provides indispensable information for rational development of freeze-dried pharmaceutical products. This article provides an overview of the physical characteristics of formulations in frozen and freeze-dried solid states, which are essential to both formulation and process development. Along with a brief description of techniques often used in physical characterization for freeze-drying development, applications of and recent improvements to these techniques are discussed. While most of these techniques are used conventionally in physical characterization of pharmaceuticals, some techniques were designed or modified specifically for studies in freeze-drying. These include freeze-drying microscopy, freeze-drying X-ray powder diffractometry and cryoenvironmental scanning microscopy, which can be used to characterize the physical properties of the formulation under conditions similar to the real vial lyophilization process. Novel applications of some conventional techniques, such as microcalorimetry and near infrared (NIR) spectroscopy, which facilitated freeze-drying development, receive special attention. Research and developmental needs in the area of physical characterization for freeze-drying are also addressed, particularly the need for a better understanding of the quantitative correlation between the molecular mobility and the storage stability (shelf life).

Abstract:Freeze-drying (FD) is the most substantial drying technique utilized in the pharmaceutical and biopharmaceutical industries. It is a drying process where the solvent is crystallized at low temperatures and then sublimed from the solid-state directly into the vapor phase. Although FD possesses several merits as its suitability for thermolabile materials and its ability to produce dry products with high-quality attributes, it is a complex and prolonged process that requires optimization of both; process and formulation variables. This review attains to disassemble FD complications through a detailed explanation of the lyophilization concept, stages, the factors influencing the process including controlled ice nucleation, and the modified and innovative FD technologies proposed in recent years to overcome the shortage of traditional FD. In addition, this work points out the quality by design (QbD), critical quality of attributes (CQAs), limitations, and drawbacks of lyophilization. Lyophilization is a propitious drying technique for thermolabile materials. Optimizing the lyophilization cycle requires controlling the process parameters. Abstract:Abstract Background Process intensification is a major hurdle in pharmaceutical process scale-up. Solvent removal strategies have limited the effectiveness of the overall stability of pharmaceutical formulations. The main aim of present review article is to focus on the use of the freeze-drying process in pharmaceuticals, biopharmaceuticals and nanoderived therapeutics and their translation into commercial viable products. Unwavering efforts of scientists in the process intensification of lyophilization promote unique features of products for commercialization. Regulatory agencies are promoting the utilization of a quality-by-design approach to improve product characteristics. Among 300 FDA-approved pharmaceutical industries, 50% of products are freeze-dried. The freeze-drying process is costlier and requires more time than other drying methodologies. Unstable pharmaceutical dispersions and solutions can be preferably stabilized by using the freeze-drying method. Main text This review highlights the utilization of critical quality attributes and process parameters for the freeze-drying process, which helps to improve the integrity and stability of the formulation. The quality-by-design approach possibly cuts the cost of the process and saves money, time, and laborious work. The present review focuses preliminarily on the applications of freeze-drying in the development of biopharmaceuticals, including vaccines, proteins and peptides, and injectable products. In addition, a separate section demonstrating the potential of freeze-drying in nanoderived therapeutics has been illustrated briefly. The present clinical scenario of freeze-dried pharmaceuticals and biopharmaceuticals has also been described in later sections of the review. Conclusions This review underscores the value of integrating Quality by Design into the development of lyophilization processes for pharmaceutical and biopharmaceutical products. By identifying critical process parameters, delineating a design space, and leveraging advanced monitoring techniques, manufacturers can effectively address the intricacies of lyophilization. Abstract:Freeze-drying, also known as lyophilization, is a process in which water in the form of ice under low pressure is removed from a material by sublimation. This process has found many applications for the production of high quality food and pharmaceuticals. The main steps of the freeze-drying process, such as the freezing of the product and primary and secondary drying, are described in this paper. The problems and mechanisms of each step of the freeze-drying process are also analyzed. The methods necessary for the selection of the primary and secondary end processes are characterized. The review contains a description of the effects of process conditions and the selected physical properties of freeze-dried materials, such as structural properties (shrinkage and density porosity), color, and texture. The study shows that little attention is given to the mechanical properties and texture of freeze-dried materials obtained from different conditions of the lyophilization process.

