Abstract:A tetrasaccharide, α-l-Rhap-(1→3)-β-d-GalpNAc-(1→6)-α-d-Galp-(1→2)-α-d-Galp, the carbohydrate moiety of clarhamnoside isolated from the marine sponge Agelas clathrodes, was synthesized as its propyl glycoside via a convergent approach. The key steps to the synthetic strategy were the stereoselective construction of the reducing-end disaccharide α-d-Galp-(1→2)-d-Galp (5) and efficient coupling with the terminal disaccharide α-l-Rhap-(1→3)-d-GalpNAc building block, in which the N-phthalimido-protected trifluoroacetimidate 13 was proved to be an effective donor.

Abstract:Three new polyunsaturated lipids, (6Z,9Z,12Z,15Z)-octadeca-6,9,12,15-tetraen-3-one (1), (6Z,9Z,12Z,15Z)-1-bromooctadeca-6,9,12,15-tetraen-3-one (2), and (Z)-ethyl docos-5-enoate (3), together with two known polyunsaturated lipids, 4(Z),7(Z),10(Z)-tridecatrienoic acid (4) and (6Z,9Z,12Z,15Z)-octadeca-1,6,9,12,15-pentaen-3-one (5), were isolated from the marine sponge Haliclona sp., which was collected from Guangxi, using HSCCC and HPLC methods. Chemical structures of the five compounds were elucidated by spectroscopic techniques.

This marine small molecule inhibitor (IC(50)=20 microM) targeting type III secretory pathway of bacterial pathogenesis was assembled in good yield via a '2+2+1' strategy based on stereocontrolled construction of the four glycosidic linkages.

Abstract:Marine sponges are an excellent source of biologically active secondary metabolites. We focus on deep-sea sponges for our discovery study. A marine sponge Cladocroce sp. exhibited cytotoxic activity in the bioactivity screening. From this sponge a previously unreported cytotoxic glycosphingolipid, calyxoside B, was isolated and the structure of this compound was elucidated by analyses of MS and NMR spectra and chemical derivatization. We converted the ketone in the middle of a long aliphatic chain into an oxime to which was applied Beckmann rearrangement to afford two positional isomers of amides. The products were subjected to acidic hydrolysis followed by LC-MS analysis, permitting us to assign unequivocally the position of the ketone. Calyxoside B shows cytotoxicity against HeLa cells with an IC50 value of 31 µM and also weakly stimulated the production of cytokines in mice.

Abstract:A convergent synthesis of the dodecasaccharide scaffold of axinelloside A was achieved through Au(I)‐catalyzed [6+6] glycosylation. The initially devised [3+1+2] assembly of the nonreducing hexasaccharide fragment was low‐yielding, whereas a convergent [3+3] glycosylation under Au(I) catalysis was proven feasible, allowing for a semi‐gram scale preparation of the wanted hexasaccharide. The requisite 1,2‐cis glycosidic bonds were forged in a highly stereoselective fashion by virtue of remote acetyl group participation, and judicious manipulation of protecting groups. The synthetic dodecasaccharide has been properly protected for the downstream elaboration toward its natural form.

Abstract:A detailed chemical investigation of two specimen of South China Sea sponges Halichondria sp. (No. 19-XD-47 and No. 17-XD-87) have resulted in the isolation of three new sterols, namely, halichsterols A (1), B (2) and C (3), together with seven related known ones (4–10). Their structures were determined by extensive spectroscopic analysis and by comparison with the spectral data reported in the literature. In bioassay, compound 2 displayed significantly anti-inflammatory activity in lipopolysaccharide (LPS)-stimulated BV-2 microglial cells.

Abstract:A new steroid, 24 xi, 25-dimethyl-3 alpha-hydroxyl-cholest-5-ene-2 beta-ol sodium sulfate (1), together with a known steroid, 24 xi, 25-dimethyl-cholest-5-ene-2 beta, 3 alpha-diol disodium sulfate (2), was isolated from the ethanol extract of marine sponge Halichondria rugosa. Their structures were elucidated on the base of spectroscopic analysis. Both compounds showed cytotoxicity to four human cancer cell lines (BEL-7402, HT-29, SPC-A1 and U-251) with IC50 values between 6.5 and 23.1 mu M.

Abstract:Objective To study the chemical constituents of the sponge Haliclona cymaeformis collected from the South China Sea.Methods The compounds were isolated and purified by column chromatography on silica gel,reversed-phase silica gel(ODS) and Sephadex LH-20,and their structures were elucidated on the basis of physicochemical and spectral analysis.Results Eleven compounds were isolated from the ethanol extract of H.cymaeformis,and their structures were identified as 24-methylene-cholesta-3β-ol(1),5α,8β-epidioxy-cholesta-6-en-3-ol(2),cholesta-7-en-3β,5α,6β,9α-tetraol(3),cholesterol(4),1H-indole-3-carboxylic acid(5),uracil(6),thymine(7),uridine(8),thymidine(9),2'-deoxyuridine(10),and 2'-deoxythymidine(11),respectively.Conclusion All the isolated compounds were obtained from the sponge H.cymaeformis for the first time.Compounds 3 showed significant inhibitory activities towards chronic myelogenous leukemia(K562) and human hepatoma(SMMC-7721) cell lines.

