Abstract:Objective To clone the homologous sequences of Human Testis Development Related Gene 1 (TDRG1) from different species and to explore the potential animal models for further function investigation of TDRG1. Methods The nucleotide sequence of TDRG1 was used as a query sequence to obtain the homologous sequences from the mouse, rat, chimpanzee and rhesus macaque genome databases by BLASTN program. The homologous sequences of TDRG1 were then confirmed by RT-PCR. Immunohistochemistry was used to analyze the homologous protein expression in the testes of these species. Results Bioinformatics analysis indicated that no homologous sequences of TDRG1 were found in the mouse and rat genome databases. However, there were homologous sequences in chimpanzee and rhesus macaque genome databases, the similarity percent were 98% and 90% respectively. The homologous sequence in rhesus macaque testicle was confirmed by RT-PCR. Immunohistochemistry showed that the expression of homologous protein TDRG1 was detected in rhesus macaque testicle tissue. However, neither homologous nucleotide sequences nor homologous protein were detected in mouse and rat testes tissue; Conclusion No gene sequence was found to be homologous with TDRG1 in mouse and rat testes. But in rhesus macaque and chimpanzee testes, homologous genes of TDRG1 was found. These results provided theoretical basis for developing TDRG1 transgenic models or TDRG1 knockout animal models.