Tray style freeze dryer helps in development of long-term storage vaccines and labile drugs. It performs freeze drying in containers or in vials on large quantities. This makes tray style freeze dryer become most common for developing lyophilized product. The need for improve the shelf-life of protein-based drug and antibiotics formulations makes tremendous growth of freeze-dried product market. This review focuses on the lyophilization process, design of freeze dryer, role of excipients in lyophilization process, decision of selecting the excipients, importance of cake structure helps to develop stable lyophilized drug product for long-term usage and applications of lyophilization technology.
Preliminary study on the effect of pre-freezing methods on lyophilization quality and storage stability of probiotics

Abstract:This study investigated the impact of pre-freezing techniques on the quality and storage stability of probiotics. Pediococcus acidilactici CCFM6432, Lactiplantibacillus plantarum CN2018, and Bifidobacterium breve CCFM1025 were subjected to freezing at −80 °C and −196 °C using liquid nitrogen, respectively. The results indicated that liquid nitrogen pre-freezing did not cause cell death during the freezing phase. Following lyophilization, liquid nitrogen pre-freezing did not affect the survival rates of P. acidilactici CCFM6432 and L. plantarum CN2018; however, it significantly decreased the survival rate of B. breve CCFM1025 ( p < 0.05). Differential Scanning Calorimetry (DSC) analysis revealed that liquid nitrogen pre-freezing increased the melting enthalpy of the cell suspension. A linear correlation between the bacterial cell survival rate and moisture content was observed, with survival rates at the pre-freezing temperature of −196 °C slightly lower than those at −80 °C. Liquid nitrogen granulation pre-freezing enhanced the storage stability of B. breve CCFM1025. This research offers valuable insights into the potential development of pre-freezing techniques for application in the food industry to enhance the quality and storage stability of probiotics.

Abstract:Freeze-drying is commonly used to increase the shelf-life of pharmaceuticals and biopharmaceuticals. Freezing represents a crucial phase in the freeze-drying process, as it determines both cycle efficiency and product quality. For this reason, different strategies have been developed to allow for a better control of freezing, among them, the so-called vacuum-induced surface freezing (VISF), which makes it possible to trigger nucleation at the same time in all the vials being processed. We studied the effect of different vial types, characterized by the presence of hydrophilic (sulfate treatment) or hydrophobic (siliconization and TopLyo Si-O-C-H layer) inner coatings, on the application of VISF. We observed that hydrophobic coatings promoted boiling and blow-up phenomena, resulting in unacceptable aesthetic defects in the final product. In contrast, hydrophilic coatings increased the risk of fogging (i.e., the undesired creeping of the product upward along the inner vial surface). We also found that the addition of a surfactant (Tween 80) to the formulation suppressed boiling in hydrophobic-coated vials, but it enhanced the formation of bubbles. This undesired bubbling events induced by the surfactant could, however, be eliminated by a degassing step prior to the application of VISF. Overall, the combination of degasification and surfactant addition seems to be a promising strategy for the successful induction of nucleation by VISF in hydrophobic vials.

Abstract:The control of freezing is crucial as it can influence the freeze-drying operation as well as the chemical and physical properties of the lyophilized product such as the specific surface area, dissolution rate, and drug stability. In this review article, we focus on two areas related to lyophilization in the pharmaceutical industry: (1) advances in our understanding of the relationship between the freezing conditions, product morphology and drying efficiency, and (2) the control of nucleation in solutions to be frozen. The final objective is to accelerate the transition toward a more robust design of freeze-drying cycles, based on deep knowledge of the phenomena involved rather than on empirical observations. Abstract:Background In recent years, as the beneficial effects of probiotics have been gradually discovered, the use of drying methods to stabilize probiotic ingredients has been more deeply applied to food, medicine, and other fields. The probiotic powder obtained by freeze-drying can effectively maintain the viability of probiotics and has good fermentation performance. Scope and approach Freeze-drying of probiotics allows the strain to maintain its original chemical composition and physical properties, and the energy consumption is lower than other drying methods. This is also a promising way for long-term storage of probiotics and satisfies more possible fermentation applications. Key findings and conclusions This review mainly introduces the effect of freeze-drying process on probiotics and proposes some key strategies that can effectively improve the viability of probiotics after freeze-drying, such as changing the culture conditions and media components of the strain, adding cryoprotectants, etc. The future application of freeze-drying to prepare probiotic microbe powder is also prospected.

Abstract:Lyophilization is a pivotal manufacturing process to obtain a stable drug product that is unstable as a ready-to-use formulation. Some formulations may require the addition of drug-specific excipients such as stabilizers, buffers, and bulking agents to support the cake appearance and ensure long-term stability of the drug product. Optimization of the lyophilization process parameters at each stage including freezing and primary and secondary drying is important because these parameters can have a direct impact on the process efficiency (shortened cycle time) and product performance (cake appearance and homogeneous moisture content). Several parameters of the formulation, including properties of the active pharmaceutical ingredient, excipients, solvent system, and container closure, determine the success of lyophilization. Development, scale-up, and transfer of the lyophilization cycle are challenging; hence, a comprehensive understanding of the critical parameters related to the formulation, lyophilization process, and lyophilizer design allows designing a quality drug product. One approach for a successful transfer of the lyophilization cycle between the laboratory and commercial-scale lyophilizer is using vial heat transfer coefficient and ice slab test to establish a maximum sublimation rate. This review provides a general overview of the lyophilization process and discusses several key considerations and product development aspects of formulation, process optimization, container closure system, scale-up principles, and drug product quality attributes from the industrial viewpoint. Abstract:Cell-free protein synthesis system is emerging as a powerful tool for rapid expression of therapeutic proteins. However, one drawback of cell-free technology is the necessity to store the major components below freezing in bulky aqueous solutions. To preserve the cell-free synthesis system for a longer time, the lyophilized cell-free system was developed, and its stability under freeze-drying conditions was explored. The novelty and the difference of this study from the others lie in determining more detailed storage conditions, which could provide a more accurate storage strategy. The effects of the treatment time, the storage temperature, and the component-mixing mode on the freeze-dried cell-free system were investigated. This study explored a robust storage strategy for freeze-dried cell-free systems and could further open up a better opportunity for on-demand synthesis of therapeutic proteins.