Abstract:Serinolamide A, isolated from a species of marine cyanobacteria, exhibits a moderate agonist effect and selectivity for the CB1 cannabinoid receptor, which is unusual for marine natural products. Herein, we reported a highly efficient enantiospecific first total synthesis of (+)-serinolamide A from l-serine in nine steps with 30% overall yield. The synthesis method provides a facile, practicable, and economical approach for the preparation of other similar endocanabinoid lipids.

Abstract:A new bisuracil analogue, 3,3-bis(uracil-1-yl)-propan-1-aminium (1), together with four known N-containing metabolites (2-5), were isolated from the South China Sea sponge Agelas clathrodes. Their chemical structures were established on the basis of spectroscopic and spectrometric analysis and comparison with known compounds. Compound 1 is an unusual naturally occurring bisuracil analogue, and compound 2 was isolated from a natural source for the first time. Compounds 2 and 4 exhibit moderate cytotoxicity against cancer cell line SGC7901.

Abstract:A new dimeric macrolide xylopyranoside, cocosolide (1), was isolated from the marine cyanobacterium preliminarily identified as Symploca sp. from Guam. The structure was determined by a combination of NMR spectroscopy, HRMS, X-ray diffraction studies and Mosher's analysis of the base hydrolysis product. Its carbon skeleton closely resembles that of clavosolides A-D isolated from the sponge Myriastra clavosa, for which no bioactivity is known. We performed the first total synthesis of cocosolide (1) along with its [α,α]-anomer (26) and macrocyclic core (28), thus leading to the confirmation of the structure of natural 1. The convergent synthesis featured Wadsworth-Emmons cyclopropanation, Sakurai annulation, Yamaguchi macrocyclization/dimerization reaction, α-selective glycosidation and β-selective glycosidation. Compounds 1 and 26 potently inhibited IL-2 production in both T-cell receptor dependent and independent manners. Full activity requires the presence of the sugar moiety as well as the intact dimeric structure. Cocosolide also suppressed the proliferation of anti-CD3-stimulated T-cells in a dose-dependent manner.

Abstract:The glycan portion of ganglioside HLG-2, which was identified in the extracts of the sea cucumber Holothuria leucospilota, was synthesized in a highly efficient and stereoselective manner. The unusual sequence of the trisaccharide moiety, alpha-N-glycolylsialy1-(2,4)-alpha-N-acetylsialyl-(2,6)-glucoside, was assembled by stereoselective coupling of a 5-N,4-O-carbonyl-protected sialyl phosphate donor, a N-2,2,2-trichloroethoxycarbonyl (Troc)-protected sialyl acceptor, and a (trimethylsily)ethyl-beta-glucosyl acceptor in high yield. The synthesis featured the high-yielding construction of two alpha-sialyl linkages.

Abstract:Fucosylated chondroitin sulfate (fCS), a representative marine polysaccharide isolated from sea cucumber, possesses diverse biological functions especially as a promising anticoagulant. However, its supply suffers from the challenges of high-cost materials, different species, and batch-to-batch variability. In the present study, we designed a concise route for the synthesis of functional glycomimetics by natural fCS as a template. 4-(4,6-Dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride-mediated amidation was applied on chondroitin sulfates for site-selective alkynylation with controllable ratios between 0.15 and 0.78. A small library of 12 fCS glycomimetics with specific sulfation patterns and fucose branches was prepared through copper-catalyzed azide-alkyne cycloaddition, which was fully characterized by nuclear magnetic resonance spectroscopy and size-exclusion chromatography with multiangle light scattering and refractive index. Through screening of their biological activities, CSE-F1 and CSE-SF1 exhibited anticoagulant activities through intrinsic pathway and inhibition of factor Xa by antithrombin III. The concise approach developed herein supplies novel glycopolymers to mimic the distinct functions of natural polysaccharides and promote the development of marine carbohydrate-based drugs.

Abstract:Comprehensive Summary Ten new cembrane‐type diterpenoids, namely xishaglaucumins A—J (1—10), along with ten known related ones (11—20), have been isolated from the soft coral Sarcophyton glaucum collected off the Xisha Island in the South China Sea. Their structures were elucidated by extensive spectroscopic analysis, quantum mechanical nuclear magnetic resonance (QM‐NMR) methods, X‐ray diffraction analysis, chemical methods and comparison with the reported data in the literature. The absolute configuration of new compounds 1, 5 and known compounds 11, 12, 16 and 20 were determined either by chemical methods or by X‐ray diffraction analysis using Cu Kα ( λ = 1.5417 Å). In in vitro bioassay, compound 4 exhibited inhibitory effects on lipopolysaccharide (LPS)‐induced inflammatory responses in BV‐2 microglial cells.
