Abstract:[Purpose] To investigate the expression of Toll-like receptor 3 (TLR3) in human head and neck squamous cell carcinoma (HNSCC), and the correlation between TLR3 and NF-κB. [Methods] Protein levels of TLR3 and NF-κB in 7 species of HNSCC cell line, HNSCC tissues and para-tumor tissues, leukoplakia and oral and nasal normal mucosae were measured by Western blot. [Results] The proteins of TLR3 and NF-κB were expressed in all HNSCC cell lines; 70% of analyzed HNSCC tissues showed a significant expression of TLR3, which correlated positively with NF-κB expression (r=0.952, P0.001); however no TLR3 expression was detected in para-tumor tissues, leukoplakia, and normal mucosae in oral and nasal cavities. [Conclusion] TLR3 probably participates in oncogenesis of HNSCC, and therefore presents a potential target protein for anti-tumor therapy.