Abstract:Quality control still remains a big issue, affecting herbs, formulations, and even the practice of Traditional Chinese Medicine. A promising solution to the problem is to take advantage of innovative process techniques including alternative drying methods within the pharmaceutical processing. Nowadays, vacuum belt drying (VBD) has gained more and more importance in food and pharmaceutical processing. In this work, Panax notoginseng extract was made into powder by the gentle VBD as well as by classical spray drying (SD), freeze drying (FD), and vacuum drying (VD). Certain chemical and physical properties of the dried powders such as moisture content, drying time, porosity, density, saponins content, pH value and hydroxyl radicals scavenging activity were evaluated. The results showed the qualities of dried products were influenced to different extent by drying methods certainly. Vacuum belt drying was suggested to dry P. notoginseng extract for its high quality assurance of hygroscopic products and suitable for sticky or high viscous products.

Suitable parameters of process application allow us to obtain best quality products compared to products dried with traditional methods. Very good physical and chemical properties of food and biotechnological products make this method the best for drying exclusive products. On the domestic market there is a large selection of different types of freeze-dried products, and there is still increasing interest of consumers in these products. A high cost of the freeze-drying still limits the wide-scale application in the food industry. Equipment innovation and pretreatment of raw material can reduce the time and energy needed for this process.

This paper will explore the characteristics of freeze-drying and its possibilities for developing stable drugs in more detail. Especially complex protein-based drugs present significant challenges to manufacturers because their stability is low, and they often have harsh conditions that need to be stored such as being kept frozen or cooled on ice at all times.

Abstract:Vacuum freeze-drying technology is the best way to preserve the quality of food at present, which can maintain the nutrition and flavor of food to the maximum extent. It has the characteristics of being green, convenient, and nutritious. Abstract:Freeze-drying is usually applied to dry bioproducts, pharmaceuticals, and high-valued foodstuffs. Due to high running cost and energy consumption of conventional freeze dryers, wider applications of freeze-drying technology are restricted. A freeze dryer based on a closed regeneration loop of hygroscopic solution is proposed here. Hygroscopic solutions such as LiCl-water solution are used as working fluid to absorb the vapor sublimated from the stuffs. The low vapor pressure of hygroscopic solution leads to a higher operating temperature of water vapor absorber compared with the operating temperature of traditional vapor condenser. Thermodynamic analyses are conducted to illustrate the advantages of this new freeze dryer. A case study performed with coffee shows a coefficient of performance improvement of 25.1% of the refrigeration system and a reduction of 14.6% of energy consumption, in which 8.0% cut of heat consumption is obtained because of the reuse of regeneration heat. Parametric analyses on the impacts of evaporation temperature and condensing temperature are also performed. It is demonstrated that the new freeze dryer has high potential of energy saving in wide operation ranges. The security issue of using hygroscopic solution in food freeze-drying has also been discussed. Considering other benefits altogether, it is believed that such a freeze dryer is promising in practical freeze-drying of foodstuffs.

Abstract:Drying is an indispensable operation in the preparation of pharmaceutical powders and always remained one of the energetic tasks in the pharmaceutical industry. Improving the stability, solubility, and dissolution of pharmaceutical products are being prime objectives of the drying process, intending to produce the products loving the dry state. Although there are voluminous literatures available concerning drying operations, there is scant information available regarding the applicability of drying in drug delivery and process scale-up. The current communication embodies the different particle engineering technologies of drying viz. spray-, freeze-, and spray-freeze drying. In addition, potential uses of drying in the taste masking, and the development of inhalable powders presented briefly. Recent advancements in the drying of novel drug delivery systems is the major focus of the present review. In our opinion, the commercial aspects, regulatory guidelines, and scale-up strategies presented herein provide an opportunity to readers, researchers, and industrialists to ruin the critical issues during drying operations and aid in developing quality pharmaceutical technologies.

Abstract:Most biopharmaceuticals are developed in liquid dosage forms that are less stable than solid forms. To ensure the stability of biopharmaceuticals, it is critical to use an effective drying technique in the presence of an appropriate stabilizing excipient. Various drying techniques are available for this purpose, such as freeze drying or lyophilization, spray drying, spray freeze-drying, supercritical fluid drying, particle replication in nonwetting templates, and fluidized bed drying.
